Abstract
与T细胞或NK细胞表面的P-选择素糖蛋白配体1(PSGL-1)结合的化合物，可用于诱发T细胞或NK细胞的消减和/或诱发T细胞或NK细胞的细胞凋亡。本发明的化合物及方法可用来调节自身免疫性疾病、器官移植排斥以及过敏性疾病中的不需要的T细胞或NK细胞诱发的免疫应答。

本发明乃是有关于调节免疫应答的组合物与方法。
发明背景
如何调节不需要的免疫应答对治疗如自身免疫性疾病、器官移植排斥、过敏疾病以及一些癌而言，至今仍是最关键性的课题之一。过度攻击性的T细胞的活性可用抑制免疫或诱导免疫耐受的方法来加以控制。耐受是免疫系统对某一抗原不应答的一种状态，而免疫抑制是降低对抗原的免疫应答，因此通常需要持续性的给药。在器官移植中，T细胞在对同种抗原的免疫应答中扮演了不可或缺的角色。现行免疫抑制的治疗法通常包括采用皮质类固醇、环胞菌素A(cyclosporin)或雷帕霉素(rapamycin)，其可阻断对T细胞而言是关键的生长因子的白介素-2(IL-2)的转录，或抑制白介素-2依赖型的细胞增生。但是有一些单克隆抗体，或为T细胞消减试剂(T cell-depletingagent)(如抗CD3、CD4、CD8的抗体)，或为细胞因子信号传递的抑制物或T细胞共同刺激路径的抑制物(如抗CD25、B7-1、B7-2、CD152、CTLA4的抗体)，这些单克隆抗体显示对减少排斥作用的发生有效，而副作用及毒性不高。这些抗体药物中有部分显示对于治疗自身免疫性疾病与延长移植体存活具有某种程度的成效。
目前已经广泛公认细胞凋亡对维持免疫系统正常功能极为重要，也是移除不必要细胞的主要机制(Kabelitz et al.Immunol.Today 14：338-340(1993)；Raff，Nature 356：397-399(1992))。不论是源自细胞内部或外部的种种信号皆会影响细胞的生与死。抗T细胞表面分子的抗体如抗Fas(或称CD95，分子量43kD)、TNFR2(分子量75kD)、CD2(分子量45kD)及CTLA-4(分子量33kD)的抗体均会诱发T细胞凋亡(Osborne，Curr.Opin.Immunol.8：245-248(1996)；Lin et al.J.Immunol.158：598-603(1997)；Zhang et al.Nature：377：348-350(1995)；Lai et al.Eur.J.Immunol.25：3243-3248(1995)；Mollereau et al.J.Immunol.156：3184-3190(1996)；Gribben et al.Proc.Natl.Acad，Sci.USA 92：811-815(1995))。企图利用Fas与TNFR2分子来控制不需要的T细胞时，会因为这两种分子并不只在免疫细胞上表达，而在其他如肝脏等重要脏器的细胞表面也有表达而在应用上受到妨碍，这种分子表达模式可能限制了这两种抗体在治疗上的运用(Ogasawara et al.Nature 364：806-809(1993)；Pfeffer et al.Cell：73：457-467(1993)；Engelmann et al.J.Biological Chemical264：14497-14504(1990))。
发明概述
本发明是基于如下发现即T细胞表面抗原P-选择素糖蛋白配体1(PSGL-1)可用来消减T细胞或诱发T细胞进行细胞凋亡。T细胞消减对治疗过量或不需要的T细胞介导的免疫应答，或是与过量或不需要的T细胞增殖相关的病症特别有用，例如T细胞的消减可降低或是除去不需要的T细胞活性或增殖，而该T细胞的活性或增殖与自身免疫疾病、器官移植排斥、过敏性疾病和/或T细胞癌(T cell-derived cancers)等相关联。因此，本发明包含使用PSGL-1功能的调节剂以避免或减少经由T细胞介导的免疫应答的方法，以及筛选PSGL-1功能的调节剂的方法。
一方面，本发明的特征为一种在个体中预防或降低由T细胞介导的免疫应答的方法。该方法包括下列步骤：选择一个体，其被诊断出患有以过强或不需要的由T细胞介导的免疫应答的疾病，或有患该疾病的危险，以及对该个体给药能与T细胞表面的PSGL-1结合的化合物，其中所述化合物与T细胞表面的PSGL-1的结合可诱发造成T细胞死亡的信号传导途径，因而预防或降低该个体中T细胞介导的免疫应答。
用于本方法的化合物可以涵盖与PSGL-1能特异性结合的抗体，或是抗体的抗原结合片段。于一例示中，此化合物为与PSGL-1能特异性结合的单克隆抗体。于一实施方案中，该方法包括给药能与单克隆抗体结合的试剂(agent)及诱发T细胞表面多个PSGL-1抗原交联的步骤。于一实施方案中，该方法包括诱发T细胞表面多个PSGL-1抗原的交联，其中所述交联会诱发信号传递过程而造成T细胞的死亡。
在一例示中，本方法包括选择一被诊断出有自身免疫性疾病的个体；另一例示中，该方法包括选择已接受或计划接受同种异体或异种器官移植的个体；于另一例示中，本方法包括选择一个诊断出有过敏病症的个体；于另一例示中，本方法包括选择一个诊断出有T细胞癌的个体的步骤。
在一个实施方案中，所述T细胞为活化的T细胞。在一个实施例中，所述T细胞是CD4+的T细胞。在另一实施例中，所述T细胞是CD8+的T细胞。
在一个实施方案中，本方法包括检测采集自给药化合物之前的个体样品(第一生物样品)的T细胞数目，并将该数目与采集自给药化合物后的个体样品(第二生物样品)的T细胞的数目相比较。
在另一实施方案中，本方法包括检测采集自给药化合物之前的个体样品(第一生物样品)的T细胞的生物活性，并将该生物活性与采集自给药化合物后的个体样品(第二生物样品)的T细胞的生物活性相比较。
在一个实施方案中，在个体给药后造成外周血液中至少消减20％的CD3+细胞。在一些实施方案中，在个体给药后造成外周血液中至少30％、40％、50％或更多的CD3+细胞的消减。
在一个实施方案中，在接触抗体或其抗原结合片段后，抗体或其抗原结合片段诱导了外周血液中至少20％的CD3+细胞死亡；在一些实施方案中，在个体给药后会诱发外周血液中至少30％、40％、50％或更多的CD3+细胞的死亡。细胞死亡现象可于任何时候检测，如在接触抗体或其抗原结合片段后的一、二、三、四、五、六、七或更多天。
另一方面，本发明的特征为一种诱发T细胞或自然杀伤(NK)细胞死亡的方法。本方法包括以下步骤：提供细胞表面表达PSGL-1的T细胞或自然杀伤细胞，以及将能与T细胞或自然杀伤细胞表面的PSGL-1结合的化合物与T细胞或自然杀伤细胞接触，其中该化合物与T细胞或自然杀伤细胞表面的PSGL-1的结合会诱发信号传递造成T细胞或自然杀伤细胞的死亡。
在本方法中使用的化合物可包括对PSGL-1具特异性结合的抗体或其抗原结合片段。在一个实施例中，所述化合物是对PSGL-1具特异性结合的单克隆抗体。在一个实施方案中，该方法包括使此单克隆抗体与结合该单克隆抗体的试剂接触的步骤，诱导T细胞或自然杀伤细胞表面的多个PSGL-1抗原的交联。
在一个实施方案中，该方法包括诱导T细胞或自然杀伤细胞表面的多个PSGL-1抗原的交联的步骤，其中所述交联诱导导致T细胞或NK细胞死亡的信号传导过程。

本发明的优点为其容许在不并发不希望有或损伤性免疫应答的情况下消减T细胞数目或诱导T细胞的细胞凋亡。譬如，向个体给药抗PSGL-1的抗体不会造成炎性细胞因子如IL-2或TNF-α水平的不期望的升高。
本发明的另一优点是以细胞表面蛋白质PSGL-1为标靶，因其主要表达在白细胞，尤其是T细胞与自然杀伤细胞上，因此，本文所述的化合物不诱发其他细胞如肝细胞等的明显水平的凋亡。在无明显诱导威胁生命的全身性细胞因子应答及在不伤害其他脏器系统的情况下，以T细胞与自然杀伤细胞(参与器官移植排斥的重要的CD3-细胞类型)为标靶的选择性消减，是期望免疫抑制剂所具有的特征。
除非有其他的定义，在此使用的所有技术及科学名词的意义，与本发明所属领域内的普通技术人员所理解的定义相同。虽然相似或等同于在此所述的材料与方法的那些材料和方法也可用在本发明的实施和测试中，适合的材料与方法将述如下。所有的出版品、专利申请、专利及其他在此提及的参考文献完全纳入参考之用。万一有专门用语上的抵触，则以本说明书为依归。再者，所叙述的材料与方法仅为说明例证，而并非用以限定本发明。

发明详述
本发明是一种藉由调节T细胞表面的PSGL-1分子的功能来调节T细胞活性的方法。PSGL-1与在此所述的组合物结合后可造成T细胞数目消减或诱发T细胞进行细胞凋亡。这些组合物因此可作为控制免疫相关疾病，如：自身免疫性疾病、器官移植排斥、过敏性疾病和/或T细胞癌的治疗剂。该组合物对任何生物样本(其中，T细胞的存在和活性是不希望的)中T细胞的消减也是有用的。
PSGL-1蛋白质
PSGL-1为一细胞表面粘附分子，其表达于嗜中性白细胞、T及B淋巴细胞、自然杀伤细胞、单核细胞、树突细胞以及有CD34的原始的人造血前体细胞上。通过与选择素(Selectins)结合的能力，PSGL-1会诱导白细胞滚动到内皮组织上及白细胞渗入炎症组织中。PSGL-1介导的T细胞与E选择素和P选择素的结合，或迁移是受到不同调节的。例如，在T细胞由初始T细胞向记忆性T细胞转化时，T细胞上才被诱导出CLA(皮肤淋巴细胞抗原)表位。只有活化的1型T辅助细胞而非2型T辅助细胞才表达功能性的PSGL-1，并且有能力移行到皮肤炎症区域。
PSGL-1为一唾液酸化粘蛋白(Sialomucin)，必须加以特别的唾液酸化、海藻糖基化、硫酸化才能与P-选择素结合。PSGL-1分子因在其N末端存在不同程度的糖基化和硫酸化位点而有多个同工型(isoform)。静止的外周血T细胞和B细胞、淋巴细胞系及在体外活化的外周血T细胞，有相近水平的PSGL-1表达量。然而，只有活化的T细胞才会表达出功能性型态的PSGL-1，并与P-选择素产生高亲和性的结合。这种活化依存性的结合活性看来是不同的翻译后修饰作用所造成的结果，在活化的T细胞中α(1，3)海藻糖基化转移酶活性水平升高也提示了这一推断。PSGL-1同工型对L-选择素和E-选择素也显示不同的亲和性，例如，CLA阳性同工型的人类T细胞既可以在E-选择素上又能在P-选择素上粘附和滚动，仅表达PSGL-1而无CLA表位的T细胞就只能与P-选择素结合。再者，PSGL-1与P-选择素的结合取决于其N末端十个氨基酸构成的肽，该末端十个氨基酸的肽包括三个用于硫酸化的酪氨酸残基，及一个用于糖基化的苏氨酸残基。
PSGL-1蛋白质可以藉由DNA重组方法和/或经由生物材料中分离出天然PSGL-1来制备。重组的PSGL-1蛋白质可在原核或真核细胞中，体内或体外制备，编码PSGL-1的核酸可用于该蛋白的重组制备(参考，例如GenBank Accession NM_003006中编码PSGL-1多肽的核酸实例)；抗PSGL-1的抗体也可用来做PSGL-1抗原的纯化(Herron at al.(2000)Science Jun 2；288(5471)：1653-56；WO 00/25808)和/或用于本文所述的方法。对PSGL-1的进一步研究描述在如下文献中，但不限于这些文献：Sako et al.(1993)Cell75：1179，Vechino et al.(1995)J.Biol.Chem.270：21966和Veldman et al.(1995)J.Biol.Chem.270：16470。
对PSGL-1的重组制备而言，同时表达PSGL-1及负责其糖基化修饰的α(1，3)海藻糖基化转移酶Fuc-TVII，对PSGL-1的功能表现都可能是需要的，另外/或者，在制备重组PSGL-1时，同时用编码JPACE的核酸进行共转染，以除去前肽和/或编码酪氨酸磺基转移酶的核酸。
抗PSGL-1的抗体可用来分离及纯化来自生物材料中的PSGL-1抗原，任何表达PSGL-1蛋白的细胞如来源于个体的T细胞或T细胞系，均可作为纯化该蛋白质的供应源。一经纯化，PSGL-1蛋白可以用在本文所述的各种方法中。例如，纯化的PSGL-1蛋白可用于T细胞上PSGL-1功能的调节剂的筛选，或做为免疫原以制备抗PSGL-1蛋白的抗体。
另外，也可以利用选择素-Fc融合蛋白(如p-选择素-Fc融合蛋白)进行PSGL-1抗原的纯化。
抗PSGL-1抗体
PSGL-1多肽(或其免疫原性片段或类似物)在本发明的方法中可以用来产生抗体。如上所述，PSGL-1多肽或其肽片段的生产可以用重组技术或固相合成的方法来合成。重组的PSGL-1多肽或其肽片段可当成免疫原来制备抗PSGL-1的抗体。另外，抗PSGL-1抗体，如TAB4单克隆抗体，则可用来纯化PSGL-1多肽，例如，天然构象的PSGL-1多肽，继而再将该多肽用作免疫原来制备额外的抗PSGL-1抗体。
本发明的抗体可以是特异性结合PSGL-1多肽的单克隆抗体，多克隆抗体或基因工程抗体。“特异性结合”特定抗原，如PSGL-1多肽的抗体基本不识别或结合样本中的其它分子。因此，本发明的特征也表现在用如下方法来鉴定与本发明多肽结合的测试化合物(例如，抗体)，所述方法是将所述多肽与测试化合物接触，然后确定所述多肽是否与测试化合物相结合(例如，通过直接检测结合，直接检测干扰测试化合物与所述多肽结合的竞争化合物和/或用检测凋亡诱导活性的检测法直接检测结合来确定)。
一般而言，PSGL-1多肽可与载体蛋白如KHL进行连结，并混合佐剂后注射到哺乳动物宿主体内。该动物体内产生的抗体可藉由肽抗原亲和层析加以纯化。
具体地，有多种动物宿主可注射PSGL-1多肽或其抗原片段进行免疫。所述动物宿主通常包括兔、小鼠、豚鼠及大鼠。依宿主种类不同会采用不同的佐剂以增加免疫应答。所述佐剂包括弗氏佐剂(完全及不完全)、矿物胶(如氢氧化铝)、表面活性剂(如溶血卵磷脂(lysolecithin))、多聚醇(pluronicpolyols)、聚阴离子、肽、油性乳剂、匙孔血蓝蛋白及二硝基酚等。可能应用在人类的佐剂有BCG(卡介苗bacilli Calmette-Guerin)及小棒杆菌(Corynebacterium parvum)。经由上述免疫过程产生的多克隆抗体是抗体分子的异质群(heterogeneous populations)，可以从免疫过的动物血清中获得。
因此，本发明中的抗体包括多克隆抗体，另外尚有单克隆抗体、人源化(humanized)抗体或嵌合抗体、单链抗体、Fab片段、F(ab′)2片段、以及使用Fab表达文文库产生的分子。
单克隆抗体是针对特定抗原的抗体的均质群(homogeneouspopulations)，可采用上述PSGL-1多肽及标准杂交瘤技术而制备(参照，例如Kohler et al.，Nature 256：495(1975)；Kohler et al.，Eur J Immunol 6：511(1976)；Kohler et al.，Eur.J Immunol 6：292(1976)；Hammerling et al.，Monoclonal Antibodies and T cell Hybridomas，Elseveir，N.Y.(1981))。
具体地，单克隆抗体可以由如下技术来获得：用连续传代的培养的细胞系生产抗体分子的任何技术(如Kohler et al.，Nature 256：495(1975)和美国专利4,376,110所述)，人类B细胞杂交瘤技术(Kosber et al.，ImmunologyToday 4：72(1983)；Cole et al.，Proc Natl Acad Sci USA 80：2026(1983))以及EBV杂交瘤技术(Cole et al.，Monoclonal Antibodies and Cancer Therapy，AlanR.Liss，Inc.，pp.77-96(1983))。所述抗体可以是任何种类的免疫球蛋白，包括IgG、IgM、IgE、IgA、IgD及它们的任何亚类。本发明产生单克隆抗体的杂交瘤可以在体外或体内培养。在体内制备高滴度单克隆抗体的能力使得杂交瘤技术成为生产单克隆抗体特别有用的方法。
一旦产生，可采用标准的Western印迹或免疫沉淀分析法(例如，如Ausubel等(同上)所述的方法)来测试多克隆或单克隆抗体对PSGL-1特异性的识别。能特异性识别和结合PSGL-1的抗体可用于本发明。那些可结合T细胞(例如CD3+细胞)表面PSGL-1抗原并能诱导T细胞消减和/或凋亡的抗PSGL-1抗体尤其有用。
上述抗体可以例如用作治疗方案的一部分(例如，所述治疗方案用来降低或消除不期望的免疫应答，诸如与自身免疫性疾病，移植排斥，过敏性疾病和T细胞癌等疾病有关的T细胞介导的免疫应答)。这些抗体也可以应用于筛选具结合PSGL-1能力的候选化合物。
再者，制备“嵌合抗体”的技术已广为使用(Morrison et al.，Proc.Natl.Acad.Sci.USA 81：6851(1984)；Neuberger et al.，Nature 312：604(1984)；Takadaet al.，Nature 314：452(1984))，所述技术是将具有适当抗原特异性的小鼠抗体分子的基因与具有适当生物活性的人抗体分子的基因剪接在一起。所述嵌合抗体的分子中，不同的部分来源于不同的动物种类，例如一些嵌合抗体具有来源于小鼠单克隆抗体的可变区和人免疫球蛋白的恒定区。
另外还有制备单链抗体的技术(美国专利4946778及4704692)，可适用于制备抗PSGL-1多肽的抗体或其片段。单链抗体藉由氨基酸桥来结合重链与轻链的Fv区片段，形成单链多肽。
能够识别并结合特异性表位的抗体片段可经由已知的技术来生产。例如所述片段包括但不限于由木瓜蛋白酶分解抗体分子而产生的F(ab′)2片段，和还原F(ab′)2片段间的二硫键而生成的Fab片段。或者，可以建构Fab表达文文库(Huse et al.，Science 246：1275(1989))，以快速及容易地鉴定具有所需特异性的Fab片段。
目前已有本领域已知的方法将抗体加以人源化。例如，具有所需的特异性的单克隆抗体可通过商业途径进行人源化(Scotgene，Scotland；OxfordMolecular，Palo Alto，Calif)。而完全人类化的抗体，如在转基因动物中表达的人类抗体也是本发明的特点之一(Green et al.，Nature Genetics 7：13(1994)；和U.S.Pat.No.5,545,806和5,569,825)。
筛选具调节PSGL-1功能的化合物的方法
本发明也包含鉴定能与PSGL-1(或PSGL-1的某区域)相互作用的化合物的方法，所述化合物包括但不限于与PSGL-1结合时能诱发T细胞消减和/或T细胞凋亡的化合物。所述化合物还包括调节PSGL-1与调节PSGL-1活性的跨膜蛋白，胞外蛋白或胞内蛋白之间相互作用的化合物，以及调节PSGL-1活性的化合物。
依照本发明筛选的化合物包括但不限于肽，抗体及其片段，以及结合PSGL-1并调节PSGL-1介导的生物功能的其它有机化合物(如本文所述)。
这些化合物也包括但不限于肽，如可溶性肽，包括但不限于随机肽文库的成员(Lam et al.，Nature 354：82[1991]；Houghten et al.，Nature354：84[1991])，由D和/或L构型氨基酸，磷肽(phosphopeptides)制成的组合的化学衍生的分子文库成员(包括，但不限于，随机或部分简并的，定向磷肽文库(directed phosphopeptide libraries)的成员；Songyang et al.，Cell72：767[1993])，抗体(包括，但不限于多克隆抗体，单克隆抗体，人源化抗体，抗独特型抗体，嵌合抗体或单链抗体，Fab，F(ab′)2和Fab表达文库片段以及其表位结合片段)以及小的有机或无机分子。
可依照本发明筛选的其它化合物包括，但不限于影响PSGL-1蛋白活性的小有机分子(如本文所述)。
可以藉助于电脑模拟和搜寻技术，提供化合物的鉴定或对已知化合物的改善，所述化合物能调节PSGL-1的表达或活性。一旦确认出这样的化合物或组合物，其活性位点或区域即可分析得知。通常，这些活性位点可能是天然活性调节剂的结合位点。这些活性位点可以由本领域已知的技术加以确认，包括从肽的氨基酸序列、核酸的核苷酸序列、或从相关化合物或组合物与其天然配体的复合物的研究得知。在后一种情况下，利用化学或X线晶体学方法，可通过寻找到调节剂(或配体)结合的位置来找到活性位点。
虽然可以如上所述对能改变结合的化合物进行设计和制备，人们也可以筛选已知的化合物文库，包括天然产物文库，合成的化合物文库以及生物活性材料(包括蛋白)文库，从中筛选出能结合PSGL-1蛋白并能导致T细胞消减和/或诱发T细胞凋亡的化合物。
在体外，可设计一些系统来鉴定能与PSGL-1(或PSGL-1某区域)相互作用的化合物。所鉴定的化合物可以用来例如调节本文所述的T细胞活性，从而用于治疗如自身免疫性疾病，移植排斥，过敏性疾病以及T细胞癌等疾病。
用来鉴定与PSGL-1结合的化合物的试验的要素包括，在一定条件下制备PSGL-1(或其某区域)的反应混合物和测试化合物，然后在充足时间下使这两种化合物相互作用并结合，从而形成可从反应混合物中除去和/或检测到的复合物。所采用的PSGL-1种类可基于筛选目的不同而改变。在一些情况下，优选对应于PSGL-1区域的肽与赋予检测系统优点(例如，标记、分离产生的复合物等)的异源蛋白或多肽融合。
筛选分析可以多种方式进行。例如：一实现此分析的方法涉及将固定PSGL-1蛋白、多肽、肽、融合蛋白质或测试物质锚定到一固相上，并检测在反应终了时附着于固相的PSGL-1与测试物形成的复合体。在本方法的实施方案中，PSGL-1反应物可被锚定于固体表面，而并未固定的测试化合物可能被直接或间接地标记。
在实施时，微量滴定板可以便利地用作固相。锚定的化合物可用非共价或共价附着方式加以固定。非共价附着可能由简单地在固体表面覆盖蛋白质溶液并风干来完成；或者，也可以先将蛋白特异性的抗体，优选单克隆抗体加以固定，继而利用所固定的抗体将所述蛋白锚定于固体表面。此覆盖蛋白质的固相可事先制备并储存。
为了完成此分析，加入非固定成分至含有锚定成分的被覆盖的表面。当反应完成后，藉由例如洗涤方式去除未反应成分，以使所形成的任何复合物仍然固定于所述固相表面。检测固定在固相表面的复合物可用许多方法来完成，其中可以将预先未固定的成分预先标记，然后检测固定于表面的标记即代表复合物已形成。另外也可使用没有预先标记的未固定成分，然后再利用间接标记来检测固定于表面的复合物，如采用对预先未固定成分具特异性的标记抗体等。
或者，反应可在液相中进行，再将反应产物与未反应的成分分离开，并检测产生的复合体，如使用固定的PSGL-1蛋白、多肽、肽、融合蛋白或检测化合物特异性的抗体，来锚定在溶液中形成的任何复合物，然后用对上述可能复合物的其它成分特异的标记抗体来检测锚定复合物的存在。
另外，以细胞为基础的分析方法也可以用来鉴定能与PSGL-1反应的化合物。例如使用表达PSGL-1的细胞系，或经由基因工程方法使其表达PSGL-1的细胞系。以细胞为基础的分析方法在鉴定化合物功能效用上特别有其价值，例如一旦鉴定出某化合物具有PSGL-1蛋白的结合能力后，该化合物便可在体外或体内检测是否具有诱发T细胞凋亡或导致T细胞消减的能力。
药物组合物
本发明的目的是改变个体中的免疫应答，而包括例如，抗体、小分子、或其他可特异性结合PSGL-1多肽的化合物的药物组合物也是本发明的一个特征。在一优选实施例中，该化合物可用作PSGL-1的激动剂(Agonist)。
使用于本发明的药物组合物可以是传统方法的配方，采用一种或多种生理上可接受的载体或赋形剂。因此该化合物及其生理上可接受的盐类和溶剂化物可根据多种不同的给药路径而进行配制。
化合物可配制成胃肠道外注射给药的制剂，例如快速大量(bolus)注射或连续输液的制剂。注射用制剂可以用单位剂量方式来取用，例如保存在安瓶中或在多剂容器中，并加入防腐剂。该组合物可制备成悬浮液、溶液、或在油性或水性载体中的乳液的形式，并可包含如悬浮剂，稳定剂和/或分散剂等配药试剂。或者，活性成分可以是粉末形式，在使用前用适当的载体如灭菌的无热原的水来配制。
控制T细胞介导的免疫应答及消减T细胞群的方法
经本文详细筛检分析后所获得的那些化合物，可用来例如调节经由PSGL-1多肽所介导的生物功能和/或治疗与过强或不需要的细胞应答，例如T细胞介导的免疫应答有关的疾病。这些化合物包括但不限于肽、抗体及其片段、以及其他有机化合物，它们结合于T细胞表面的PSGL-1并诱发信号传递路径造成T细胞的死亡。本发明方法任选包括加入诱导细胞表面PSGL-1交联的交联剂。本发明所述的化合物可用于任何期望消减或除去T细胞活性的情况，特别应用于治疗包括自身免疫性疾病、器官移植排斥、过敏性疾病及T细胞癌。
此处所述的抗PSGL-1的化合物可治疗的疾病实例包括但不限于：糖尿病、关节炎(包括类风湿性关节炎、青少年风湿性关节炎、骨关节炎及牛皮癣性关节炎)、多发性硬化症、脑脊髓炎、重症肌无力、系统性红斑性狼疮、自体免疫性甲状腺炎、皮炎(包括特应性皮炎、湿疹性皮炎)、牛皮癣、斯耶格伦氏综合征(sjogen′s syndrome)、克罗恩氏病(crohn′s disease)、鹅口疮性溃疡、虹膜炎、结膜炎、角膜结膜炎、I型糖尿病、炎症性肠管疾病、溃疡性结肠炎、哮喘、过敏性哮喘、皮肤红斑狼疮(cutaneous lupus erythematosus)、硬皮病、阴道炎、直肠炎、药疹、麻风回变反应(leprosy reversal reaction)、红斑性麻风节结(erythema nodosum leprosum)、自体免疫性葡萄膜炎、过敏性脑脊髓炎、急性坏死出血性脑病、自发性进行性双侧感觉神经听觉丧失(idiopathic bilateral progressive sensorineural hearing loss)、再生障碍性贫血、纯红细胞性贫血、自发性血小板减少症、多发性软骨炎、韦格纳氏肉芽肿病(Wegener′s granulomatosis)、慢性活动性肝炎、史提夫-强生氏综合征(Stevens-Johnson syndrome)、自发性热带口疮、扁平苔癣，奎夫氏病(Graves′disease)、类肉瘤病、原发性胆管硬化、眼后房葡萄膜炎、间质性肺脏纤维化、移植物抗宿主疾病、移植病例(包括使用异源或异种组织的移植)如骨髓移植、肝脏移植、或任何器官组织的移植、过敏性疾病如特应性过敏(atopicallergy)、爱滋病及T细胞肿瘤如白血病和/或淋巴瘤。
本发明方法可用于不论是体外或体内于细胞群里消减T细胞。例如来自于个体的生物样本可藉由在体外与本发明所述的抗PSGL-1的化合物混合，并任选加入交联试剂来消减T细胞。本方法可应用于如在细胞群中提升非T细胞的细胞数目，以及减少或除去细胞群中T细胞的活性。
以下为本发明的实施例，本发明的范围并不仅限于下述实施例中。
实施例
实施例1：抗T细胞凋亡诱导蛋白(“TAIP”)单克隆抗体的制备
生产TAIP特异性的单克隆抗体的方法是采用Kohler与Milstein的人们熟知的细胞融合方法(European Journal of Immunology(1976)6：511-519)以制备分泌所需抗体的杂交瘤。将刀豆蛋白A(Con A)活化后的Balb/c小鼠脾脏T细胞注射给仓鼠后，取得其抗体生成细胞并与骨髓瘤细胞系融合，形成分泌抗体的杂交瘤。两种细胞群是用聚乙二醇融合的，并克隆出所产生的抗体生成细胞，再以标准的组织培养方法进行扩增。根据这些方法生产的杂交瘤分泌单克隆抗体，命名为TAB4，此单克隆抗体可在体外诱发T细胞凋亡，及在体内导致T细胞消减。而由TAB4识别的蛋白质命名为T细胞凋亡诱导蛋白(T cell apoptosis inducing protein，TAIP)。
C57BL/6J(B6)和BALB/c小鼠购自Jackson实验室(Bar Harbor，ME)。叙利亚仓鼠购自国立台湾大学医学院动物中心。
TAB4杂交瘤的浓缩的培养上清液以20000×g离心10分钟，所得上清以1∶1的比例用结合缓冲液(0.1M醋酸钠，pH5.0)进行稀释。将蛋白质G柱(柱床体积约1ml)以3-5ml结合缓冲液洗三次。将澄清的培养上清液上样至蛋白质G柱，且将流出的上清液再上样至柱中。以6-10ml结合缓冲液洗柱，并用5ml洗脱缓冲液(0.1M甘氨酸-盐酸，pH2.8)从所述柱中洗脱出结合的抗体。每一级分包含1ml洗脱的抗体，通过使每个1ml的级分与50μl 1MTris-HCl，pH7.5混合，将洗脱的级分调整至中性pH值。汇集包含抗体的级分，并在2升PBS(pH7.4)中透析三次，每次透析三小时。之后的抗体样品用Bradford所述的方法用Bio-Rad Protein Assay(BIO-RAD，Hercules，CA)检测其蛋白浓度。
实施例2：小鼠脾细胞悬浮液的制备及T细胞的活化与富集
小鼠脾脏浸渍于8ml的Hank′s平衡盐溶液(HBSS)中，用灭菌盖玻片小心磨碎，细胞液移入15ml的离心管(Coaster)，以200×g离心5分钟，倒掉上清液，细胞沉淀层藉由轻拍管壁使之重新悬浮于剩余的缓冲溶液中。加入1ml红细胞裂解缓冲液(0.6M NH4Cl，0.17M Tris碱，pH7.65)，置于室温孵育2分钟以裂解混入的红细胞，再加入9ml HBSS快速终止反应。以200×g离心5分钟后倒掉上清液，所得的细胞沉淀层以HBSS洗两次后悬浮于RPMI培养液中。用血球计数器(Cambridge Scientific Inc.)及台盼蓝排除法来检测上述混合物中细胞的浓度及存活率。
将上述脾细胞用RPMI培养液调整至终浓度3×106/ml，并加入终浓度为2μg/ml的刀豆蛋白A来活化T细胞，细胞悬浮液移至6孔培养板(每孔5ml)或10cm的培养皿中(每一培养皿10ml)，在37℃，5％二氧化碳培养环境下培养48小时，然后收获培养物。这样活化的脾细胞(包括活化的T细胞)，重新悬浮于5ml的HBSS中，并小心覆盖到离心管中5ml 55％的Percoll溶液层上，此时小心勿破坏已分开的溶液层。在25℃以1900×g离心细胞13分钟且不使用煞车减速。从两层间的交界面收集富集的T细胞，并用HBSS洗二次后即可用于实验中。
实施例3：活化T细胞的凋亡
被活化的T细胞(见实施例2)以含有5ng/ml IL-2的PRMI培养液稀释到终浓度5×105个细胞/ml，并依据表1的处理条件用对照Ig、TAB4或抗CD3抗体进行处理。

在培养18-24小时后，使用7-AAD细胞凋亡分析来检测每一培养液中的细胞凋亡程度。处理过的细胞移至流式细胞仪(FACS)分析管(Falcon)，用冰冷的FACS溶液(PBS中含1％胎牛血清，0.05％叠氮钠)洗两次，在4℃，200×g离心得到细胞沉淀层。细胞重新悬浮于冰冷的FACS溶液至终浓度1-2×107个细胞/ml。此时使0.1ml细胞悬浮液与7-AAD混合，至7-AAD的终浓度为2μg/ml，然后于4℃避光培养20分钟进行染色。染色后的细胞以冰冷的FACS溶液洗二次，再悬浮于0.5ml的FACS溶液中，并以BD LSR流式细胞仪(Becton Dickison)进行分析。
图1显示代表性的时程实验结果，所述实验研究活化的T细胞对TAB4(抗TAIP)介导的凋亡信号获得敏感性的时间。实验方法为小鼠脾细胞以Con-A活化并维持在含有IL-2的培养液中。收获并悬浮活化的T细胞，然后用TAB4单克隆抗体或对照仓鼠Ig进行攻击，并以抗仓鼠IgG抗体做为交联剂进行反应。于第一天即观察到TAIP交联具有诱发低水平(6.5％)凋亡细胞死亡的能力，但是TAB4诱发细胞凋亡的程度在第2天增加至17％，第4天至52％的高峰，而在第6天下降至44％。而与仅用IL-2的培养物比较，对照仓鼠IgG不诱发特异的凋亡T细胞的死亡。抗CD3抗体(做为阳性对照)在活化48小时后，可以诱发38％的T细胞凋亡(数据未显示)。
实施例4：在不同组织中TAIP抗原的表达
细胞用冰冷的FACS溶液(PBS中含1％胎牛血清，0.05％叠氮钠)洗两次，装入FACS分析管(Falcon)中于4℃200×g离心，细胞重新悬浮于冰冷的FACS溶液至细胞终浓度为1×107个细胞/ml，并分成0.1ml的等份，装入FACS分析管(Falcon)中以备每次分析之用。此时加入TAB4单克隆抗体或对照仓鼠Ig(终浓度2μg/ml)至细胞中，在蔽光及4℃条件下孵育30分钟进行表面染色。细胞以冰冷的FACS溶液洗一次后再进行后续的染色：(1)向脾细胞加入含有细胞色素(cychrome)偶联的抗CD3抗体(2μg/ml)、FITC偶联的抗仓鼠Ig及PE偶联的抗CD8/CD4/CD19/CD11b/泛(pan)-NK/I-A/I-E/Mac-3抗体(2μg/ml)的100μl冰冷的FACS溶液；(2)向胸腺细胞加入含有FITC偶联的抗仓鼠Ig、PE偶联的抗CD8抗体及细胞色素偶联的抗CD4抗体(2μg/ml)的100μl冰冷的FACS溶液。在蔽光及4℃条件下反应30分钟。染色后的细胞以冰冷的FACS溶液洗两次，重新悬浮于1毫升的FACS溶液中，并以BD LSR流式细胞仪(Becton Dickinson)分析。
图3和图4显示经由FACS分析得知TAIP抗原在脾细胞与胸腺细胞不同亚群上的分布。如图3所示，CD19+B细胞表面表达低水平但可检测到的TAIP蛋白。然而在CD3+T细胞及部分自然杀伤细胞表面则可检测到大量的TAIP蛋白。大部分的CD4+、CD8+及CD4+8+胸腺T细胞表达大量的ATIP蛋白，相反的只有少量的CD4-8-胸腺T细胞表达TAIP蛋白(图4)。
收集来源于B6和BALB/c小鼠的组织，包括脑、胸腺、心脏、肺脏、肝脏、胃、肾脏、脾脏以及皮肤，于室温过夜固定于10％甲醛，并包埋于石腊块中。石腊块以Leica RM2135切片机切成4μm厚的组织切片，延展于45℃水中并平铺于有涂层的玻片上，玻片在37℃干燥以便使用于后续实验。
含有组织石腊切片的玻片依据标准程序加以脱腊及经二甲苯-100％乙醇的系列干燥之，最后保存在100％乙醇中。组织切片依据标准程序经由100％乙醇-90％乙醇-85％乙醇-70％乙醇-PBS的系列进行再水化(rehydrated)，最后置于PBS溶液中。接下来的反应都在潮湿箱中进行。将组织切片置于封闭(Blocking)缓冲液(含1％正常山羊血清)，于室温孵育1小时(或4℃过夜)以封闭非特异性的结合。移去封闭缓冲液，切片加入TAB4或正常仓鼠Ig(1∶200稀释)继续在室温中孵育1小时(或4℃过夜)。切片以PBS洗二次，每次5分钟以移去第一抗体(primary antibody)，再与1∶250稀释的碱性磷酸酶偶联的山羊抗仓鼠Ig反应，并在室温下孵育1小时。切片再次以PBS洗二次，每次5分钟，以移去酶偶联的抗体，并以BCIP/NBT底物溶液于室温避光呈色30分钟。之后再次以PBS洗切片，以移去多余的酶底物，并以PBS-乙醇-二甲苯的系列进行脱水，封固以用于显微镜下观察。
结果显示只在骨髓来源的组织中可检测到TAIP蛋白的表达，在所检测的其它组织上没有检测到该蛋白的表达。
实施例5：细胞表面TAIP抗原的生物素化及免疫沉淀
5×107个RL♂1或NIH-3T3细胞于含有0.5mg/ml Sulfo-NHS-生物素(Pierce)的1ml PBS中，置于冰上30分钟进行表面生物素化反应。反应后将细胞孵育于0.5ml Dulbecco的更改了的Eagle′s培养液(Life Technologies，Inc.)，置于冰上10分钟以终止反应，细胞以1ml的Dulbecco的更改了的Eagle′s培养液洗一次，再用1ml PBS洗两次。
已标记的细胞以5.0×107个细胞/ml的密度在含有完全蛋白酶抑制物混合剂(Roche)的冷的溶解缓冲液(1％ Triton X-100，20mM Tris-HCl，pH8.0，160mM NaCl，1mM CaCl2)中溶解15分钟，而不溶解的物质以10,000×g离心10分钟沉淀下来，这些步骤及所有后续步骤皆在4℃下进行。为了进行免疫沉淀，事先将溶解产物与50μl压紧的蛋白质G-Sepharose(AmershamPharmacia Biotech)孵育30分钟以去除非特异性结合蛋白质，粒状物被沉淀，将上清液分成多个等份(通常相当于5.0×107个细胞)与事先加入了10μgTAB4单克隆抗体或来自正常仓鼠血清的IgG的蛋白质G-Sepharose 20μl共同孵育。在4℃孵育4小时后，树脂以洗涤缓冲液(0.05％ Triton X-100，50mMTris-HCl，pH8.5，400mM NaCl，1mM CaCl2，1mg/ml卵白蛋白)洗四次，再以相似的洗涤缓冲液(250mM NaCl代替400mM NaCl)洗两次。特异性结合于TAB4的蛋白质以50μl的1×SDS样品缓冲液洗脱。洗脱的蛋白质以8％SDS-PAGE电泳分离并转移至硝酸纤维素膜(Millipore)上。滤膜用过氧化物酶偶联的卵白素(Avidin)(PharMingen)进行反应并以化学发光试剂(NEMTMLife Science Products)呈色来分析生物素化的蛋白。
如图2所示，在RL♂1细胞(TAIP+T细胞)中由TAB4鉴定出来的生物素化的表面蛋白的分子量约120kDa，而在3T3细胞(TAIP-细胞)中则未鉴定出所述蛋白。而且包含有仓鼠正常血清的蛋白质G-Sepharose没能回收到此120kDa的蛋白质。这些结果显示这种120kDa的蛋白质是TAB4单克隆抗体所识别的T细胞表面抗原。
实施例6：体内T细胞的消减
为检验体内TAB4对T细胞或其他细胞群的作用，在小鼠腹膜内注射300μg TAB4或对照仓鼠Ig，在第4天收取脾细胞、胸腺细胞及外周血单个核细胞，计算总细胞数并以FACS分析细胞表面标志。
为了FACS分析，细胞于4℃以2％多聚甲醛固定20分钟，洗二次，再悬浮于冰冷的FACS溶液至终浓度为1×107个细胞/ml。每一次分析FACS分析管(Falcon)中加入100μl等份的细胞悬浮液。TAB4或对照仓鼠Ig加到细胞中混合至终浓度2μg/ml，并于4℃避光孵育30分钟。细胞以冰冷的FACS溶液洗一次后，依下列条件进行反应：(1)脾细胞加入100μl冰冷的FACS溶液，其中含有细胞色素偶联的抗CD3抗体(2μg/ml)、FITC偶联的抗仓鼠Ig及PE偶联的抗CD8/CD4/CD19/CD11b/泛NK/I-A/I-E/Mac-3抗体(2μg/ml)；和(2)胸腺细胞加入100μl冰冷的FACS溶液，其中含有FITC偶联的抗仓鼠Ig、PE偶联的抗CD8抗体及细胞色素偶联的抗CD4抗体(2μg/ml)。在4℃避光进行反应30分钟。最后，染色的细胞以冰冷的FACS溶液洗两次，重新悬浮于1,000μl的FACS溶液中，之后以BD LSR流式细胞仪(BectonDickinson)分析。
注射后4天，在TAB4处理的小鼠的外周血白细胞(PBL)中，CD3+T细胞百分比从对照组小鼠的36.7％降至4.1％(表2)。而TAB4处理仅造成脾细胞总数些微下降。然而在在TAB4处理的小鼠中，CD3+T细胞数目下降62％，自然杀伤细胞数目下降50％，CD19+B细胞总数目稍微增加。从TAB4处理的小鼠取得的胸腺细胞总数仅是对照小鼠中所观察到的胸腺细胞水平的48％(减少了52％)。此外，除了CD4+T细胞，所有其他CD8+、CD4+CD8+及CD4-CD8-T细胞数目也都有减少，其中CD4+CD8+的T细胞亚群受到的影响最大(减少了64.7％)。

实施例7：抗TAIP抗体不诱导IL-2或TNF-α的分泌
Balb/c小鼠(H-2d)腹膜内注射300μg TAB4或对照仓鼠Ig。注射后7天分离脾细胞(做为应答细胞)，与丝裂霉素C处理的C3H(H-2k)脾细胞(做为刺激细胞)共同培养，3天后收取培养上清液，并以ELISA试剂盒(PharMingen)测量IL-2含量。如图5所示，与对照组小鼠相比较，来自TAB4处理小鼠的反应细胞其IL-2分泌被抑制。但是分析血浆中IL-2与TNF-α水平时，在对照组与TAB4处理小鼠的血清中其含量并无明显不同。既然IL-2的分泌对T细胞的活化是重要的，此结果显示特异性抗TAIP抗体如TAB4，可以应用于体内调节T细胞并控制不需要的T细胞介导的免疫应答，如与自身免疫性疾病或器官移植排斥有关的那些免疫应答。
实施例8：使用抗TAIP抗体预防移植排斥
8至12周龄小鼠(取自杰克森实验室)以马来酸乙酰丙嗪(Acepromazinemeleate)(Fermenta Animal Health Co.，Kansas City，MO)麻醉。皮肤移植前(皮肤移植手术前7天)，无胸腺切除的C57BL/6受体小鼠(H-2b)腹膜内注射500μg TAB4或同型对照抗体。七天后，将完全异基因的不相容的Balb/cj小鼠(H-2d)的胁腹外侧皮肤移植到经抗体预先处理过的C57BL/6小鼠的胁腹外侧。移植后七天，小鼠再次注射500μg TAB4或同型对照抗体。移值后每天监测小鼠。当有50％捐赠皮肤坏死时视为排斥。移植物存活的百分比如图7(n＝8)所示。数据显示TAB4处理延长了异基因皮肤移植物的存活率。
实施例9：TAIP经鉴定为PSGL-1
P-选择素糖蛋白配体-1(PSGL-1)，又名CD162，是白细胞上表达的主要的P-选择素配体，此白细胞包括T细胞(Sako et al.，1993，Cell 75：1179；Vachino et al.，1995，J.Biol.Chem.270：21966；Veldman et al.，1995，J.Biol.Chem.270：16470)。TAIP的生化特性如分子量与其二聚化的趋势，提示TAIP与PSGL-1可能为相似物的可能性。为了研究此两种抗原的关系，进行了以下测试：1)由TAB4沉淀的抗原是否也可被市面上可购得的抗PSGL-1抗体所识别；和2)从细胞溶解物中抗PSGL-1抗体是否能消减TAB4。
RL♂1T细胞以1.0×108个细胞/ml的细胞密度在含有完整蛋白酶抑制物混合剂的裂解缓冲液(1％ Triton X-100，20mM Tris-HCl，pH8.0，160mMNaCl，1mM CaCl2)中裂解一小时，而未裂解的物质则以10,000×g离心10分钟沉淀，上述及后续步骤皆在4℃下进行。相当于5.0×107个细胞的裂解物与事先加入10μg抗PSGL-1单克隆抗体(clone 2PHl，PharMingen，SanDiego，CA)、抗TAIP单克隆抗体、TAB4或来自正常仓鼠血清的IgG的20μl蛋白质G-Sepharose孵育，于4℃孵育4小时后，粒状物以洗涤缓冲液(0.05％ Triton X-100，50mM Tris-HCL，pH8.5，400mM NaCl，1mM CaCl2，1mg/ml卵白蛋白)洗五次，再以相似的洗涤缓冲液(250mM代替400mM NaCl)洗两次。结合的蛋白以40μl的1×SDS样品缓冲液洗脱。洗脱的蛋白以6％SDS-PAGE分离并转移至硝酸纤维素膜上，该膜以抗PSGL-1单克隆抗体进行免疫印迹，并用过氧化物酶偶联的山羊抗大鼠IgG(重链+轻链)及化学发光剂(Renaissance，NEN)进行显示。
表面生物素化的RL♂1T细胞在1.0×108个细胞/ml的细胞密度下在裂解缓冲液中裂解。细胞提取物与结合到40μl蛋白质G-Sepharose上的20μg抗体在4℃下过夜孵育。用抗PSGL-1单克隆抗体(2PHl)或对照大鼠IgG进行消减，或用TAB4或正常仓鼠对照血清进行消减。消减后的裂解物分别以TAB4或抗PSGL-1单克隆抗体进行免疫沉淀。免疫沉淀物于6％ SDS-聚丙烯酰胺凝胶上分离并以荧光自显影法检测。如图6所示，抗PSGL-1抗体可以从T细胞裂解物中消减TAIP蛋白。再者，用抗TAIP抗体(TAB4)免疫沉淀下来的蛋白质，可经由Western印迹分析得知能够被抗PSGL-1抗体所识别。
实施例10：抗PSGL-1抗体可诱发人类T细胞凋亡
为了确定PSGL-1在人类T细胞凋亡中所扮演的角色，对活化的T细胞进行时程实验，以找到该细胞对PSGL-1介导的细胞凋亡信号获得敏感性的时间。以植物血凝素(PHA)丝裂原刺激人类T细胞并使其在包含IL-2的培养液中进一步扩增。之后收取活化的T细胞，在IL-2与交联抗体存在下用抗PSGL-1抗体进行攻击。
从健康成人获得的人外周血加入肝素抗凝后，依不同细胞密度以Ficoll-Paque Plus(Pharmacia Biotech)富集外周血单个核细胞(PBMC)。此细胞以1％ PHA(Life Technologies，GibcoBRL)活化48小时，并将该细胞在整个分析期间培养在含有重组人IL-2(5ng/ml)的培养液中。为了评估抗人PSGL-1抗体诱导凋亡的能力，活化的细胞进行如下处理：(1)加入1μg/ml的抗PSGL-1抗体克隆KPL-1(BD PharMingen)及0.5μg/ml的交联剂兔抗小鼠Ig(Jackson ImmunoResearch Laboratory)；(2)加入纯化的同型对照小鼠Ig及交联剂兔抗小鼠Ig；或(3)仅加入交联剂兔抗小鼠Ig。处理六小时后，细胞以抗膜联蛋白V抗体(BD PharMingen)和PI(Sigma)进行染色，以FACS分析确定早期凋亡细胞的百分数。
如图8所示，在应用抗PSGL-1抗体加交联剂时，由PSGL-1激发的信号，可导致PHA活化的人PBMC(主要为T细胞)的明显水平的凋亡。经抗PSGL-1抗体处理过的培养物中凋亡细胞的百分比，从第三天的8.5％增加至第八天的24％。而用同位吻合(isotopic-matched)的对照抗体或仅用交联抗体的处理对上述细胞皆无任何作用。
实施例11：使用抗PSGL-1激动剂型抗体治疗自身免疫性疾病
非肥胖型糖尿病(NOD)小鼠，是一种被充分认可的自体免疫糖尿病动物，饲养于标准环境下。在大约20周龄时NOD小鼠会产生自发性糖尿病。在实验组中，每只小鼠在第14、15和17周龄分别接受300μg剂量的抗PSGL-1抗体(TAB4)腹膜内注射。在第24和26周龄各追加一次相同剂量的注射。而对照组则给予相同剂量的仓鼠Ig。在15周龄后以Medi-Test葡萄糖试纸(Macherery-Nagel，Germany)每周两次监测小鼠的尿糖。连续两次测量非空腹尿中葡萄糖水平超过300mg/dl时视为糖尿病。
如图9所示，与对照抗体处理相比，TAB4(抗PSGL-1抗体)处理产生明显的保护作用。因此抗PSGL-1抗体处理可抑制自身免疫T细胞的活性，并可延迟I型糖尿病的发生。
其他实施方案
首先必须要了解上述的发明以及其细节叙述仅是以阐明本发明为目的，但非限制本发明的范畴。本发明的其他观点、优点及修改皆包含于下述的权利要求范围之内。
Claims (8)
1.包含特异性结合T细胞表面P-选择素糖蛋白配体-1(PSGL-1)的抗体或其抗原结合片段的组合物制备用于降低个体的T细胞介导的免疫应答的药物的用途，其中所述个体已诊断患有以过强或不需要的T细胞介导的免疫应答为特征的疾病；其中所述抗体或其抗原结合片段与T细胞表面PSGL-1的结合可诱导导致T细胞死亡的信号转导途径，从而降低所述过强或不需要的T细胞介导的免疫应答；且其中所述以过强或不需要的T细胞介导的免疫应答为特征的疾病是牛皮癣。
2.包含特异性结合T细胞表面P-选择素糖蛋白配体-1(PSGL-1)的抗体或其抗原结合片段的组合物制备用于降低个体的T细胞介导的免疫应答的药物的用途，其中所述个体已诊断患有以过强或不需要的T细胞介导的免疫应答为特征的疾病；其中所述抗体或其抗原结合片段与T细胞表面PSGL-1的结合可诱导导致T细胞死亡的信号转导途径，从而降低所述过强或不需要的T细胞介导的免疫应答；且其中所述以过强或不需要的T细胞介导的免疫应答为特征的疾病是类风湿性关节炎。
3.包含特异性结合T细胞表面P-选择素糖蛋白配体-1(PSGL-1)的抗体或其抗原结合片段的组合物制备用于降低个体的T细胞介导的免疫应答的药物的用途，其中所述个体已诊断患有以过强或不需要的T细胞介导的免疫应答为特征的疾病；其中所述抗体或其抗原结合片段与T细胞表面PSGL-1的结合可诱导导致T细胞死亡的信号转导途径，从而降低所述过强或不需要的T细胞介导的免疫应答；且其中所述以过强或不需要的T细胞介导的免疫应答为特征的疾病是自身免疫性糖尿病。
4.包含特异性结合T细胞表面P-选择素糖蛋白配体-1(PSGL-1)的抗体或其抗原结合片段的组合物制备用于降低个体的T细胞介导的免疫应答的药物的用途，其中所述个体已诊断患有以过强或不需要的T细胞介导的免疫应答为特征的疾病；其中所述抗体或其抗原结合片段与T细胞表面PSGL-1的结合可诱导导致T细胞死亡的信号转导途径，从而降低所述过强或不需要的T细胞介导的免疫应答；且其中所述以过强或不需要的T细胞介导的免疫应答为特征的疾病是多发性硬化症。
5.包含特异性结合T细胞表面P-选择素糖蛋白配体-1(PSGL-1)的抗体或其抗原结合片段的组合物制备用于降低个体的T细胞介导的免疫应答的药物的用途，其中所述个体已诊断患有以过强或不需要的T细胞介导的免疫应答为特征的疾病；其中所述抗体或其抗原结合片段与T细胞表面PSGL-1的结合可诱导导致T细胞死亡的信号转导途径，从而降低所述过强或不需要的T细胞介导的免疫应答；且其中所述以过强或不需要的T细胞介导的免疫应答为特征的疾病是移植物抗宿主疾病。
6.权利要求1-5任一项所述的用途，其中所述抗体是单克隆抗体。
7.权利要求1-5任一项所述的用途，其中所述药物配制成胃肠道外注射给药的剂型。
8.权利要求6的用途，其中所述药物配制成胃肠道外注射给药的剂型。
