囊膜糖蛋白G蛋白是狂犬病病毒最主要的致病因子(6,7)。LEP株小鼠脑内接种时仍感染并致死,额外增加一个G蛋白是否会增强其致病能力。通过测定LD50并观察小鼠状态,我们发现rLEP-G株的LD50与其亲本株LEP没有显著差异,这表明rLEP-G株的毒力与亲本株LEP几乎是一样的。LEP高滴度外周感染小鼠可引起小鼠发病并致死,可见LEP还残留一定的神经侵害能力,rLEP-G株高滴度外周感染小鼠引起小鼠体重下降及个别后肢麻痹但并不引起小鼠死亡。影响RV致病性的其中一个因素是逃避细胞凋亡的能力(8)。增加G蛋白含量的重组病毒诱导细胞凋亡能力增强,外周感染时对小鼠的致病力下降,与之相应的是病毒的中和抗体滴度显著上升,这可能是因为内源性佐剂的释放刺激了细胞毒性T细胞反应且能促进树突状细胞的成熟和抗原递呈。The envelope glycoprotein G protein is the most important virulence factor of rabies virus (6,7) . Mice of LEP strain are still infected and lethal when inoculated in the brain, whether the addition of an additional G protein will enhance their pathogenicity. By measuring the LD50 and observing the state of the mice, we found that the LD 50 of the rLEP-G strain was not significantly different from its parent strain LEP, which indicated that the virulence of the rLEP-G strain was almost the same as that of the parent strain LEP. Peripheral infection of mice with high titer of LEP can cause morbidity and death in mice. It can be seen that LEP still has a certain nerve damage ability. Mice die. One of the factors influencing RV pathogenicity is the ability to evade apoptosis (8) . The ability of the recombinant virus with increased G protein content to induce apoptosis is enhanced, and the pathogenicity to mice is reduced during peripheral infection. Correspondingly, the neutralizing antibody titer of the virus is significantly increased, which may be due to the endogenous adjuvant Release stimulates cytotoxic T cell responses and promotes dendritic cell maturation and antigen presentation.

狂犬病病毒的RNA基因组以RNP复合物形式存在,RNA基因组模板及其转录子均被N蛋白紧密包裹起来,使其不受RNA酶的降解,维持高活性。Mebatsion等(9)首次拯救的表达外源蛋白CAT的重组病毒传代25次后仍能检测到CAT活性。插入外源片段后的重组RV RNA基因组的高遗传稳定性是其作为载体使用的优势之一。rLEP-G株在BHK-21细胞上连续多次传代后仍保有额外的G基因并维持病毒的高滴度和G蛋白高表达,表明在培养细胞中rLEP-G株的遗传稳定性。The RNA genome of rabies virus exists in the form of RNP complex, and the RNA genome template and its transcripts are tightly wrapped by N protein, so that it is not degraded by RNase and maintains high activity. Mebatsion et al . (9) rescued the recombinant virus expressing foreign protein CAT for the first time and could still detect CAT activity after 25 passages. The high genetic stability of the recombinant RV RNA genome after insertion of foreign fragments is one of the advantages of its use as a vector. The rLEP-G strain still retained the extra G gene and maintained a high titer of the virus and high expression of the G protein after multiple passages on BHK-21 cells, indicating the genetic stability of the rLEP-G strain in cultured cells.
