Abstract

发明领域
本发明涉及富含乳清蛋白的乳基产品(milk-based product)及其制备方法。
发明背景
已经表明乳清蛋白是优良的蛋白质源,尤其是在运动员的营养、肌肉量的增加和保持方面如此。因此,市场上有许多乳清蛋白粉和由其制备的饮料。通常,作为所述乳清蛋白产品的原料,乳清蛋白浓缩物作为粉末被使用,它通过将干酪(cheese)、夸克(quark)或酪蛋白乳清超滤以及通过随后将由超滤接收到的浓缩物干燥来制备。这些产品具有味道恶臭的问题,这是由蛋白水解(由发酵剂如干酪发酵剂和凝乳酶引起)、残余脂肪的氧化和其它味道缺点导致的。而且,在乳清产品的制备过程中除去矿物质使得味道比一般乳的味道更平淡。人们已经试图消除与味道有关的问题,由此已经使用各种食品添加剂、调味物质、调味制品和操作助剂对乳清产品进行了提味。
目前的乳清蛋白产品除了有味道问题外,还有所有乳清蛋白的营养价值不相等的问题。例如,在干酪生产过程中由酪蛋白释放到乳清中的糖巨肽(glycomacropeptide)的营养价值比α-乳清蛋白和β-乳球蛋白的营养价值小。糖巨肽构成了干酪乳清的总蛋白质的重要部分。
在已知乳清产品中包含的高含量乳糖还可引起另外的问题。众所周知,乳糖在世界上大批成人中引起耐受不良症状。
还普遍知道,对基于乳清蛋白的产品进行热处理可在产品中引起结构缺陷。这些产品通常被描述为片状的、粗糙的、结块的或沙状的。
鉴于以上问题,已知乳清蛋白产品的性价比是不吸引人的。因此,所述产品通常不是可以大规模获得的,而是作为在有限的机构如健康中心可获得的特供产品被提供给消费者。
乳基乳清蛋白产品通常是人们所熟知的。而且,用于将乳组分分离成单独部分的各种膜技术及其组合在文献中有大量描述。例如,WO94/13148公开了通过将脱脂乳进行微滤和超滤来制备未变性的乳清蛋白浓缩物的方法。酪蛋白被滞留在微滤保留物中,而α-乳清蛋白和β-乳球蛋白相当容易地穿过孔径为约0.1微米的微滤膜。
WO96/08155公开了采用微滤和超滤从脱脂乳原料中分离酪蛋白和乳清蛋白。例如,可以通过该方法制备乳清蛋白含量降低的乳饮料。
WO00/30461公开了在婴儿配方产品的制备中可以采用微滤以使氨基酸组成与人乳相似。
WO03/094623A1公开了采用数种膜技术、即超滤、纳滤和反渗透来制备不含乳糖的乳饮料。
人们希望可以提供一种没有已知产品的缺点、而是具有舒适味道和有利营养组成的乳清蛋白产品。
发明简述
我们已经出人意料地发现,与已知乳清产品有关的问题可以通过在通过膜技术制备的并且富含α-乳清蛋白和β-乳球蛋白的乳基乳清蛋白部分中加入酪蛋白来避免。出人意料的是,即使是少量的酪蛋白也足以改善产品的感官特性,如保持味道柔和可口。出人意料的是,本发明的乳清蛋白产品的结构和稳定性也是良好的,没有任何沙状物、薄片、沉积物或凝胶形成等。而且,产品的营养价值也得到提高。
在本发明的实施方案中,能够制备外观和味道象乳、但是具有更有利于运动员和其它运动爱好者的组成的乳清蛋白饮料。

发明详述
本发明的目的是提供一种乳清蛋白产品,其具有约25:75至约50:50以下的乳清蛋白:酪蛋白比例、基于干物质计算至少约20%的总蛋白质含量以及基于产品重量计算约2.5%至约8%重量的蛋白质含量。在本发明的实施方案中,乳清蛋白:酪蛋白比例为约25:75至约40:60。在本发明的特定实施方案中,所述比例为约30:70。在本发明的其它实施方案中,所述比例是约49:51、48:52、47:53、46:54和45:55。在本发明的其它实施方案中,所述比例是约26:74、27:73、28:72和29:71。
在本发明的实施方案中,产品的总蛋白质含量基于干物质计算为约30%至约60%。在另一项实施方案中,总蛋白质含量基于干物质计算为30%至60%。在本发明的其它实施方案中,总蛋白质含量基于干物质计算为约40%至约60%。在其它实施方案中,总蛋白质含量基于干物质计算为40%至60%。在其它实施方案中,乳清蛋白产品的总蛋白质含量基于干物质计算为至少20%。
本发明的乳清蛋白产品具有良好的感官特性,具体而言,它没有由在常规的干酪、凝乳和酪蛋白乳清中存在的糖巨肽和使人不愉快的代谢物引起的异味。此外,本发明的乳清蛋白产品还具有有利的营养特性和对健康的有利作用。而且,本发明的乳清蛋白产品的稳定性也是好的,其中没有观察到成片、沉降、胶凝或其它引起不希望的结构改变的现象。
在本发明的上下文中,术语“乳基”表示含有乳清蛋白和酪蛋白的乳来源的产品。
乳清蛋白产品可以由一种或多种通过各种膜技术或其组合由乳原料获得的不同组分来制备。乳清蛋白产品还可以包含乳来源的矿物质。乳原料可以是乳本身或者是浓缩物或按照预期方式经过预处理。乳原料可以补充有在制备乳产品中通常使用的成分,例如脂肪、蛋白质或糖部分等等。因此,乳原料可以例如是全脂乳、奶油(cream)、低脂乳或脱脂乳、超滤乳、透滤乳、微滤乳、不含乳糖或低乳糖的乳、经蛋白酶处理的乳、由乳粉重构的乳、有机乳或这些乳的组合或者任意这些乳的稀释物。乳可以来源于牛、绵羊、山羊、骆驼、马或产生适于营养的乳的任意其它动物。乳优选是低脂或脱脂乳。在本发明的更优选的实施方案中,乳清蛋白产品由脱脂乳制备。
本发明的乳清蛋白产品可以作为液体如饮料、浓缩物或粉末来提供。在本发明的特定实施方案中,乳清蛋白产品是饮料。在本发明的实施方案中,基于饮料的重量计算,饮料的总蛋白质含量为约2.5%至约8%。在另一项实施方案中,基于饮料的重量计算,饮料的总蛋白质含量为2.5%至8%。在另一项实施方案中,饮料的总蛋白质含量为约3.5%至约7%。在另一项实施方案中,饮料的总蛋白质含量为3.5%至7%。酪蛋白占总蛋白质含量的75%至50%、优选70%至50%,而富含α-乳清蛋白和β-乳球蛋白的乳清蛋白占25%至50%、优选30%至50%。
本发明的乳清蛋白产品的特征是它不含糖、甜味剂或调味剂,但是不限于该实施方案。在本发明的特定实施方案中,当乳清蛋白产品是即可使用的饮料时,则饮料中不包括糖、甜味剂或调味剂。
与牛乳的矿物质组成一样,本发明的乳清蛋白产品的矿物质组成是高度生理的。例如,本发明的乳清蛋白饮料通常可以含有0.5%至1.5%、优选0.6%至0.8%的矿物质。但是,本发明的乳清蛋白产品的钙含量低于普通乳的钙含量。因此,乳清蛋白产品可以被提供有补充性的钙和其它乳矿物质,例如由下述的本发明的方法接收到的纳滤渗透物,以使得钙含量达到正常乳中存在的水平或更高。因此,补充性的钙可以作为任意钙源如乳钙、葡萄糖酸钙、枸橼酸钙、乳酸钙等或其混合物来提供。
本发明的乳清蛋白产品还可以包括脂肪。产品的脂肪含量通常为约0%至3.5%。
在本发明的实施方案中,乳清蛋白产品是低乳糖或无乳糖产品。可以通过用于制备该产品的膜技术而获得低乳糖或无乳糖产品。而且,乳清蛋白产品中任何残留的乳糖可以通过酶被水解。在本发明的上下文中,“低乳糖”表示在乳清蛋白产品中乳糖含量低于1%。“无乳糖”表示乳清蛋白产品的乳糖含量为0.5g/份(例如,对于液体乳0.5g/244g,乳糖含量最多为0.21%),但是不超过0.5%。根据本发明,还可以制备含有少量碳水化合物和具有无缺点的感官特性的乳清蛋白饮料。
本发明的乳清蛋白产品可以用作原料来制备所有种类的酸乳产品和/或酸化的新鲜产品,通常为酸奶、发酵乳、viili和发酵奶油、酸奶油、夸克、酪乳、开菲尔(kefir)、乳制子弹饮料(dairy shot drinks)和其它酸乳产品。我们出人意料地发现,由本发明的乳清蛋白产品制备的酸乳产品的感官特性与常规酸乳产品的感官特性相似。
本发明的产品可以选自但不限于食物产品、动物饲料、营养产品、食品补充剂、食物成分、保健食品和药物产品。在本发明的实施方案中,产品是食物或饲料产品。在本发明的另一项实施方案中,产品是功能食品,即具有任意促进健康和/或预防和/或减轻疾病的性质的食品。食物产品、食物材料和/或药物产品和动物饲料各自的形式不受特别限制。
如上所述,由于其具有有利的营养组成,所以本发明的乳清蛋白产品本身或作为常规饮食的一部分适合于运动员和其它运动爱好者。本发明提供了用于支持和改善健康进食的包含乳清蛋白的组合物。该产品还可用于尤其是联合地用于减轻和/或预防成年型糖尿病、代谢综合征和少肌症(sarcopenia)。
本发明的另一个目的是提供乳清蛋白产品作为食物产品、动物饲料、营养产品、食品补充剂、食物成分、保健食品和药物产品的用途。在本发明的实施方案中,产品作为功能食品和/或营养产品被提供。在另一项实施方案中,产品作为药物被提供。
乳清蛋白产品可以由一种或多种通过膜技术获得的部分来制备。两种或更多种技术可以以适当的方式联用,包括微滤、超滤、纳滤和反渗透。
因此,本发明的另一个目的是提供制备乳清蛋白产品的方法,该方法包括:
-使乳基原料进行微滤,以分离出作为微滤渗透物的理想的乳清和作为微滤保留物的酪蛋白浓缩物,
-使至少一部分微滤渗透物进行超滤,以提供超滤渗透物和作为超滤保留物的乳清蛋白浓缩物,
-由超滤保留物和含酪蛋白的材料并且如果需要的话还由其它成分配制乳清蛋白产品,以便提供约20:80至约50:50以下的乳清蛋白:酪蛋白比例和基于干物质计算为至少约20%的总蛋白质含量。
乳基原料优选是脱脂乳。
任意含有酪蛋白的材料都可以在本发明中用作含酪蛋白的材料。在本发明的实施方案中,含酪蛋白的材料选自本发明的方法中获得的微滤保留物、乳、酸乳产品、酸化乳产品、发酵乳产品以及它们的任意组合。酸化乳产品可以例如是酸乳或酸乳酪(yoghurt)或它们的组合。如本文所用的术语“乳”表示从哺乳动物的乳腺获得的任意的正常分泌物,例如牛乳、山羊乳、骆驼乳、马乳或绵羊乳,或者产生适于营养的乳的任意其它动物的乳腺获得的任意的正常分泌物。乳可以补充有在制备乳产品中通常使用的成分,例如脂肪、蛋白质或糖部分等等。因此,乳包括例如全脂乳、低脂乳或脱脂乳、奶油、超滤乳(UF保留物)、透滤乳、微滤乳(MF渗透物)、由乳粉重构的乳、有机乳或任意这些乳的组合或稀释物。在一项实施方案中,乳是脱脂乳。在另一项实施方案中,乳是低乳糖或无乳糖乳。

在一项实施方案中,按照本发明的方法制备的乳清蛋白产品是其中蛋白质含量基于产品的重量计算为约2.5至约8%重量、优选约3.5%至约7%的饮料。
在配制乳清蛋白产品之后,可以酌情按照本身已知的方式进行热处理。
根据本发明,乳基原料进行微滤。然后,至少一部分微滤渗透物进行超滤。在本发明的实施方案中,可以使至少一部分包括大多数矿物质和糖(包括乳糖)在内的超滤渗透物进一步进行纳滤(NF)以将矿物质分离入NF渗透物和将糖分离入NF保留物。在另一项实施方案中,还可以使至少一部分NF渗透物进一步进行反渗透(RO)以将矿物质浓缩成RO保留物。可以利用由所述的进一步膜过滤得到的这些部分来配制本发明的乳清蛋白产品。在本发明的实施方案中,微滤保留物、超滤保留物和纳滤保留物用于制备本发明的乳清蛋白产品。在另一项实施方案中,超滤保留物和纳滤保留物用于制备本发明的乳清蛋白产率。在本发明的另一项实施方案中,微滤保留物、超滤保留物和反渗透保留物用于制备本发明的乳清蛋白产品。在本发明的一项实施方案中,乳清蛋白产品由乳、超滤保留物、纳滤保留物和水组成。
在本发明的另一项实施方案中,通过用水或由膜过滤获得的适宜部分进行透滤促进了微滤(MF)、超滤(UF)和/或纳滤(NF)。当透滤与微滤联用时,由将MF渗透物进行超滤获得的UF渗透物适合用作透析液(diawater)。当UF渗透物进一步进行纳滤时,NF渗透物适合用作超滤中的透析液。当NF渗透物还进一步进行反渗透(RO)时,RO渗透物适合用作纳滤中的透析液。在本发明的方法中可以使用一个或多个所述的透滤步骤。所述级分以及它们的组合可以来自于单个步骤或者分离的步骤。
本发明的方法在图1中以通用术语进行了解释。乳依次进行微滤、超滤、纳滤和反渗透。在图中,任选的操作以虚线表示。微滤、超滤和纳滤可以通过透滤技术、分别采用至少一部分超滤渗透物、纳滤渗透物或反渗透(RO)渗透物作为透析液(diawater)进行。本发明的乳清蛋白产品由UF保留物和含酪蛋白的材料组成。如果期望的话,将NF保留物和RO渗透物引入乳清蛋白产品中。含酪蛋白的材料可以包括由微滤获得的MF保留物。同样,含酪蛋白的材料可以完全地或部分地由MF保留物组成。
本发明的方法提供了具有良好感官特性如味道和口感、同时具有良好稳定性的乳清蛋白产品。通过该方法能够阻止使乳清蛋白产品具有令人不愉快的异味的糖巨肽和代谢物的释放。因此,通过实施本发明的方法能够减少、消除或掩盖乳清蛋白产品的异味。
以前的研究表明乳清蛋白的营养质量是有区别的。更具体而言,已经发现α-乳清蛋白比β-乳球蛋白具有更好的营养价值。根据该认识,可以以适当的方式调整本发明的乳清蛋白产品的组成以适应各种用途。在本发明中,通过对乳原料进行热处理或通过选择膜来实现乳清蛋白组成的调整。本发明的方法使用了在乳产品的热处理中本身已知的技术。可以在本发明的方法中使用的热处理的实例有巴氏灭菌法、高温巴氏灭菌法或在低于巴氏灭菌法温度的温度下加热足够长的时间。具体而言,可以提到的有UHT处理(例如将乳于138℃处理2至4秒)、ESL处理(例如将乳于130℃处理1至2秒)、巴氏灭菌法(例如将乳于72℃处理15秒)或高温巴氏灭菌法(95℃,5分钟)。热处理可以是直接的(蒸汽接触乳,乳接触蒸汽)或间接的(管式热交换器、板式热交换器、刮板式热交换器)。
在本发明的实施方案中,在微滤之前将乳在65℃至95℃的温度范围进行热处理15秒至10分钟以选择性地分离乳清蛋白成分。热处理的结果是,β-乳球蛋白发生变性并与酪蛋白结合,而α-乳清蛋白穿过膜。这样,可以增加微滤渗透物中的α-乳清蛋白含量。
在本发明的实施方案中,本发明的乳清蛋白产品中的乳糖被水解为单糖,如同本领域所熟知的那样。这可以使用可市售获得的乳糖酶、按照本身已知的方式进行。在本发明的实施方案中,乳糖水解在对配制的乳清蛋白产品进行膜过滤之后实现。在本发明的另一项实施方案中,乳糖水解步骤和微滤步骤相互同时开始。在本发明的另一项实施方案中,乳原料的乳糖水解在膜过滤步骤之前开始。
乳糖水解可以继续,只要乳糖酶例如被对乳清蛋白产品进行的热处理所灭活,所述乳清蛋白产品是在本发明的方法中接收到的各部分(UF保留物和MF保留物)的稍后阶段配制的。
给出以下实施例以进一步解释本发明而非将本发明限于此。
实施例1
将脱脂乳(1000L)通过孔径为800kDa的聚合物滤膜(Synder FR)进行微滤。使用95的浓集因数,包括透滤步骤。通过方程式1计算浓集因数。所形成的微滤保留物的量为190L,其具有的干物质含量为20.0%。
将在微滤中形成的渗透物(1890L)通过孔径为10kDa的聚合物超滤(UF)膜(Koch HFK-131)进一步过滤。将由超滤得到的渗透物进一步进行纳滤(NF),产生NF保留物和渗透物(130L)。
通过透滤、采用以上130L NF渗透物作为透析液进行超滤。超滤的总浓集因数为24(方程式1)。在超滤中,形成100L超滤保留物和1920L超滤渗透物,其中1080L用于微滤的透滤。将剩余的超滤渗透物(840L)通过截止值为200Da的滤膜(Desal5-DK)进行纳滤。纳滤的浓集因数为4.25(方程式1),由此形成197L纳滤保留物和644L纳滤渗透物,130L后者在如上所述的微滤渗透物的超滤的透滤中用作透析液。
未在微滤渗透物的超滤的透滤中用作透析液的剩余的纳滤渗透物被用于其它目的,或者通过反渗透膜(Koch HR)、通过采用浓集因数10(方程式1)进行浓缩。所形成的纳滤渗透物的反渗透渗透物的量为500L,其中44L在纳滤的透滤中用作透析液。所形成的纳滤渗透物的反渗透保留物的量为55L。
实施例2
在热处理仪器中在65℃至95℃的温度范围对脱脂乳(1000L)进行热处理15秒至10分钟以选择性地分离乳清蛋白成分。脱脂乳的热处理影响了乳清蛋白在微滤中的渗透,从而微滤渗透物富含不耐热性较低的α-乳清蛋白,其变性程度为0至26%,而β-乳球蛋白以1至90%的程度变性。对脱脂乳进行热处理后,将乳进行如实施例1中所述的过滤操作。
作为举例,在微滤渗透物中α-乳清蛋白和β-乳球蛋白的总量中的α-乳清蛋白比例(%重量)为38%(于75℃热处理30秒)至45%(于90℃热处理30秒)。
实施例3
如表1所示由实施例1的微滤保留物、超滤保留物和纳滤保留物以及乳矿物质粉和水配制了本发明的乳清蛋白产品。产品的乳清蛋白:酪蛋白比例为21:79,蛋白质含量基于干物质计算为52%。产品是不含乳糖的乳饮料,其中乳糖在配制后被酶水解至0.1%以下的水平。

Claims (23)
1.乳清蛋白产品,其具有约25:75至约50:50以下的乳清蛋白:酪蛋白比例、基于干物质计算至少约20%的总蛋白质含量以及基于产品重量计算约2.5%至约8%重量的蛋白质含量。
2.权利要求1的乳清蛋白产品,其中乳清蛋白:酪蛋白比例为约25:75至约40:60,优选约30:70。
3.权利要求1或2的乳清蛋白产品,其中总蛋白质含量基于干物质计算为约30%至约60%、优选基于干物质计算为约40%至约60%。
4.上述权利要求任一项的乳清蛋白产品,其中所述产品是其中蛋白质含量基于产品的重量计算为约3.5至约7%的饮料。
5.上述权利要求任一项的乳清蛋白产品,其中所述产品还包含补充性的乳矿物质。
6.制备乳清蛋白产品的方法,该方法包括:
-使乳基原料进行微滤,以分离出作为微滤渗透物的理想的乳清和作为微滤保留物的酪蛋白浓缩物,
-使至少一部分微滤渗透物进行超滤,以提供超滤渗透物和作为超滤保留物的乳清蛋白浓缩物,
-由超滤保留物和含酪蛋白的材料并且如果需要的话还由其它成分配制乳清蛋白产品,以便提供约20:80至约50:50以下的乳清蛋白:酪蛋白比例和基于干物质计算为至少约20%的总蛋白质含量。
7.权利要求6的方法,其中乳清蛋白产品的乳清蛋白:酪蛋白比例为约25:75至约50:50以下、优选约25:75至约40:60、更优选约30:70。
8.权利要求6或7的方法,其中总蛋白质含量基于干物质计算为约30%至约60%、优选基于干物质计算为约40%至约60%。
9.权利要求6-8任一项的方法,其中产品是其中蛋白质含量基于产品的重量计算为约2.5%至约8%重量、优选约3.5%至约7%的饮料。
10.权利要求6-9任一项的方法,其中含酪蛋白的材料选自微滤保留物、乳、酸乳产品、酸化乳产品、发酵乳产品以及它们的任意组合。
11.权利要求6-10任一项的方法,其中乳基原料是脱脂乳。
12.权利要求6-11任一项的方法,其中将超滤渗透物进行纳滤以提供纳滤保留物和渗透物。
13.权利要求12的方法,其中将纳滤渗透物进行反渗透以提供反渗透保留物和渗透物。
14.权利要求6-13任一项的方法,其中将透滤与微滤、超滤和纳滤一起使用。
15.权利要求14的方法,其中将微滤保留物、超滤保留物和纳滤保留物用于配制乳清蛋白产品。
16.权利要求6-15任一项的方法,其中在微滤之前将乳原料在65℃至95℃的温度范围进行热处理15秒至10分钟。
17.权利要求6-16任一项的方法,其中向乳清蛋白产品中加入补充性的乳矿物质。
18.权利要求6-17任一项的方法,其中乳清蛋白产品中的糖巨肽的释放被阻止。
19.权利要求6-18任一项的方法,其中乳清蛋白产品的异味被减少、消除或掩盖。
20.权利要求1-5任一项的或者通过权利要求6-19任一项的方法制备的乳清蛋白产品,用作食物产品、动物饲料、营养产品、食品补充剂、食物成分、保健食品和药物产品。
21.权利要求1-5任一项的或者通过权利要求6-19任一项的方法制备的乳清蛋白产品作为食物产品、动物饲料、营养产品、食品补充剂、食物成分或保健食品的用途。
22.权利要求21的用途,其中乳清蛋白产品被用作功能食品和/或营养产品。
23.权利要求1-5任一项的或者通过权利要求6-19任一项的方法制备的乳清蛋白产品在制备酸乳产品和/或酸化的新鲜产品如酸奶、发酵乳、viili、发酵奶油、酸奶油、夸克、酪乳、开菲尔和乳制子弹饮料中的用途。
