A method of preparing a special dietary supplement, the method comprising the steps of:
mixing grapes and water, squeezing the mixture into pulp by using a juicer, filtering the pulp to obtain a grape-water crude extract, and treating the grape-water crude extract with pectolytic enzyme for 5 days at the temperature of 25 ℃ to obtain a grape extract;
step two, mixing 10 parts of grape extract and 5 parts of haematococcus pluvialis to prepare a first mixture;
step three, adding 100 parts of clean peanut meal into a high-speed mixer, uniformly mixing, crushing, sieving, adding water, uniformly stirring, adding 5 parts of serine protease, adding a sodium hydroxide solution to adjust the pH value to 8, heating to 60 ℃, hydrolyzing for 5 hours, adding 2 parts of protease into the hydrolysate, adding dilute hydrochloric acid to adjust the pH value to 6, hydrolyzing for 3 hours, filtering to remove impurities to obtain an enzymolysis solution, adding 10 parts of peptone, 5 parts of skim milk powder and 10 parts of white granulated sugar into the enzymolysis solution, adjusting the pH value to 6.5 by adding dilute hydrochloric acid or a sodium hydroxide solution, sterilizing at high temperature, and cooling to obtain a culture medium;
inoculating bacillus subtilis into a culture medium for fermentation culture, and fermenting for 72 hours to obtain a fermentation liquid;
step five, centrifuging the fermentation liquor, collecting bacterial sludge, and then resuspending the bacterial sludge in normal saline to obtain a resuspension liquid;
and step six, adding the first mixture prepared in the step two, 20 parts of vitamins and 20 parts of glycine into the heavy suspension, fully mixing, adding dilute hydrochloric acid or sodium hydroxide solution to adjust the pH value, and performing vacuum freeze drying to obtain the special dietary supplement.
Example 2
A method of preparing a special dietary supplement, the method comprising the steps of:
mixing grapes and water, squeezing the mixture into pulp by using a juicer, filtering the pulp to obtain a grape-water crude extract, and treating the grape-water crude extract with pectolytic enzyme for 4 days at the temperature of 30 ℃ to obtain a grape extract;
step two, mixing 15 parts of grape extract and 8 parts of haematococcus pluvialis to prepare a first mixture;
step three, adding 120 parts of clean peanut meal into a high-speed mixer, uniformly mixing, crushing, sieving, adding water, uniformly stirring, adding 8 parts of serine protease, adding a sodium hydroxide solution to adjust the pH value to 8, heating to 60 ℃, hydrolyzing for 4.5 hours, adding 3 parts of protease into the hydrolysate, adding dilute hydrochloric acid to adjust the pH value to 6, hydrolyzing for 2.5 hours, filtering to remove impurities to obtain an enzymolysis solution, adding 12 parts of peptone, 7 parts of skim milk powder and 15 parts of white granulated sugar into the enzymolysis solution, adjusting the pH value to 6.8 by adding dilute hydrochloric acid or a sodium hydroxide solution, sterilizing at high temperature, and cooling to obtain a culture medium;
inoculating bacillus subtilis into a culture medium for fermentation culture for 60 hours to prepare a fermentation liquid;
step five, centrifuging the fermentation liquor, collecting bacterial sludge, and then resuspending the bacterial sludge in normal saline to obtain a resuspension liquid;
and step six, adding the first mixture prepared in the step two, 25 parts of vitamins and 30 parts of lysine into the heavy suspension, fully mixing, adding dilute hydrochloric acid or sodium hydroxide solution to adjust the pH value, and performing vacuum freeze drying to obtain the special dietary supplement.
Example 3
A method of preparing a special dietary supplement, the method comprising the steps of:
mixing grapes and water, squeezing the mixture into pulp by using a juicer, filtering the pulp to obtain a grape-water crude extract, and treating the grape-water crude extract with pectolytic enzyme for 3 days at the temperature of 35 ℃ to obtain a grape extract;
step two, mixing 20 parts of grape extract and 10 parts of haematococcus pluvialis to prepare a first mixture;
step three, adding 150 parts of clean peanut meal into a high-speed mixer, uniformly mixing, crushing, sieving, adding water, uniformly stirring, adding 10 parts of serine protease, adding a sodium hydroxide solution to adjust the pH value to 8, heating to 60 ℃, hydrolyzing for 4 hours, adding 4 parts of protease into the hydrolysate, adding dilute hydrochloric acid to adjust the pH value to 6, hydrolyzing for 2 hours, filtering to remove impurities to obtain an enzymolysis solution, adding 15 parts of peptone, 10 parts of skim milk powder and 20 parts of white granulated sugar into the enzymolysis solution, adjusting the pH value to 7.0 by adding dilute hydrochloric acid or a sodium hydroxide solution, sterilizing at high temperature, and cooling to obtain a culture medium;
inoculating bacillus subtilis into a culture medium for fermentation culture, and fermenting for 48 hours to obtain a fermentation liquid;
step five, centrifuging the fermentation liquor, collecting bacterial sludge, and then resuspending the bacterial sludge in normal saline to obtain a resuspension liquid;
and step six, adding the first mixture prepared in the step two, 30 parts of vitamins, 50 parts of arginine and glycine into the heavy suspension, fully mixing, adding dilute hydrochloric acid or sodium hydroxide solution to adjust the pH value, and performing vacuum freeze drying to obtain the special dietary supplement.
The technical solutions described above only represent the preferred technical solutions of the present invention, and some possible modifications to some parts of the technical solutions by those skilled in the art all represent the principles of the present invention, and fall within the protection scope of the present invention.
Claims (8)
1. A method for preparing a special dietary supplement, comprising the steps of:
mixing grapes and water, squeezing the mixture into pulp by using a juicer, filtering the pulp to obtain a grape-water crude extract, and treating the grape-water crude extract by using pectolytic enzyme at a certain temperature for a certain time to obtain a grape extract;
step two, mixing the grape extract and haematococcus pluvialis to prepare a first mixture;
step three, adding clean peanut meal into a high-speed mixer, uniformly mixing, crushing, sieving, adding water, uniformly stirring, adding a certain amount of serine protease, adding a sodium hydroxide solution to adjust the pH value to 8, heating to a certain temperature, hydrolyzing for a period of time, adding a certain amount of protease into the hydrolysate, adding dilute hydrochloric acid to adjust the pH value to 6, hydrolyzing for a period of time, filtering to remove impurities to obtain an enzymolysis solution, adding a certain amount of peptone, skim milk powder and white granulated sugar into the enzymolysis solution, adjusting the pH value to 6.5-7.0 by adding a dilute hydrochloric acid or a sodium hydroxide solution, sterilizing at high temperature, and cooling to obtain a culture medium;
inoculating bacillus subtilis into a culture medium for fermentation culture, and fermenting for a period of time to obtain a fermentation liquid;
step five, centrifuging the fermentation liquor, collecting bacterial sludge, and then resuspending the bacterial sludge in normal saline to obtain a resuspension liquid;
and step six, adding the first mixture prepared in the step two and a certain amount of vitamins and amino acids into the heavy suspension, fully mixing, adding dilute hydrochloric acid or sodium hydroxide solution to adjust the pH value, and performing vacuum freeze drying to obtain the special dietary supplement.
2. The method of claim 1, wherein the treatment temperature of the pectolyase enzyme in step one is 25-35 ℃ and the treatment time is 3-5 days.
3. The method of claim 1, wherein in step two, the weight parts of the grape extract and the haematococcus pluvialis are 10-20 parts and 5-10 parts respectively.
4. The method for preparing a special dietary supplement according to claim 1, wherein in the third step, 100-150 parts of peanut meal, 5-10 parts of serine protease, 2-4 parts of protease, 10-15 parts of peptone, 5-10 parts of skim milk powder, and 10-20 parts of white granulated sugar.
5. A process for the preparation of a dietary supplement according to claim 1, wherein the temperature for hydrolysis is 60 ℃ when the pH is adjusted to 8 in said step three, the time for hydrolysis is 4-5 hours and the time for hydrolysis is 2-3 hours when the pH is adjusted to 6.
6. A method for the preparation of a dietary supplement according to claim 1, wherein the fermentation time in step four is 48-72 hours.
7. A method of preparing a dietary supplement of claim 1, wherein said step six comprises vitamins 20-30 parts, amino acids 20-50 parts by weight.
8. A method of preparing a dietary supplement of claim 1, wherein said amino acids in step six are one or more of arginine, lysine, glycine, leucine.
