Claims (1)
- 【特許請求の範囲】 1.(a)バシラス(Bacillus)細胞の突然変異株を培養し、ここで、(i) 該突然変異株は、ポリペプチドの産生を促す条件下で、ポリペプチドをコードす る一次核酸配列とサーファクチンまたはそのアイソフォームの生合成または分泌 に関与する少なくとも1つの遺伝子の修飾を含有する二次核酸配列とを含み、そ して(ii)突然変異株は、同一条件下で培養された場合、バシラス細胞より少 ないサーファクチンまたはそのアイソフォームを産生し;そして (b)ポリペプチドを培地から単離する; 工程を含んでなるポリペプチドの製造方法。 2.バシラス細胞がバシラス・アルカロフィルス(Bacillus alkalophilus)、 バシラス・アミロリクマファシエンス(Bacillus amyloliquefaciens)、バシラ ス・ブレビス(Bacillus brevis)、バシラス・サーキュランス(Bacillus circul ans)、バシラス・コアギュランス(Bacillus coagulans)、バシラス・フィル ムス(Bacillus firmus)、バシラス・ラウトス(Bacillus lautus)、バシラス・レ ンタス(Bacillus lentus)、バシラス・リケンホルミス(Bacillus licheniformi s)、バシラス・メガテリウム(Bacillus megaterium)、バシラス・プミルス( Bacillus pumilus)、バシラス・ステアロサーモフィルス(Bacillus stearother mophilus)、バシラス・ズブチリス(Bacillus subtilis)またはバシラス・チュリ ンジエンシス(Bacillus thuringiensis)細胞である請求項1の方法。 3.バシラス細胞がバシラス・ズブチリス細胞である請求項2の方法。 4.バシラス細胞がバシラス・ズブチリスATCC 6051またはバシラス ・ズブチリスATCC 6051Aである請求項3の 方法。 5.バシラス細胞がバシラス・ズブチリスNCFB 736である請求項3の 方法。 6.遺伝子がsrfオペロンの遺伝子である請求項1の方法。 7.遺伝子がsrfAである請求項6の方法。 8.遺伝子がsrfBである請求項6の方法。 9.遺伝子がsrfCである請求項6の方法。 10.遺伝子がsrfDである請求項6の方法。 11.遺伝子がsfpである請求項1の方法。 12.同一条件下で培養した場合に、突然変異株がバシラス細胞よりも少なく とも約25%少ないサーファクチンまたはそのアイソフォームを産生する請求項 1の方法。 13.修飾がサーファクチンまたはそのアイソフオームの非発泡性変異体の産 生を引き起こす請求項1の方法。 14.ポリペプチドがバシラス細胞と異種である請求項1の方法。 15.突然変異株がプロテアーゼをコードする1つ又はそれ以上の遺伝子の修 飾をさらに包含する請求項1の方法。 16.遺伝子がnprEおよび/またはaprEである請求項15の方法。 17.突然変異株がspoIIACおよび/またはamyE遺伝子の修飾をさ らに包含する請求項1の方法。 18.異種ポリペプチドをコードする一次核酸配列とサーファクチンまたはそ のアイソフォームの生合成または分泌に関与する少なくとも1つの遺伝子の修飾 を含有する二次核酸配列とを含むバラス細胞の突然変異株であって、同一条件下 で培養した場合にバシラス細胞より少ないサーファクチンまたはそのアイソフォ ームを産生す る突然変異株。 19.生来のポリペプチドをコードする一次核酸配列とサーファクチンまたは そのアイソフォームの生合成または分泌に関与する少なくとも1つの遺伝子の修 飾を含有する二次核酸配列との少なくとも2つのコピーを包含するバシラス細胞 の突然変異株であって、同一条件下で培養した場合にバシラス細胞より少ないサ ーファクチンまたはそのアイソフォームを産生する突然変異株。 20.(a)サーファクチンまたはそのアイソフォームの生合成または分泌に 関与する少なくとも1つの遺伝子の修飾を含有する一次核酸配列とバシラス細胞 にとって異種性であるポリペプチドをコードする二次核酸配列とをバシラス細胞 中に導入し;そして (b)核酸配列を包含する工程(a)からの突然変異株を同定する; 工程を含んで成る請求項19の突然変異株の獲得方法。 21.(a)サーファクチンまたはそのアイソフォームの生合成または分泌に 関与する少なくとも1つの遺伝子の修飾を包含する一次核酸配列とバシラス細胞 にとって生来のポリペプチドをコードする二次核酸配列とをバシラス細胞中に導 入し;そして (b)核酸配列を包含する工程(a)からの突然変異株を同定する; 工程を含んで成る請求項19の突然変異株の獲得方法。
