Abstract
Description
本出願は、2003年12月30日に出願された、仮出願番号60/533,169号の米国仮特許出願に基づく国内優先権を主張するものである。なお、この仮特許出願の全体を本明細書に援用する。
米国政府は、国防総省により認められたDAAD 13−03−C−0047の条項に従って、本発明に対して特定の権利を有し得る。
本発明のシステム及び方法は、好ましくは、波動減衰、相変化、周波数変動、及び/又は共振周波数変動を決定するように、測定、又はさもなければ検知される音響−機械エネルギーを用いて、試験試料中の標的の生物学的検体の存在を検出し得る。
Claims (40)
- 内部容積を有するハウジングと、
前記ハウジングに動作可能に取り付けられ、検出面を有するセンサと、
前記ハウジングの前記内部容積内に位置し、前記検出面により画定される容積と、前記検出面から離間されて該検出面に対面し流れ前方制御部を備える対向面とを有する検出室と、
前記ハウジングの前記内部容積内に位置し、前記検出室と流体連通する廃棄室とを備える、検出カートリッジ。 - 前記検出面は、音響機械的導波管を備える、請求項1に記載のカートリッジ。
- 前記センサは、弾性表面波音響機械的センサを備える、請求項1に記載のカートリッジ。
- 前記流れ前方制御部は、前記検出室の前記対向面上のランド領域から突出し該ランド領域により分離される複数の離散構造体を備える、請求項1に記載のカートリッジ。
- 前記流れ前方制御部は、前記検出室の前記対向面において1つ以上の流路を備える、請求項1に記載のカートリッジ。
- 前記1つ以上の流路のうちの少なくとも1つの流路は、廃棄室の入力端と出力端との間で、前記検出室内に画定された長手軸に略垂直に配向される、請求項5に記載のカートリッジ。
- 前記流れ前方制御部は、前記対向面の一部を占める疎水性材料の1つ以上の領域と、前記対向面の一部を占める親水性材料の1つ以上の領域とを備える、請求項1に記載のカートリッジ。
- 疎水性材料及び親水性材料の少なくとも1対の連続帯をさらに備え、各対の連続帯は、前記検出室の幅にわたって延びている、請求項7に記載のカートリッジ。
- 前記流れ前方制御部は、前記検出室の前記対向面上のランド領域から突出し該ランド領域により分離される複数の離散構造体と、前記対向面の一部を占める疎水性材料の1つ以上の領域と、前記対向面の一部を占める親水性材料の1つ以上の領域とを備える、請求項1に記載のカートリッジ。
- 前記流れ前方制御部は、前記検出室の前記対向面における1つ以上の流路と、前記対向面の一部を占める疎水性材料の1つ以上の領域と、前記対向面の一部を占める親水性材料の1つ以上の領域とを備える、請求項1に記載のカートリッジ。
- 前記廃棄室内に位置する吸収材料をさらに備える、請求項1に記載のカートリッジ。
- 前記カートリッジは、前記検出室と前記廃棄室との間に位置する毛細管構造をさらに備える、請求項1に記載のカートリッジ。
- 開放時に前記ハウジングの前記内部容積を前記カートリッジ周りの周囲大気と流体連通させる弁をさらに備える、請求項1に記載のカートリッジ。
- 前記弁は前記廃棄室内に位置する、請求項13に記載のカートリッジ。
- 前記弁は閉鎖要素を備える、請求項13に記載のカートリッジ。
- 前記ハウジングに動作可能に接続された流体モニタをさらに備え、前記ハウジングの前記内部容積内に位置する液体が前記流体モニタにより検知可能となされる、請求項1に記載のカートリッジ。
- 内部容積を有するハウジングと、
該ハウジングに動作可能に取り付けられ、弾性表面波音響機械的センサを備えるセンサと、
前記ハウジングの前記内部容積内に位置し、検出面により画定される容積と、前記検出面から離間されて該検出面に対面し1つ以上の流路を備える対向面とを有する検出室と、
前記ハウジングの前記内部容積内に位置し、前記検出室と流体連通する廃棄室と、
前記廃棄室内に位置する吸収材料と、
前記検出室と前記廃棄室との間に位置する毛細管構造とを備える、検出カートリッジ。 - 内部容積を有するカートリッジハウジングと、
該カートリッジハウジングに動作可能に取り付けられ、検出面を有するセンサと、
前記カートリッジハウジングの前記内部容積内に位置し、前記検出面により画定される容積と、前記検出面から離間されて該検出面に対面し流れ前方制御部を備える対向面とを有する検出室と、
前記カートリッジハウジングの前記内部容積内に位置し、前記検出室と流体連通する廃棄室と、
各々が前記カートリッジハウジングの前記内部容積内に開放する1つ以上のモジュール口を通して前記カートリッジハウジングに取り付けられた出口を備える1つ以上の封止されたモジュールとを備え、各モジュールは、
出口と封止された内部容積とを有するモジュールハウジングと、
前記モジュールの前記出口上に位置する出口封止材と、
前記モジュールハウジングの前記内部容積内に位置するプランジャとをさらに備え、前記プランジャは、該プランジャが前記出口から離れている充填位置から、前記プランジャが前記出口に近接している放出位置まで移動可能であり、
前記プランジャが前記出口に向かって移動すると、前記モジュールハウジングの前記内部容積からの材料が前記カートリッジハウジングの前記内部容積内に前記出口を通って出ていくように、前記出口封止材を開放する、検出カートリッジ。 - 前記カートリッジハウジングの前記内部容積内に中間準備室をさらに備え、該中間準備室は前記検出室から上流側に位置し、前記モジュール口は前記中間準備室内に開放する、請求項18に記載のカートリッジ。
- 前記1つ以上の封止されたモジュールのうちの少なくとも1つのモジュールの前記内部容積は、
内部に液体を含む第1の室と、
前記モジュールハウジングの前記内部容積内に位置し、内部に試薬を含む第2の室と、
前記モジュールハウジング内で前記第1の室から前記第2の室を隔離する室間封止材とを備え、
前記第1の室、前記室間封止材、及び前記第2の室は、前記プランジャと前記出口封止材との間に位置し、
前記プランジャが前記出口に向かって移動すると、前記第1の室内の前記液体が前記第2の室内の前記試薬と接触するように、前記室間封止材を開放する、請求項18に記載のカートリッジ。 - 請求項1に記載の前記検出カートリッジを通して試料材料を移動する方法であって、
請求項1に記載の検出カートリッジを提供する工程と、
前記検出カートリッジの前記ハウジングの前記内部容積内に試料材料を送出する工程とを含み、前記試料材料が前記検出室に流入し、該検出室を通して前記廃棄室に向かう前記試料材料の流れ前方の進行が、前記検出室内の前記対向面上の前記流れ前方制御部により少なくとも部分的に制御される、試料材料を移動する方法。 - 試料材料を前記検出室内に送出する工程は、前記ハウジングの前記内部容積内に位置する中間準備室内に前記試料材料を送出する工程を含み、前記試料材料は前記中間準備室から前記検出室内に流出する、請求項21に記載の方法。
- 前記検出カートリッジは、前記廃棄室内に吸収材料をさらに備え、前記吸収材料は試料材料を前記廃棄室内に吸引する、請求項21に記載の方法。
- 前記検出室と前記廃棄室との間に位置する毛細管構造をさらに備え、前記毛細管構造は前記検出室から試料材料を吸引する、請求項21に記載の方法。
- 開放時に前記ハウジングの前記内部容積を前記カートリッジ周りの周囲大気と流体連通させる弁をさらに備え、前記方法は、前記検出室を通る試料材料流を制御するために前記弁を開放する工程を含む、請求項21に記載の方法。
- 前記弁を開放する工程は、前記検出室を通る試料材料流の流量を調整するために前記弁の大きさを調整する工程を含む、請求項25に記載の方法。
- 出口と封止された内部容積とを有するハウジングと、
前記出口上に位置する出口封止材と、
前記ハウジングの前記内部容積内に位置し、内部に液体を含む第1の室と、
前記ハウジングの前記内部容積内に位置し、内部に試薬を含む第2の室と、
前記ハウジング内で前記第1の室から前記第2の室を隔離する室間封止材と、
プランジャとを備え、前記第1の室、前記室間封止材、前記第2の室、及び前記出口封止材は、前記プランジャと前記出口との間に位置し、前記プランジャは、該プランジャが前記出口から離れている充填位置から、該プランジャが前記出口に近接している放出位置まで移動可能であり、
前記プランジャが前記出口に向かって移動すると、前記第1の室内の前記液体が前記第2の室内の前記試薬と接触するように、前記室間封止材を開放し、前記プランジャが前記放出位置内にさらに移動すると、前記ハウジングの前記内部容積からの前記液体及び前記試薬が前記出口を通って出ていくように、前記出口封止材を開放する、封止されたモジュール。 - 前記プランジャは先端部を有し、該先端部は、前記室間封止材と対面し、該室間封止材を突き破り、該室間封止材を開放する、請求項27に記載のモジュール。
- 前記第1の室及び前記第2の室は密閉封止された区間室を備える、請求項27に記載のモジュール。
- 前記プランジャは、該プランジャが前記充填位置にある際に前記第1の室の容積の一部を画定する、請求項27に記載のモジュール。
- 前記プランジャは、該プランジャが前記放出位置にある際に前記出口と噛み合う、請求項27に記載のモジュール。
- 前記第1の室内の前記液体は水を含み、前記第2の室内の前記試薬は、加水分解性材料を含む、請求項27に記載のモジュール。
- 請求項27に記載の封止されたモジュールを用いて材料を送出する方法であって、
請求項27に記載の封止されたモジュールを提供する工程と、
前記室間封止材を開放し、前記第1の室からの前記液体を前記第2の室内の前記試薬に接触させるために、前記プランジャを前記封止モジュールの前記出口に向かって移動する工程と、
前記出口封止材を開放し、前記ハウジングの前記内部容積からの前記液体及び前記試薬を前記出口を介して追い出すために、前記プランジャを前記出口に向かって移動する工程とを含む、材料を送出する方法。 - 前記プランジャは前記室間封止材を突き破る先端部を備える、請求項33に記載の方法。
- 出口と封止された内部容積とを有するハウジングと、
前記出口上に位置する出口封止材と、
前記ハウジングの前記内部容積内に位置し、内部に1つ以上の試薬を含む室と、
前記出口から離れた充填位置から前記出口に近接する放出位置まで移動可能なプランジャと、
前記室と流体連通する投入口とを備え、該投入口は、前記プランジャが前記充填位置にある際に前記プランジャと前記出口との間で前記室に進入し、
前記プランジャが前記出口に向かって動くと、前記ハウジングの前記内部容積からの材料が前記出口を通って出ていくように、前記出口封止材を開放する、モジュール。 - 前記投入口を閉じる封止材をさらに備える、請求項35に記載のモジュール。
- 前記プランジャは先端部を有し、該先端部は、前記出口封止材に対面し、該出口封止材を突き破り、該出口封止材を開放する、請求項35に記載のモジュール。
- 前記プランジャは、該プランジャが前記充填位置にある際に前記室の容積の一部を画定する、請求項35に記載のモジュール。
- 前記プランジャは、該プランジャが前記放出位置にある際に前記出口と噛み合う、請求項35に記載のモジュール。
- 請求項35に記載のモジュールを用いて材料を送出する方法であって、
請求項35に記載のモジュールを提供する工程と、
液体を含む試料材料を、前記投入口を通って前記室内に送出し、前記試料材料を前記室内に位置する前記試薬と接触させる工程と、
前記液体が前記出口を通って前記室から出ていくように、前記出口封止材を開放するために、前記プランジャを前記出口に向かって移動する工程とを含む、材料を送出する方法。
