Abstract
Description
本出願は、発明の名称「Dietary Supplement and Method of Processing Same」であり、2003年10月23日に出願された米国仮特許出願番号60/59,955に基づく優先権を主張する。上記の仮特許出願の開示は、その全体が本明細書中に参考として援用される。
本発明は、生物学的に活性な低分子を含有するカプセル化された組成物および、生物活性を維持(特に、サーチュイン(Sirtuin)1遺伝子を活性化(発現)または非活性化(サイレント)する能力を維持)するように組成物を処理する方法に関する。
したがって、上記に照らし、そして本発明が記載される場合に明らかとなる他の理由に関して、本発明の目的は、生物学的に活性なレスベラトロルを含有するカプセル化された組成物を提供し、そしてレスベラトロルが、消費者に摂取される場合に生物学的に活性なままであるように、この組成物をカプセル化する方法を提供することである。
本発明の1つの実施形態は、植物に由来するか、または合成的に生成される低分子を含有する組成物、および延長された期間、その生物活性を維持するようにこの組成物をカプセル化する方法に関する。植物の供給源または合成の供給源による生物学的に活性な低分子は、植物性低分子ポリフェノール(例えば、レスベラトロル、ケルセチン、フィセチン、ブテイン(butein)、ピセアトノール(piceatonol)、イソリキリチゲニン(isoliquiritigenin))を含み得、これらは、その小さいサイズおよび低分子量によって、細胞壁を通過し、細胞核に進入し、そして特定の遺伝子制御された機構を変化させ得る。好ましい植物性低分子ポリフェノールは、レスベラトロル(分子量228.25)であり、これは、他のポリフェノールより良好にサーチュイン−1制御性の酵素活性を活性化することが示された。しかし、他の低分子は、利用され得る。レスベラトロルは、合成的に生成されても、植物材料(ブドウおよびタデが挙げられる)のような供給源から天然に得られてもよい。好ましくは、レスベラトロルとしては、トランス立体異性体の分子(すなわち、トランス−3,4’,5トリヒドロキシスチルベン)が挙げられ、これは、以下:
Claims (28)
- 組成物をカプセル化するプロセスであって、以下:
(a)生物学的に活性な低分子を含む物質を得る工程;および
(b)実質的に酸素を含まない環境において生物学的に活性な低分子をカプセル化する工程、
を包含する、プロセス。 - 前記生物学的に活性な低分子が、レスベラトロルを含む、請求項1に記載のプロセス。
- 請求項2に記載のプロセスであって、(c)キレート剤、抗酸化剤、および乳化剤のうちの少なくとも1つと前記レスベラトロルとを混合する工程をさらに包含する、プロセス。
- 請求項2に記載のプロセスであって、ここで工程(a)が、以下:
(a.1)前記レスベラトロルを抽出するために溶媒を使用する工程であって、ここで該溶媒としては、水性溶媒、有機溶媒、およびそれらの混合物が挙げられる、工程、
を包含する、プロセス。 - 請求項2に記載のプロセスであって、ここで工程(b)が、以下:
(b.1)気密充填チャンバー中に頭部分および胴体部分を備えるカプセルを受容させる工程;
(b.2)生物学的に活性なレスベラトロルを含有する組成物で該胴体部分を少なくとも部分的に満たす工程;
(b.3)該頭部分を、該頭部分の位置が少なくとも部分的に重なるように該胴体部分に対して位置決めする工程;および
(b.4)該重なる部分に沿って流体気密シールを形成する工程、
を包含する、プロセス。 - 前記カプセル部分が、100μmについて約165cm3/m2/日未満の酸素透過度(ASTM D3985で測定した場合)を有する物質を含む、請求項5に記載のプロセス。
- 前記カプセル部分が、100μmについて約4cm3/m2/日未満の酸素透過度(ASTM D3985で測定した場合)を有する物質を含む、請求項5に記載のプロセス。
- 前記カプセル部分が、100μmについて約1cm3/m2/日未満の酸素透過度(ASTM D3985で測定した場合)を有する物質を含む、請求項5に記載のプロセス。
- 前記カプセル部分が、ゼラチン、ヒドロキシプロピルメチルセルロース、およびデンプンのうちの少なくとも1つを含有する、請求項5に記載のプロセス。
- 請求項5に記載のプロセスであって、ここで工程(b)が、以下:
(b.5)前記実質的に酸素を含まない環境を形成するために、前記充填チャンバー内に窒素ブランケットを形成する工程、
をさらに包含する、プロセス。 - 請求項4に記載のプロセスであって、ここで工程(b)が、以下:

(a.1)Vinis ViniferaおよびPolygonumからなる群より選択される天然の供給源から前記レスベラトロルを得る工程、
をさらに包含する、プロセス。 - 生物学的に活性な低分子、キレート剤、抗酸化剤、および乳化剤を含有するカプセル化された組成物であって、ここで該低分子が、実質的に酸素を含まない環境においてカプセル化される、組成物。
- 前記生物学的に活性な低分子が、レスベラトロルを含む、請求項13に記載のカプセル化された組成物。
- 請求項14に記載のカプセル化された組成物であって、以下:
前記キレート剤が、約5重量%〜約15重量%の量で存在し、
前記抗酸化剤が、約15重量%〜約35重量%の量で存在し、
前記乳化剤が、約15重量%〜約45重量%の量で存在し、そして
前記レスベラトロルが、約5重量%〜約30重量%の量で存在する、組成物。 - 請求項14に記載のカプセル化された組成物であって、以下:
前記キレート剤が、約7重量%〜約10重量%の量で存在し、
前記抗酸化剤が、約20重量%〜約30重量%の量で存在し、
前記乳化剤が、約25重量%〜約40重量%の量で存在し、そして
前記レスベラトロルが、約10重量%〜約20重量%の量で存在する、組成物。 - 前記キレート剤が、レスベラトロルの金属誘導性の酸化を遮断し得る化合物を含む、請求項14に記載のカプセル化された組成物。
- 前記キレート剤が、フィチン酸およびNDGAのうちの少なくとも1つを含む、請求項17に記載のカプセル化された組成物。
- 前記抗酸化剤が、フェノール性抗酸化剤を含む、請求項14に記載のカプセル化された組成物。
- 前記フェノール性抗酸化剤が、ケルセチンを含む、請求項19に記載のカプセル化された組成物。
- 前記乳化剤が、レシチン(ホスファチジルコリン)を含む、請求項14に記載のカプセル化された組成物。
- 前記レスベラトロルが、天然の供給源に由来する、請求項14に記載のカプセル化された組成物。
- 前記生物活性が、サーチュイン1酵素の活性化を含む、請求項14に記載のカプセル化された組成物。
- 栄養補助サプリメントであって、以下
(a)生物学的に活性なレスベラトロル物質を得るプロセス;
(b)実質的に酸素を含まない環境を備える充填チャンバー中に頭部分および胴体部分を備えるカプセルを受容させるプロセス;
(c)該生物学的に活性なレスベラトロル物質で該胴体部分を少なくとも部分的に満たすプロセス;
(d)該頭部分を、該頭部分の位置が少なくとも部分的に重なるように該胴体部分に対して位置決めするプロセス;および
(e)該重なる部分に沿って流体気密シールを形成するプロセス、
によって生成される、栄養補助サプリメント。 - 生物学的に活性な形態のレスベラトロルを含有するカプセル化された組成物を含む栄養補助サプリメントであって、ここで該レスベラトロルが、カプセル化後に少なくとも4ヶ月間、生物活性を維持する、栄養補助サプリメント。
- 前記生物活性が、サーチュイン1酵素活性を刺激することを含む、請求項25に記載の栄養補助サプリメント。
- 生物学的に活性な低分子、ならびにキレート剤、抗酸化剤、および乳化剤のうちの少なくとも1つを含有するカプセル化された組成物であって、ここで該低分子が、細胞壁を通過し、細胞核に進入し、そして該核内にある遺伝子制御された酵素を変化させ得る、組成物。
- 前記遺伝子制御された酵素が、サーチュイン1酵素である、請求項27に記載のカプセル化された組成物。
