Abstract
【解決手段】チョウジエキス及びボタンエキスからなる群から選択される少なくとも1種の植物エキスを有効成分とする発毛促進剤。

Description
チョウジエキスを調製するために使用するチョウジは、特に限定されないが、医薬部外品に使用が認められているという理由から、チョウジ(学名:Syzygium aromaticum)を使用することが好ましい。
ボタンエキスを調製するために使用するボタンは、特に限定されないが、医薬部外品に使用が認められているという理由から、ボタン(学名:Paeonia suffruticosa)を使用することが好ましい。
本発明に関するチョウジ、ボタンの抽出物(エキス)を作製する方法としては、抽出工程及び分離工程を組み合わせる方法、上記方法に更に分画工程を組み合わせる方法等があげられるが、これらに限定されない。
本発明の発毛促進剤は、前記チョウジエキス及びボタンエキスからなる群から選択される少なくとも1種の植物エキスを有効成分とするものである。本発明の発毛促進剤は、特に、前記チョウジエキスを有効成分として含有することでVEGF遺伝子発現量増加効果を発揮するVEGF遺伝子発現量増加剤として用いることができる。本発明の発毛促進剤は、特に、前記チョウジエキスを有効成分として含有することでFGF−7遺伝子発現量増加効果を発揮するFGF−7遺伝子発現量増加剤として用いることができる。本発明の発毛促進剤は、特に、前記ボタンエキスを有効成分として含有することでVEGF産生促進効果を発揮するVEGF産生促進剤として用いることができる。
本発明のVEGF(血管内皮増殖因子、vascular endothelial growth factor)遺伝子発現量増加剤は、前記チョウジエキスを有効成分として含有するものである。
本発明のVEGF産生促進剤は、前記ボタンエキスを有効成分として含有するものである。
本発明のFGF−7(発毛因子、fibroblast growth factor 7)遺伝子発現量増加剤は、前記チョウジエキスを有効成分として含有するものである。
本発明の発毛促進剤(医薬品の形態を含む)(VEGF遺伝子発現量増加剤、VEGF産生促進剤、FGF−7遺伝子発現量増加剤)は、その形態は特に制限されず、非経口投与形態及び経口投与形態のいずれもが含まれる。
チョウジエキスのVEGF遺伝子発現量増加及びボタンエキスのVEGF産生促進は、ヒト頭髪毛乳頭細胞(HFDPC)を使用して、評価することができる。
以下の手順で、毛乳頭細胞によって培養液中に分泌されたVEGFをELISAにより測定する。
以下の手順で、毛乳頭細胞からtotal RNAを抽出し、VEGFの遺伝子発現解析を行う。
3.毛乳頭細胞でのFGF−7の遺伝子発現の解析
以下の手順で、毛乳頭細胞からtotal RNAを抽出し、FGF−7の遺伝子発現解析を行う。
チョウジ(つぼみ:乾燥物の植物体として10g)を、細かく刻み、その粉砕物に、10倍量の含水エタノール(100mL)を加えて、2日間静置し、エキス(抽出液)を得た。
VEGF産生量及びVEGF遺伝子発現量を、ヒト頭髪毛乳頭細胞(HFDPC)(東洋紡績株式会社製)を使用して、評価した。
以下の手順で、毛乳頭細胞によって培養液中に分泌されたVEGFをELISAにより測定した。
以下の手順で、毛乳頭細胞からtotal RNAを抽出し、VEGFの遺伝子発現解析を行った。
上記「2.毛乳頭細胞でのVEGFの遺伝子発現の解析」と同様に、毛乳頭細胞からtotal RNAを抽出し、FGF-7遺伝子発現解析も行った。FGF-7の発現誘導剤としてアデノシンを100μM(最終濃度)で使用した。これまで、FGF-7は、毛乳頭細胞から産生され、毛母細胞に作用し、毛母細胞の増殖、分裂を促すことで毛髪成長をさせること、そして、アデノシンが毛乳頭細胞表面の受容体に直接作用し、FGF-7(fibroblast growth factor 7、発毛因子)の産生を高めることが、報告されている。
更に、以下の手順で、5-α-リダクターゼの活性測定を行った。
毛乳頭細胞が産生したVEGF量、VEGFの遺伝子発現量、FGF-7の遺伝子発現量及び5-α-リダクターゼ活性抑制程度について、有意差検定を行い、下記の基準に従って評価した。
○:有意に効果有り(p<0.05)
△:効果有り(cut off値以上、ポジティブコントロールの10%以上の効果有り)
−:効果なし
N.D.:未測定又は評価不能
試験結果を下記表3及び4に示した。
Claims (2)
- チョウジエキス及びボタンエキスからなる群から選択される少なくとも1種の植物エキスを有効成分とする発毛促進剤。
- 前記植物エキスが、チョウジ及びボタンからなる群から選択される少なくとも1種の植物体の水、エタノール及び1,3−ブチレングリコールよりなる群から選ばれる少なくとも1種の抽出溶媒で抽出されたものである、請求項1に記載の発毛促進剤。
