Description
従来技術の問題は、単一ウェル又はマルチウェルの試料調製用デバイス、試料をMALDI TOF質量分析による更なる分析の前に脱塩して濃縮する方法、及び該デバイスを形成する方法を提供する本発明によって克服された。例えば、本発明の一つの具体例によるデバイスは、複数のウェルを含み、各ウェルは、それぞれの三次元膜構造を含む出口又は排水口と流体連絡しており、該三次元膜構造は、好ましくは、固相抽出を実施できるデバイスを形成するように該出口内に固定された多孔性ポリマーマトリクス中に捕捉された複数の吸着性粒子を含む。本発明は又、試料調製用デバイスを形成する方法、及び本発明のデバイスを利用する試料調製方法にも向けられている。

用語「膜」は、ここで用いる場合、所望の適用に適した多孔度を有する透過性及び半透性の三次元構造(粒子を含むもの又は含まないもの)を包含する。用語「複合体構造」は、ここで用いる場合、充填膜を包含する。
50fモルの大腸菌β−ガラクトシダーゼ(B−gal)トリプシン処理したペプチドを含む100μlの試料を、127mmHg(5インチHg)の真空を利用する単一通過にて、本発明の密な複合体C18膜(二側面冷却により形成したもの)上に積載した。結合したペプチドを、100μlの0.1%(v/v)トリフルオロ酢酸で、再び真空を利用して洗う。次いで、これらのペプチドを、1.5μlの50%アセトニトリル(10mg/mlのα−シアノ−4−ヒドロキシ−桂皮酸を含むDIW中)にて溶出させた。この溶離液を、次いで、MALDI TOF MS標的上にスポットして、Bruker Daltronics Auto Flex MALDI TOF MSにより分析した。10種類のペプチドが、図5Aに示したように、データベース検索によって、陽性に同定された。

Claims (18)
- ハウジング内に複合体構造を形成する方法であって、該方法は、下記:
少なくとも第一及び第二の開口を有する該ハウジングを用意し、該第一の開口は、該第二の開口から離れており;
該ハウジング内に予め決めた量の、膜形成ポリマーを含むキャスト用溶液を分配し;
該キャスト用溶液を、該ポリマーが不溶性である冷却(quenching) 流体に、該冷却流体が該ポリマーを該ハウジング内で沈殿させるために該第一及び第二の開口の各々に同時に入るように、曝す
ことを含み、
該沈殿したポリマーにより形成された複合体構造が、約10未満のアスペクト比を有し、沈殿したポリマーにより形成された複合体構造が、0.703kg/cm 2 ( 10psi ) で該複合体構造を通る空気流が51リットル/分/cm 2 未満である密度を有し且つ多孔性ポリマーマトリクス中に捕捉された複数の吸着性粒子を含む三次元構造を含む当該方法。 - 前記の膜形成ポリマー中に複数の吸着性粒子を与えることを更に含む、請求項1に記載の方法。
- 前記の冷却流体が、前記の開口の各々に等圧的に入る、請求項1に記載の方法。
- 前記の冷却流体が、アルコールと水を含む、請求項1に記載の方法。

- 前記の複合体構造を通る空気流が、0.703kg/cm 2 ( 10psi )で約23リットル/分/cm2である、請求項7に記載の試料調製用デバイス。
- 前記のハウジングが、マルチウェルプレートのウェルの排水口である、請求項7に記載の試料調製用デバイス。
- 前記のハウジングが、前記の複合体構造を支持する少なくとも一つの支持エレメントを含む、請求項7に記載の試料調製用デバイス。
- 試料を予め決めた時間にわたって三次元構造を多数回通過させた後に該試料が予め決めた程度まで精製されるように溶質を捕捉するための、多孔性ポリマーマトリクス中に捕捉された複数の吸着性粒子を含み且つ約10より小さいアスペクト比を有する三次元構造を含む型の試料調製用デバイスであって、下記構成を含む請求項1の方法により形成されたデバイス:
該デバイス中に該三次元構造を、駆動力を加えた際に、該試料が、該試料を一回だけ該構造を予め決めた時間にわたって通過させた後に、少なくとも該予め決めた程度まで精製されるように、十分な密度を有するように形成された構成。 - 前記のデバイスが、複数の開口を含み、各開口が、開いた上端と開いた底部を有し、前記の三次元構造が、該開口の各々において形成される、請求項11に記載の試料調製用デバイス。
- 前記の開口の各々が、多数の孔よりなる、請求項12に記載の試料調製用デバイス。
- 開いた上端が、開いた底部より一層大きい、請求項12に記載の試料調製用デバイス。
- 前記の開口が、前記の構造を支持するための少なくとも一つの支持エレメントを含む、請求項11に記載の試料調製用デバイス。

多孔性ポリマーマトリクス中に捕捉された複数の吸着性粒子を含み且つ約10より小さいアスペクト比を有し且つ、該試料を、予め決めた時間にわたって該構造を一回だけ通過させることにより、予め決めた程度まで精製するのに十分な密度を有する三次元構造を用意する。 - 前記のハウジングが、ピペットチップである、請求項7に記載の試料調製用デバイス。
