Description
インビトロで展開するためのプロトタイプPGSモジュールを構築し、慢性前立腺炎及び他のUTIのために一般に処方されるフルオロキノロンであるシプロフロキサシンを用いて薬物放出動態について試験した。プロトタイプモジュール900を図9に示す。プロトタイプモジュール900は、形状が矩形であり、直径300μmの薬物ロッド902を収容するための内部円柱状コアを有し、1つの100μmの放出オリフィス904を含んでいた。モジュールは、ワイヤを張り渡したアルミニウム鋳型内にPGSプレポリマーを融解させ、その後、加熱真空下で48時間重合反応させることによって形成した。PGS鋳造物は、鋳造物の頂部表面上の選択位置に、レーザーマイクロアブレーションプロセスによりオリフィスをあけた際に、鋳型内に残存しており、オリフィスは、埋め込まれた直径300μmの長手方向のワイヤまで突き抜けた。
放出動態をインビトロで測定するために、シプロフロキサシンを装填したプロトタイプPGSデバイスを、実施例1に記載したように作製し、ガラスバイアルの内部に取り付け、脱イオン水2mL中に浸漬した。周囲の媒質中のシプロフロキサシン−HCl(CIP)濃度の時点測定を、CIPのために開発した定量的なHPLC−UV検出法を使用して8〜10日の期間にわたっておよそ12時間毎に行った。
ウサギ(2.7kg、ニュージーランド白色、雄性)の精嚢内に移植した非吸収性シリコーンデバイスを用いてパイロットインビボ実験を行なった。使用した薬物はリドカインであり、総装填量は2mgであった。この実験は、デバイスが、男性の精嚢などの膀胱以外の位置に移植される場合の状況をシミュレートするために設計した。ウサギ実験用の非吸収性デバイスを図13に示し、経時的なリドカイン血漿濃度を図14に示す。
鋳造法を使用することによって、約7.62cmの長さ、直径が約330μmの中空リザーバ、デバイスの第1の端部から約2.81cmに位置した第1のオリフィス、及びデバイスの第2の端部から約2.81cmに位置した第2のオリフィスを有するデバイス本体を作製した。オリフィスの直径は約100μmであった。そのようなデバイスは、鋼ワイヤを埋め込んだ鋳型中にPGSをキャストすることによって形成した。鋳型の長さは約7.62cmであった。いくつかの鋼ワイヤを、鋳型を通してその長さに沿って張り渡した。各ワイヤの直径は約330μmであった。PGSをキャストした後、デバイス本体中にオリフィスをレーザーであけた。PGSを鋳型から取り出し、切断して個々のデバイス本体にした。
薬物ロッドを作製し、薬物ロッドを送達デバイス本体、又は収納容器に付随させる方法を試験した。
Claims (7)
- 生体吸収性エラストマー製であり、少なくとも1つの開口部を有する収納容器と、
前記収納容器に入れられた薬物と、
を備える、患者の尿生殖器部位内に配置するための浸透圧ポンプデバイスであって、
前記少なくとも1つの開口部を通じて、浸透圧によって推進されて、インビボで前記薬物を分配するように構成されており、
当該デバイスは患者の尿生殖器部位内で当該デバイスを保持するための弾性保持フレームを有し、
前記弾性保持フレームは、フープ、コイル、バネ、2次元らせん状、又は3次元らせん状形状を備える、保持のための形状を有する、
デバイス。 - 前記生体吸収性エラストマーがポリ(グリセロール−セバシン酸)を含む、請求項1に記載の浸透圧ポンプデバイス。
- 前記薬物が固体フォーム又は半固体フォームである、請求項1又は2に記載の浸透圧ポンプデバイス。
- 前記収納容器が、水に対して選択的に浸透性であり、前記薬物に対して不浸透性である側壁を備える、請求項1に記載の浸透圧ポンプデバイス。
- 当該デバイスが、当該デバイスを、尿道カテーテルによって前記患者の膀胱に挿入し、その中で保持するのに適した寸法とされており、弾性を有する、請求項1に記載の浸透圧ポンプデバイス。
- 挿入のためのより低レベルの輪郭形状、及び膀胱内に保持するためのより高度な輪郭形状をとるよう、形状を変化させるよう構成されている、請求項5に記載の浸透圧ポンプデバイス。
- 前記薬物が、抗生物質、免疫抑制剤、抗炎症剤、化学療法剤、麻酔薬、又はこれらの組合せを含む、請求項1ないし6のいずれか1項に記載の浸透圧ポンプデバイス。
