Abstract

PURPOSE:To provide a hard gelatin capsule to be filled with an oily liquid agent to form a capsule preparation almost free from the leakage of the liquid from the fitting part. CONSTITUTION:The fitting part of a capsule cap 1 and a capsule cap 2 constituting the objective hard gelatin capsule has a notch mechanism consisting of notches 3 having a diameter of about 1.2-2mm and a depth (height) of about 0.2-0.4mm and formed with nearly equal spacings at >=6 parts on the circumference of the fitting part of the capsule cap 1 or the capsule cap 1 and the capsule cap 2 in the circumferential direction. The rigidity of the fitting part is increased, the roundness of the opening of the fitting part is improved and a close fitting is ensured by the >=6 notches formed on the fitting part. Accordingly, the leakage of liquid from the fitting part of the capsule is almost completely prevented without sealing the fitting part with a viscous gelatin liquid.

Description

【発明の詳細な説明】 〔産業上の利用分野〕本発明は、油性液状物の充填用カ
プセルとして液漏れのないカプセル剤を製造することが
できる硬ゼラチンカプセルに関する。 〔従来の技術〕カプセル剤の製造に使用されるカプセル
は、軟ゼラチンカプセル(以下、「軟カプセル」)と、
硬ゼラチンカプセル(以下、「硬カプセル」)に大別さ
れる。油性液状物の充填には専ら軟カプセルが使用さ
れ、硬カプセルは主として、粉状ないし塊状等の固形の
充填物に対して使用されている。硬カプセルは、第2図
に示すようにカプセルキヤツプ1とカプセルボデイ2と
からなる。医薬品には、00号〜5号(号数字が大きい
程、カプセルサイズは小さい)の7種のサイズのものが
使用されている。その硬カプセルは、キヤツプ1とボデ
イ2との嵌合部の離脱防止のためのロツク機構として、
キヤツプ1の嵌合部周面に、直径方向に向い合う2個所
に微小円形状のノツチ3,3が形成されている。なお、
キヤツプ1とボデイ2の嵌め合い深さを規定し両者のず
れを防止するために、キヤツプ1には内側に突出する円
周突起4が、ボデイ2にはそれに対応する円周溝5がそ
れぞれ所定の位置に形成されている。硬カプセルは、粉
状・塊状等の固形物の充填のほか、油性液状物の充填に
使用することもむろん可能ではあるが、その場合は、嵌
合部からの液漏れを防止するために、上記のノツチに代
え、そのシール手段として嵌合部の外周にゼラチン粘稠
液が塗布される。 〔発明が解決しようとする課題〕軟カプセルは、油性液
状物を完全な密封状態に充填することができるが、硬カ
プセルに比べ、充填量の偏差が大きい、充填速度、濃度
(粘度)、液温度の影響を受け易い、製品同士が癒着し
易い、等の難点があり、またその充填には特殊な技術と
高価な機器装置を必要とする。他方、硬カプセルは、取
扱いが容易であり、そのカプセル剤の製造には、軟カプ
セルを使用する場合のような特殊な技術や設備を必要と
しないが、油性液状物を充填する場合には、カプセルの
1個ずつにその嵌合部の液漏れ防止としてゼラチン粘稠
液を塗布するという煩瑣なシール工程が必要となり、工
業的生産に不向きである。そこで、本発明は、油性液状
物の充填に硬カプセルを使用し、嵌合部にゼラチン粘稠
液を塗布するシール工程を必要とすることなく、液漏れ
をほゞ完全に防止することができる改良された硬カプセ
ルを提供するものである。 〔課題を解決するための手段および作用〕本発明の硬カ
プセルは、カプセルキヤツプとカプセルボデイの嵌合部
のロツク機構として、カプセルキヤツプ、またはカプセ
ルキヤツプとカプセルボデイの嵌合部周面の円周方向の
少くとも6個所にほゞ等間隔をおいてノツチが形成され
ていることを特徴としている。第1図は本発明の硬カプ
セルの実施例を示している。図の例では、キヤツプ1
に、ノツチ3,3,…が円周方向にほゞ等間隔をなす6
個所に形成され、ボデイ2には、そのキヤツプ1の6個
のノツチ3,3,…に対応するノツチ3,3,…が円周
方向にほゞ等間隔をおいて6個形成されている。本発明
の硬カプセルは、そのキヤツプ1とボデイ2との嵌合部
に6個以上のノツチが形成されている点を除き、その材
質、形状・サイズ、肉厚、キヤツプとボデイの嵌合部ク
リアランス、あるいはキヤツプの円周突起4,ボデイの
円周溝5の形設態様等は、従来より粉末ないし塊状物等
の充填に使用されている2ノツチ型硬カプセルのそれと
何ら異ならず、また改変を加える必要もない。本発明の
硬カプセルにおけるキヤツプおよびボデイのノツチ形設
態様も、そのノツチ個数が6個以上である点を除けば、
従来の2ノツチ型硬カプセルと異ならず、そのサイズは
カプセルの大きさに応じて適宜設定すればよく、ノツチ
の直径は約1.2〜2.2mm程度、深さ(高さ)は約
0.2〜0.4mm程度としてよい。本発明の硬カプセ
ルのサイズを、医薬品の充填に使用されている各種号サ
イズ(00号〜5号)のうち、例えば00号サイズと
し、これに6個のノツチを形成する場合、キヤツプにお
ける隣り合うノツチの中心点間距離(周面上)は約4.
6mm,ボデイにおける隣り合うノツチの中心点間距離
は約4.5mm、1号サイズのカプセルでは、キヤツプ
のノツチ中心点間距離は約4.0mm、ボデイのノツチ
中心点間距離は約3.9mmであり、また5号サイズの
カプセルでは、キヤツプのノツチ中心点間距離は約2.
7mmで、ボデイのノツチ中心点間距離は約2.6mm
となる。上記説明では、カプセルのキヤツプ1およびボ
デイ2の双方にノツチを形成することとしているが、必
ずしもそうである必要はなく、キヤツプ1にのみ6個以
上のノツチを形成し、ボデイ2のノツチ形成は省略する
こともできる。本発明の硬カプセルを製造するには、成
形原型として、6個以上のノツチ形成部分を設けたもの
を使用する点を除いて、従来の2ノツチ型硬カプセルの
製造装置をそのまゝ使用すればよく、特別の機器の使用
や工程の付加を必要とせず、2ノツチ型の硬カプセルと
全く同様に連続的に製造することができる。また、本発
明の硬カプセルの保管および内容物の充填時の取扱い等
に何ら特別の処置は必要としない。本発明の硬カプセル
は、そのキヤツプとボデイとの嵌合部に6個以上のノツ
チが形成されていることにより、嵌合部の剛性が高く、
また2ノツチ型のものと異なつて嵌合部の開口形状の真
円度が高く良好な密着性が付与されているので、中鎖の
直鎖脂肪酸であるトリグリセライド等に代表される油性
液状物の充填用カプセルとして使用することができ、ゼ
ラチン粘稠液によるシールを施すことなく、嵌合部から
の液漏れをほゞ完全に防止したカプセル剤を製造するこ
とができる。 〔実施例〕カプセル カプセルのキヤツプ1とボデイ2のそれぞれの嵌合部の
周面に、第1図に示したように6個の円形ノツチを等間
隔に形成した硬ゼラチンカプセル カプセルサイズ:2号サイズ(キヤツプ嵌合部内径
6.8mm,ボデイ嵌合部外径 6.6mm,クリアラ
ンス 0.2mm) ノツチサイズ:直径約1.6mm,隣り合うノツチの中
心点間距離:キヤツプ上約3.6mm,ボデイ上約3.
5mm。カプセル充填物 トリグリセライドに微量の親油性薬剤を添加して調製し
た油性液状組成物 上記硬ゼラチンカプセルに上記油性液状組成物200m

記と同じ)を使用して上記油性液状組成物200mgを
充填した硬ゼラチンカプセル剤(カプセル剤B)、およ
びノツチを有しない硬ゼラチンカプセル(カプセルサイ
ズは上記と同じ)を使用し上記油性液状組成物200m
gを充填したうえ、嵌合部外周をゼラチン粘稠液でシー
ルしたカプセル剤(カプセル剤C)を、それぞれ製剤し
た。上記各供試カプセル剤A,B,Cの嵌合部からの液
漏れ発生率を下表に示す。 〔発明の効果〕本発明の硬ゼラチンカプセルは、油性液
状物の充填に使用して嵌合部からの液漏れがほゞ完全に
防止されたカプセル剤を製造することができる。本発明
の硬ゼラチンカプセルは、その成形原型として6個所の
ノツチ形成部分を設けたものを使用する以外は、従来の
2ノツチ型硬ゼラチンカプセルと同様の機器および工程
により安価に連続生産することができ、また油性液状物
の充填工程では、嵌合部にゼラチン粘稠液を塗布する煩
瑣なシール工程を必要とせず、油性液状物充填カプセル
剤を効率よく製剤することができる。硬ゼラチンカプセ
ルは、軟ゼラチンカプセルに比べ取扱い性が良く、かつ
そのカプセル剤の製造に特殊な技術や機器装置を必要と
せず、コスト的にも極めて有利である。また油性液状物
充填用カプセルとして軟ゼラチンカプセルに代え、本発
明の硬ゼラチンカプセルを使用することにより、軟ゼラ
チンカプセルを使用する場合の種々の不利を回避するこ
とができる。
