Claims (1)
- 【特許請求の範囲】 1.アッセイ産物をコードするレポーター遺伝子に有効に連結した、骨形成タン パク質をコードする遺伝子由来のプロモーターを含む発現ベクターを含む、変換 された組換え宿主細胞を含むことを特徴とする骨形成剤を同定するシステム。 2.当該骨形成タンパク質がBMP-2及びBMP-4タンパク質から成る群から選 択される請求項1に記載のシステム。 3.当該レポーター遺伝子が、ホタルルシフェラーゼ、クロラムフェニコールア セチルトランスフェラーゼ、β−ガラクトシダーゼ、グリーンフルオレセンタン パク質、ヒト成長ホルモン、アルカリフォスファターゼ及びβ−グルクロニダー ゼからなる群から選択されるアッセイ産物をコードする遺伝子を含む請求項1又 は2に記載のシステム。 4.当該レポーター遺伝子が、ホタルルシフェラーゼ産物をコードする遺伝子を 含む請求項3に記載のシステム。 5.当該レポーター遺伝子からの当該アッセイ産物の発現を許容する条件下、請 求項1−4の何れかに記載の細胞を培養し; 当該細胞を骨形成活性を有する可能性のある少なくとも1つの候補化合物と接触 させ; 当該候補化合物の存在下に産生されるアッセイ産物の量を測定し、及び当該量と 当該候補化合物の非存在下に産生されるアッセイ産物の量とを比較し;及び 存在するときの当該アッセイ産物の量を増加させる候補化合物を骨形成化合物と して同定する; 以上のステップを含むことを特徴とする骨形成化合物を同定する方法。 6.BMP-2及びBMP-4タンパク質からなる群から選択される骨形成タンパク 質をコードする遺伝子のプロモーター領域をコードする核酸配列を含むことを特 徴とする単離された核酸分子。 7.図1C(配列ID NO:3)に図示されたBMP-4遺伝子の-2372 から+316部位、生物的に活性なプロモーターをコードするその部位、図11に図 示されたBMP-2配列、及び生物的に活性なプロモーターをコードするその部位 からなる群から選択される核酸配列に対応する請求項6に記載の核酸分子。 8.請求項6又は7に記載の核酸配列を含むことを特徴とする組換え発現ベクタ ー。 9.当該核酸配列がアッセイ産物をコードするレポーター遺伝子に有効に連結す る請求項8に記載の組換え発現ベクター。 10.当該レポーター遺伝子が、ホタルルシフェラーゼ、クロラムフェニコール アセチルトランスフェラーゼ、β-ガラクトシダーゼ、グリーンフルオレセンタ ンパク質、ヒト成長ホルモン、アルカリフォスファターゼ、及びβ-グルクロニ ダーゼからなる群から選択されるアッセイ産物をコードする遺伝子を含む請求項 9に記載の組換え発現ベクター。
