Claims (1)
- 【特許請求の範囲】 1.植物細胞において下流に位置する異種DNAセグメントの根皮層に特異的 な転写に関与する単離されたDNA分子であって、 (a)本願明細書に記載の配列番号1、配列番号2、配列番号3、配列 番号4、配列番号5、配列番号6、配列番号7、配列番号8および配列番号9と 、 (b)60℃にて、0.3MのNaCl、0.03Mのクエン酸ナトリウ ム、0.1%のSDSという洗浄ストリンジェンシー(厳密性)で表される条件 で、上述の(a)のうちの一配列を有する単離されたDNAにハイブリダイゼー ションし、かつ、植物細胞において下流に位置する異種DNAセグメントの根皮 層に特異的な転写に関与するDNA配列と から成る群から選択される配列を有する前記単離されたDNA分子。 2.発現カセットを含むDNA構成物であって、5'から3'方向に、Tobacco RD2プロモーターと、前記プロモーターから下流に位置し、かつ前記プロモー ターと機能的に関連した異種DNAセグメントとを含む構成物。 3.発現カセットを含むDNA構成物であって、前記構成物は5'から3'方向 に、根皮層に特異的なプロモーターと、前記プロモーターから下流に位置し、か つ前記プロモーターと機能的に関連した異種DNAセグメントとを含み、前記根 皮層に特異的なプロモーターが、 (a)本願明細書に記載の配列番号1、配列番号2、配列番号3、配列 番号4、配列番号5、配列番号6、配列番号7、配列番号8および配列番号9と 、 (b)60℃にて、0.3MのNaCl、0.03Mのクエン酸ナトリウ ム、0.1%のSDSという洗浄ストリンジェンシーで表される条件で、上述の (a)のうちの一配列を有する単離されたDNAにハイブリダイゼーションし、 植物細胞において下流に位置する異種DNAセグメントの根皮層に特異的な転写 に関与 するDNA配列と から成る群から選択される配列を有することを特徴とする構成物。 4.前記構成物はプラスミドをさらに含む請求項3に記載のDNA構成物。 5.前記異種DNAセグメントが、殺虫性タンパク質をコードする遺伝子であ る請求項3に記載のDNA構成物。 6.前記異種DNAセグメントが、昆虫に有毒なバチルス・スリンギエンシス (Bacillus thuringiensis)結晶タンパク質をコードする遺伝子である請求項4 に記載のDNA構成物。 7.請求項3に記載のDNA構成物を含有する植物細胞。 8.請求項7に記載の植物細胞から植物を再生するステップを含む形質転換植 物を作成する方法。 9.請求項3に記載のDNA構成物を含有するアグロバクテリウム・チュメフ ァシエンス(Agrobacterium tumefaciens)の細胞であって、前記DNA構成物は Tiプラスミドをさらに含むことを特徴とするAgrobacterium tumefaciensの細 胞。 10.請求項9に記載のAgrobacterium tumefaciensに植物細胞を感染させて 、形質転換した植物細胞を作成するステップと、その後、前記形質転換した植物 細胞から植物を再生させるステップとを含んでなる形質転換植物を作成する方法 。 11.請求項3に記載のDNA構成物を担持する微粒子であって、植物細胞の 発射注入による形質転換に適する前記微粒子。 12.請求項11に記載の微粒子を植物細胞内に撃ち込んで、形質転換した植 物細胞を生成するステップと、その後、前記形質転換した植物細胞から植物を再 生させるステップとを含んでなる形質転換植物を作成する方法。 13.請求項3に記載のDNA構成物を含有する植物細胞プロトプラスト。 14.請求項13に記載の植物細胞プロトプラストから植物を再生させるステ ップを含む形質転換植物を作成する方法。 15.形質転換した植物細胞を含む形質転換植物であって、前記形質転換した 植物細胞は異種DNA構成物を含み、前記DNA構成物は5'から3'方向に根皮 層に特異的なプロモーターと、前記プロモーターから下流に位置し、かつ前記プ ロモーターと機能的に関連した異種DNAセグメントとを含み、前記プロモータ ーは前記異種DNAセグメントの根皮層に特異的な転写に関与することを特徴と する形質転換植物。 16.前記根皮層に特異的なプロモーターが、植物細胞において下流に位置す る異種DNAセグメントの根皮層に特異的な転写に関与するTobacco RD2プロ モーターである請求項15に記載の形質転換植物。 17.前記プロモーターが、 (a)本願明細書に記載の配列番号1、配列番号2、配列番号3、配列 番号4、配列番号5、配列番号6、配列番号7、配列番号8および配列番号9と 、 (b)60℃にて、0.3MのNaCl、0.03Mのクエン酸ナトリウ ム、0.1%のSDSという洗浄ストリンジェンシーで表される条件で、上述の (a)のうちの一配列を有する単離されたDNAにハイブリダイゼーションし、 植物細胞において下流に位置する異種DNAセグメントの根皮層に特異的な転写 に関与するDNA配列と から成る群から選択される配列を有することを特徴とする請求項15に記載の形 質転換植物。 18.前記植物は双子葉植物である請求項15に記載の形質転換植物。 19.前記植物は単子葉植物である請求項15に記載の形質転換植物。 20.前記植物はタバコ(Nicotiana tabacum)である請求項15に記載の形 質転換植物。 21.植物細胞において下流に位置する異種DNAセグメントの根皮層に特異 的な転写に関与するプロモーターから実質的に成り、かつ本願明細書に記載の配 列番号1〜9から成る群から選択される配列を有する単離されたDNA分子。 22.発現カセットを含むDNA構成物であって、5'から3'の方向に、請求 項21に記載のプロモーターと、前記プロモーターから下流に位置し、かつ前記 プロモーターと機能的に関連した異種DNAセグメントとを含むDNA構成物。 23.形質転換した植物細胞を含む形質転換植物であって、前記形質転換した 植物細胞は請求項22に記載のDNA構成物を含有する形質転換植物。
