Description

본 발명은 아크릴로일 치환된 피롤 유도체, 이의 제조방법 및 이를 함유하는 약제학적 조성물에 관한 것이다. 본 발명의 피롤 유도체는 하기구조식을 갖는 공지된 화학물인 디스타마이신 A의 유도체로 여겨진다:

본 발명은 하기 일반식(I)의 아크릴로일 치환된 피롤 유도체를 제공한다:
상기식에서, n은 1내지 5의 정수이고:

일반식(I)의 화합물은
A) 하기 일반식(Ⅱ)의 화합물 또는 이의 염을 일반식(Ⅲ)의 화합물과 반응시켜 일반식(I)의 화합물 또는 이의 염을 수득하거나;
B) 하기 일반식(Ⅳ)의 화합물 또는 이의 염을 하기일반식(Ⅴ)의 화합물과 반응시켜, A가 그룹또는인 일반식(I)의 화합물 또는 이의 염을 수득함을 특징으로 하는 방법에 의해 제조된다;
상기식에서,
R1, R2, R3, R4, A, B,Het 및 n은 상기 정의한 바와 같으며;
X는 하이드록시 또는 이탈 그룹이고;
Z은또는 H2N-Het-CO-이다.

예를들어 본 발명 화합물의 세포독성은 하기의 방법을 사용하여 뮤린 L1210 백혁병 세포상에 대해 시험되었다. 생체내 종양으로부터 세포를 추출하여 세포배양을 확립한다. 세포는 10회 계배양까지 사용된다. 세포독성은 4시간 약제 처리 및 약제 없는 배지에서 48시간 동안 성장시켜 생존한 세포의 수를 측정함으로써 측정한다. 처리된 배양물내에서 세포 성장의 백분율을 대조군의 백분율과 비교한다. ID50값(대조군에 대하여, 세포성장의 50%를 억제하는 투여량)은 투여량-반응곡선상에서 계산한다.

무수 DMF (10ml)중의 0℃로 냉각시킨, (E)--(p-메톡시벤조일)--브로모-아크릴산(428mg, J.O.C. 26, 755 (1961)에 따라 제조됨)의 용액에 N,N'-디사이클로헥실 카보디이미드(288mg)을 가하고 생성된 용액을 0℃에서 20분간 교반시킨다.

[실시예 5]
화합물 N-데포밀-N-(p-니트로벤조일)디스타마이신 A 하이드로클로라이드 (2.6g)를 2N HCL (10ml) 및 CH3OH(150ml) 혼합물에 용해시키고 H2압력 (50 p.s.i.)하에서 4시간 동안 Pd 촉매(탄소상 10%)상에서 환원시킨다. 촉매를 여과하고, 생성된 용액을 진공하에서 농축시켜 건조한다. 잔사를 에탄올(10ml)로 처리하고 1시간 동안 교반시킨후 여과하여 N-데포밀-N-(p-아미노벤조일)디스타마이신 A, 디하이드로클로라이드(2g)를 수득한다.

근육내 주사 20mg/ml
주사용 약제학적 조성물은-(N-메틸-4-(N-메틸-4-(N-메틸-4-(N-메틸-4-(α-브로모아크릴아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)프로피온아미딘, 하이드로클로라이드 20g을 주사용 물(1000ml)중에 용해시키고 1내지 5ml의 앰플에 밀봉시킴으로써 제조할 수 있다.

Claims (6)

  1. 하기 일반식(I)의 화합물 및 이의 약제학적으로 허용되는 염.
    상기식에서, n은 1내지 5의 정수이고: R1및 R2는 각각 동일하거나 상이할 수 있으며, 수소, 할로겐, -CN, -NO2, C1-C4알킬 또는 그룹이고; R3는 수소, 할로겐, -CN 또는 -NO2이며, R4는 각각 독립적으로 수소 또는 C1-C4알킬이고; A는 결합, 그룹또는 그룹 -NH-Het-CO-(여기에서, Het는 포화 또는 불포화 5원 또는 6원 헤테로모노사이클릭 환이다)이며; B는 그룹또는(여기에서, m은 1, 2 또는 3이고, R5는 각각 독립적으로 C1-C4알킬 그룹이다)이다.
  2. 제1항에 있어서, n이 3,4 또는 5이고; A가 결합 또는 그룹이며; B가 그룹또는이고; R1및 R2가 수소이며; R3가 염소 또는 브롬이고, R4가 메틸인 일반식(I)의 화합물 및 약제학적으로 허용되는 산과의 이의 염.
  3. 제1항에 있어서, N-데포밀-N-(α-클로로아크릴로일)디스타마이신 A;-(N-메틸-4-(N-메틸-4-(N-메틸-4-(N-메틸-4-(α-클로로-아크릴아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)프로피온아미딘; N-데포밀-N-(α-브로모아크릴로일)디스타마이신 A;-(N-메틸-4-(N-메틸-4-(N-메틸-4-(α-브로모아크릴아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-(카복스아미도)프로피온아미딘; β-(N-메틸-4-(N-메틸-4-(N-메틸-4-(N-메틸-4-(N-메틸-4-(α-브로모아크릴아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피로피온아미딘;N-데포밀-N-(4-(α-브로모아크릴아미도)벤조일)-디스타마이신 A; 및 3-(N-메틸-4-(N-메틸-4-(N-메틸-4-(N-메틸-4-(α-클로로-아크릴아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)피롤-2-카복스아미도)프로필-디메틸아민 중에서 선택된 일반식(I)의 화합물 및 이의 약제학적으로 혀용되는 염.
  4. 제1항 내지 제3항중 어느 한 항에 있어서, 하이드로클로라이드인 염.
  5. A)하기 일반식(Ⅱ)의 화합물 또는 이의 염을 일반식(Ⅲ)의 화합물과 반응시켜 일반식(I)의 화합물 또는 이의 염을 수득하거나; B)하기 일반식(Ⅳ)의 화합물 또는 이의 염을 하기 일반식(Ⅴ)의 화합물과 반응시켜, A가 그룹또는인 일반식(I)의 화합물 또는 이의 염을 수득함을 포함하여, 제1항에 따른 일반식(I)의 화합물 또는 이의 염을 제조하는 방법.
    상기식에서, R1, R2, R3, R4, A, B, Het 및 n은 제1항에서 정의한 바와 같으며; X는 하이드록시 또는 이탈 그룹이고; Z는또는 H2N-Het-CO-이다.
  6. 활성물질로서의 제1항에 따른 일반식(I)의 화합물 또는 이의 약제학적으로 허용되는 염을 적합함 담체, 희석제 또는 이의 혼합물과 함께 함유하는, 항종양제로서 유용한 약제학적 조성물
