Claims (6)
- 무세포시스템에서 단백질을 제조하는 방법에 있어서, 세포를 배양한 후 분쇄하여 원하는 단백질의 생산에 필요한 조직을 포함하는 세포추출물을 제조하는 단계; 및 상기 세포추출물과 함께 유전정보원, 단백질 합성을 위한 제1에너지원으로 작용하는 ATP, GTP, 제1에너지원을 반응기 내에서 바로 (in situ) 재생시키는데 필요한 제2에너지원으로서의 고에너지 인산화합물 또는 탄수화물 또는 이들의 유도체, 기질로서 아미노산 혼합물(여기에서 아미노산은 통상적으로 사용되는 20종의 L-형아미노산을 의미함) 및 직경이 0.1 내지 1000㎛이고 비표면적이 10내지 1000㎡/g인 다공성 고체물질을 포함하는 반응배지를 분리막형 단백질 합성 반응기에 공급하여 원하는 단백질을 제조하는 단계를 포함하는 것을 특징으로 하는, 무세포시스템에서 단백질을 제조하는 방법.
- 제1항에 있어서, 상기 제1에너지원의 분해산물이 미토콘드리아 또는 광합성조직을 이용한 별도의 에너지재생수단에 의해 제1에너지원을 재생된 다음 상기 단백질 합성 반응기로 순환되는 단계를 더 포함하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 분리막이 상기 단백질 합성 반응기의 출구에 설치되며, 그 외면에 접촉하고 있는 용액을 교환해 가면서 투석운전하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 다공성 고체물질이 다공성 키토산, 다공성 셀룰로오스, 다공성 젤라틴, 다공성 콜라겐 및 그들의 유도체로부터 선택되는 유기고분자물질인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 다공성 고체물질이 알루미나, 실리카, 티타니아, 지르코니아, 몰리브데나, 바나둠 옥사이드, 코발트 옥사이드, 이들의 혼합물 및 제오라이트로부터 선택되는 무기산화물인 것을 특징으로 하는 방법.
- 제1항에 있어서, 원하는 단백질에 대한 흡착성이 높은 물질로 충전된 컬럼을 포함하는 단백질 분리수단에 의해 원하는 단백질을 분리하는 단계를 포함하고 있는 것을 특징으로 하는 방법.
