Claims (9)
- YPS(앱신, yapsin(yeast aspartic protease, 효모 아스파틱 프로테아제); 이하 'YPS'라 약칭함) 6 및 YPS7로 구성된 군으로부터 선택된 하나 이상의 유전자가 결손되고, 사카로마이세스 세레비시애(Saccharomyces cerevisiae), 한세눌라 폴리모르파(Hansenula polymorpha), 피키아 패스토리스(Pichia pastoris), 야로위아 리포리티카(Yarrowia lipolytica), 스키조사카라마이세스 폼베(Schizosaccharomyces pombe)로 구성된 군으로부터 선택되는 앱신 결손 효모 변이 균주.
- 제 1항에 있어서, YPS1, YPS2 및 YPS3로 구성된 군으로부터 선택된 하나 이상의 유전자가 추가로 결손된 것을 특징으로 하는 효모 변이 균주.
- 제 1항에 있어서, 효모는 사카로마이세스 세레비시애인 것을 특징으로 하는 균주.
- 제 2항에 있어서, YPS1, YPS2, YPS3 및 YPS6 유전자가 결손된 것을 특징으로 하는 SLH23(YPS1ΔYPS2ΔYPS3ΔYPS6Δ) 균주;(수탁번호: KCTC 10384BP).
- 제 2항에 있어서, YPS1, YPS2, YPS3, YPS6 및 YPS7 유전자가 결손된 것을 특징으로 하는 SLH28(YPS1ΔYPS2ΔYPS3ΔYPS6ΔYPS7Δ)균주;(수탁번호: KCTC 10385BP).
- 1) 제 1항 내지 제 5항 중 어느 한 항의 앱신 결손 변이 균주를 숙주세포로 하여 재조합 단백질 유전자를 포함한 발현 벡터를 도입하는 단계;2) 단계 1에서 제조한 재조합 단백질 유전자가 도입된 앱신 결손 변이 균주를 배양하는 단계; 및3) 단계 2의 배양 후 분비되는 재조합 단백질을 정제하는 단계를 포함하는 것을 특징으로 하는 재조합 단백질의 대량 생산 방법.
- 제 6항에 있어서, 재조합 단백질 유전자는 단일 또는 쌍으로 존재하는 염기성 아미노산 잔기를 포함하는 재조합 단백질을 코딩하는 유전자인 것을 특징으로 하는 방법.
- 제 7항에 있어서, 재조합 단백질은 인체 혈청 알부민, 글루카곤, 글루카곤 유사 펩타이드, 인체 엘라핀 전구체, 곤충 이뇨 호르몬, 인간 부갑상선 호르몬 및 인간 리포코틴 I으로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 8항에 있어서, 재조합 단백질은 인간 부갑상선 호르몬 또는 인간 리포코틴 I인 것을 특징으로 하는 방법.
