Abstract
본 발명은, 스핑고이드 베이스를 포함하는 국소적 조성물을 적용함으로써, 국소적으로 일어나는 미생물 생장을 억제하는 것을 개시한다. 특히, 상기 스핑고이드 베이스는 계면활성제와 결합하여 효과적으로 조제된다.
Description
본 발명은 항균제로서 역할을 하는 스핑고이드 베이스를 포함하는 국소적 조성물의 분야에 관한 것이다.
건강한 사람 피부는, 피부의 자연적인 미생물적 평형을 유지하는 역할을 하는 많은 상이한 미생물에 의해 콜로니화되어 있다. 그러나, 피부 미생물상(microflora) 내의 특정 종의 생장은 쉽게 일어날 수 있으며, 그로써 미용적으로 그리고 피부과학적으로 바람직하지 못한 현상, 즉, 불쾌한 체취, 그리고 더 심한 경우에는 감염등을 일으킨다. 원하지 않는 미생물 생장과 관련된 몇몇 피부 상태들이 알려져 있다. 예를 들어, 상처입은 또는 병에 걸린 피부는 아토피성 습진을 앓는 환자에게 가장 중요한 감염제이기도 한 박테리아인 Staphylococcus aureus에 의해 중복감염(superinfection)되기 쉽다. 게다가 여드름은 Propionibacterium acnes 박테리아의 생장에 관련한다. 또한 진균성 피부 감염(진균증, mycoses)도 빈번하게 일어난다고 알려져 있다.
피부 표면 지질이, 그램-양성 박테리아에 대해 항균 활성을 갖는 한 가지 이상의 지질 화합물을 함유한다는 것은 수십년간 알려져 온 것이다(Burtenshaw (1942), J.Hyg.42, 184-209). 이들 항균성 지질 화합물은 주로 보통의 미생물상으로부터의 리파아제에 의해 피지의 트리글리세리드로부터 방출된 유리 지방산이라고 생각되었다(Kearney 외. (1984), Br.J.Dermatol. 110, 593-599). 최근에서야 이러한 점에서 스핑고지질의 역할에 대해 주의가 집중되었다.
최근의 일련의 논문에서, Bibel 외는 스핑고이드 베이스에 항균 활성이 있다고 하였다. 최초의 논문에서, 약 0.0005 내지 0.005%의 스핑고이드 베이스를 함유하는 시험 혼합물은 생체외에서 미생물 생장을 억제한다고 기술되었다(Bibel 외. (1992), J.Invest.Dermatol. 98, 269-273).
그러나 이들 혼합물 내에는 고농도의 에탄올이 추가로 존재하며, 에탄올은 그 자체가 항균 활성에 실질적으로 기여하는 용매로서 알려져 있다. 더욱이, 그램-음성 박테리아에 대하여는 아주 낮은 활성이 관찰되거나 활성이 관찰되지 않았다. 생체 내 실험에서(Bibel 외.(1992), 상기의 것; Bibel 외.(1995), Clin.Exper. Dermtol. 20, 395-400), 스핑고이드 베이스를 매우 고농도로, 즉 1.6% 에탄올 용액 또는 광유 내의 1.5% 에탄올 용액의 현탁액으로 적용하였다. 그러나, 이들 조제물들은 활성 성분을 피부까지 효과적으로 송달할 수 있다고 생각되지는 않는다. 예를 들어, Bibel 외.(1995)는 건조시 스핑가닌의 과립화는 이 화합물의 감소된 이용성을 초래한다고 보고하였다.
본 발명은 효과적으로 조제된 스핑고이드 베이스, 즉, 고농도의 에탄올 없이 조제된 스핑고이드 베이스를 포함하는, 국소적으로 일어난 미생물 생장의 억제용 조성물을 개시한다.
도 1은 Staphylococcus aureus 및 Corynebacterium xerosis를 스핑고신의 농도를 높여가면서 인큐베이트한 후 얻어지는 CFUs이다.
도 2는 Escherichia coli를 시간이 증가하는 동안 세 가지 농도의 피토스핑고신과 함께 인큐베이트한 후 얻어지는 CFUs이다.
도 3은 Pseudomonas aeruginosa를 시간이 증가하는 동안 두 가지 농도의 피토스핑고신과 함께 인큐베이트한 후 얻어지는 CFUs이다.

본 발명은 스핑고이드 베이스가 에탄올과 같은 항균 용매의 억제 농도(inhibiting concentration) 존재 없이 강력한 항균 활성을 갖는다는 것을 개시한다. 원하지 않는 미생물 생장과 관련한 다양한 피부 상태에 국소적으로 적용하기에 적절한 효과적인 조제물이 개시되어 있다.
본 발명은 더 나아가 에탄올의 억제 농도 존재 없이 생체 외에서 실질적인 항균 효과를 얻기 위해 필요한 스핑고이드 베이스의 농도는 적어도 0.005wt%여야한다는 것을 개시한다. 더 높은 농도, 즉, 0.01, 0.02, 0.04 또는 0.08wt%가 사용될 때, 스핑고이드 베이스의 항균 효과는 증가한다.
본 발명 전체를 통하여, "항균 활성/효과" 또는 "생장-억제 활성/효과"라는 용어는 동의어로 사용된다.
스핑고이드 베이스의 항균 활성을 감지할 수 있는 미생물에는 박테리아, 효모 및 진균류가 포함된다.
적어도 약 0.005%의 농도의 스핑고이드 베이스를 사용할 때, 스핑고이드 베이스는 그램-음성 박테리아에 대한 생장-억제 활성을 또한 보인다는 것을 본 발명은 개시한다. 특히, 본 발명은 스핑고이드 베이스가 그램-양성 박테리아에 대한 활성과 비교하여 거의 유사한 그램-음성 박테리아에 대한 생장-억제 활성을 보인다는 것을 개시한다.
본 발명은 더 나아가 스핑고이드 베이스가 실질적인 항진균 활성을 보인다는 것을 개시한다. 항진균 활성에는 사상균 뿐만 아니라 효모에 대한 생장-억제 활성도 포함되는 것으로 이해된다.
본 발명은 국소적 사용을 위한 조성물에 포함되는 스핑고이드 베이스는, 이온성(음이온 및/또는 양이온) 및 비이온성 계면활성제의 군에서 선택되는 계면활성제와 결합할 때 효과적으로 조제된다는 것을 개시한다. 바람직하게는, 계면활성제는 비이온성 계면활성제류의 군으로부터, 더 바람직하게는 트윈80과 같이 에톡실화-소르비탄-에스테르류의 군으로부터 선택된다.
본 발명에 따른 국소적 조성물은 용매로서 물이 사용되는 조성물, 용매로서 연화제(예를 들어, 지방 또는 오일)가 사용되는 조성물, 그리고 용매로서 물과 지방 또는 오일이 사용되는 조성물(에멀션)을 포함한다.
스핑고이드 베이스가 국소적 조성물 내에서 효과적으로 조제되는 농도는 0.001 내지 5wt%, 바람직하게는 0.005 내지 5wt%, 더 바람직하게는 0.01 내지 2.5wt%, 가장 바람직하게는 0.02 내지 1wt%, 특히 바람직하게는 0.02 내지 0.5wt%의 범위일 수 있다.
스핑고이드 베이스의 어떤 농도가 유리하게 사용되는지는 적용의 유형에 따라 달라질 수 있다. 보통, 감염, 예를 들어 부상 감염의 치료는 예방적 사용, 예를 들어 피부 미생물상의 정상화보다 더 높은 투여량의 스핑고이드 베이스를 요구할 것이다.
계면활성제는 일반적으로 0.01 내지 10%, 바람직하게는 0.1 내지 5%, 더 바람직하게는 0.5 내지 2.5%의 범위의 농도로 사용된다.
사용되는 스핑고이드 베이스의 유형은 본 발명에서 중요하지 않다. 일반적으로, 스핑고이드 베이스는 스핑가닌류, 스핑고신류 또는 피토스핑고신류의 군으로부터 선택된다. 바람직하게는, 스핑고이드 베이스는 피토스핑고신류의 군으로부터 선택된다.
본 발명에 따라 사용되는 스핑고이드 베이스는, 예를 들어 천연의 소스로부터 또는 화학 합성 공정으로부터 등 어떠한 적절한 소스로부터도 얻을 수 있다. 그러나 스핑고이드 베이스를 화학적으로 실행가능한 비용으로 충분한 양을 얻을 수 있도록 생산 공정을 적용하는 것이 바람직하다. 이러한 점에서, 현재의 일부 스핑고이드 베이스 소스는 단점을 가질 수 있다. 화학 합성의 경우에, 정확한 입체화학적 구조를 조제하는 것이 매우 어렵다. 동물 및/또는 식물 조직 추출물의 정제의 경우에는, 스핑고이드 베이스의 양이 매우 작으며 분리에 비용이 많이 든다. 더욱이, 동물 소스는 BSE(광우병)를 유발하는 병원체와 같이 다른 감염성 병원체 및 바이러스의 존재로 인하여 안전하지 않다고 생각된다.
그러므로, 스핑고이드 베이스는 바람직하게는 미생물 발효 공정으로부터 얻는다. 더 바람직하게는 효모, 특히 바람직하게는 Pichia ciferrii로부터 얻는다. 본 발명의 한 구체예에서, 스핑고이드 베이스 피토스핑고신은 Pichia ciferrii-유도된 테트라아세틸-피토스핑고신(TAPS)으로부터, 적절한 탈아세틸화 반응에 의해 얻는다. 상기 탈아세틸화는, 예를 들어 염기 촉매에 의해 수산화칼륨으로 가수분해하는 것처럼 화학적이거나 또는 효소적일 수 있다. 스핑고이드 베이스를 포함하는 본 발명에 따른 조성물은, 피부, 모발, 그리고 구강, 코, 눈, 비뇨생식기계 등의 상피 내막에 미용적 및/또는 피부과학적 적용을 포함하는 것으로 이해되는 국소적 적용에 적절하다. 스핑고이드 베이스를 포함하는 국소적 조성물은, 국소적으로 일어나는 다양한 미생물 활동과 관련한 바람직하지 못하고 그리고/또는 비정상적인 상태에 적용하기 적절하다.
스핑고이드 베이스를 포함하는 본 발명에 따른 국소적 조성물은 또한 고약, 붕대 등의 형태로도 유리하게 적용될 수 있다.
스핑고이드 베이스를 포함하는 국소적 조성물이 유리하게 적용될 수 있는, 국소적으로 일어나는 바람직하지 못한 그리고/또는 비정상적인 상태의 예는, 여드름, 비듬, 구강 및/또는 입술 감염, 진균증, 다양한 다른 피부-감염 질환 또는 질 감염 등이다. 스핑고이드 베이스를 포함하는 국소적 조성물은 더 나아가 예를 들어 화상과 같은 상처-치유, 그리고 피부 미생물상의 정상화에 유리하게 적용된다.
그것들의 항균 활성으로 인하여, 스핑고이드 베이스는 부가적으로 미용 및 피부과학적 조성물에서 방부제로서 역할을 할 수 있어서, 화학적 방부재의 존재를 감소시키고 그리고/또는 그에 대신할 수 있다.

항박테리아 활성의 측정
모든 박테리아 균주는, 37℃, 280rpm에서, 100ml의 브레인 하트 인퓨젼(BHI) 배지를 함유하는 500ml 진탕 플라스크 내에서 하룻밤 동안 배양하였다. P.acnes만 멸균 질소 가스로 세척(flush)된 BHI 배지 내에서 48-72 시간 동안 30℃에서 교반 없이 혐기적으로 배양하였다.
선택된 스핑고이드 베이스의 항박테리아 활성은, 스핑고이드 베이스와 함께 박테리아 균주를 인큐베이션한 후 얻어지는 콜로니 형성 단위(CFUs)의 양으로 측정한다. 이 목적을 위하여, 1% 네오펩톤 내의 10배 희석된 하룻밤 동안의 배양액 100㎕를 0.7% 네오펩톤, 0.6%의 에탄올, 1.1%의 트윈80, 그리고 적당량의 스핑고이드 베이스를 함유하는 용액 300㎕에 가하였다. 생성된 혼합물을 특정 시간 동안 37℃에서 인큐베이트하고, 이 때 생리학적 염 내의 106배 희석액 100㎕를 BHI 한천 평판위에 도말하였다. 생성된 콜로니를 계수하였다.
항진균 활성의 측정
96-웰 마이크로타이터플레이트 내에서의 액체 희석법.
-10mg 피토스핑고신/ml의 모액을, 20% 에탄올과 16% 트윈80을 함유하는 물 내에서 제조하였다;
-그 모액으로부터 10배 희석액을 Sabouraud 덱스트로스 브로스(Difco) 내에서 제조하였다;
-Sabouraud 덱스트로스 브로스 상에서 배양된 Saccharomyces cerevisiae 및 Candida albicans로부터, 수회 연속 희석액을 시험에 필요한 총수(total count)의 2배로 상기 배지 내에서 제조하였다;
-피토스핑고신의 10배 희석액 100㎕를 96-웰 마이크로타이터플레이트의 좌측 첫 번 째 웰들에(수직식으로) 가하였다;
-각 열의 웰들에, 적절한 배양액 및 희석액 100㎕를 가장 높은 총수가 위에 오도록 하는 식으로 가하였다;
-첫번째 웰을 혼합한 후, 이 웰로부터 취한 100㎕를 오른쪽 두번째 웰에 가하였다;
-두번째 웰을 혼합한 후, 이 웰로부터 취한 100㎕를 오른쪽 세번째 웰에 가하였다;
-각 열에 대하여 이 희석율을 만들어서, 왼쪽에서 오른쪽으로는 희석율이 1000, 500, 250㎍ 피토스핑고신/ml 등이 되게, 그리고 위에서 아래로는 총수 내에서 연속적인 희석율이 되게 제조하였다.

한천 희석법.
-20mg 피토스핑고신/ml 모액을, 20% 에탄올과 16% 트윈80을 함유하는 물 내에서 제조하였다;
-그 모액으로부터 20배 희석액을, 녹이고 식힌 Malt Extract Agar(MEA, Difco)내에서 제조하였다;
-이 희석액 50ml를, 녹이고 식힌 MEA 50ml와 혼합하였다;
-두번째 희석액 50ml을, 녹여서 식힌 50ml의 MEA 등과 혼합하여서, 1000, 500, 250㎍ 피토스핑고신/ml MEA 등의 희석 농도를 만들었다;
-각 희석액으로부터 20ml를 플라스틱 페트리접시(9cm)에 붓고, 식혀서 굳힌다;
-적절한 온도와 시간에서 Microsporum canis를 MEA 상에서 배양하였다;
-배양한 후, 생리학적 염 내의 살균 유리구슬을 가하였다;-그 배양액을 균질한 덩어리가 얻어질 때까지 진탕하였다;
-이렇게 얻어진 배양액 10㎕를 다양한 한천접시 상에 점으로 찍고, 적절한 온도와 시간에서 배양하였다;
-Sabouraud 덱스트로스 브로스 내에서 배양된 Saccharomyces cerevisiae로부터, 적당한 연속 희석을 같은 브로스 내에서 행하였다;

사용된 스핑고이드 베이스는 스핑고신(Sigma)과 피토스핑고신(Pichia ciferrii -유도된 테트라아세틸피토스핑고신의 탈아실화로부터 얻어짐)이었다.
실시예 1
여러 가지 농도의 스핑고신의 항박테리아 활성
박테리아 균주를 37℃에서 60분간 스핑고신의 농도를 높여가면서 인큐베이트함으로써, 두가지의 박테리아 균주에 대한 스핑고신(S)의 항박테리아 활성을 분석하였다(더 상세한 것은 실험을 참조).
S.aureus와 C.xerosis에 대한 스핑고신의 항박테리아 활성을 도시한 두 개의 표시 그래프(도 1)는, CFUs의 양이 스핑고신의 농도가 0.005에서 0.02%로 증가함에 따라 감소한다는 것을 보여준다.
비교가능한 스핑고신 및/또는 피토스핑고신의 항박테리아 활성은, M.Luteus, E.coli, P.aeruginosa 및 P.acnes를 사용하여 측정하였다.
실시예 2
시간과 관련한 피토스핑고신의 항박테리아 활성
선택된 박테리아 균주를 37℃에서 240분까지의 시간 동안, 피토스핑고신을 함유한 용액과 함께 인큐베이트함으로써, 시간과 관련하여 여러 박테리아에 대한 피토스핑고신(PS)의 항박테리아 활성을 분석하였다(더 상세한 것은 실험을 참조).
E.coli, P.aeruginosa, 및 P. acnes를 둘 또는 셋의 상이한 농도의 피토스핑고신과 함께 인큐베이트한 결과는 도 2, 3, 및 4에 각각 도시되어 있다. 더 긴 인큐베이션 시간에서 일어나는 박테리아의 생장은, 보다 높은 농도, 즉, 0.02 내지 0.08wt%의 피토스핑고신을 이용함으로써 예방될 수 있다.
실시예 3
피토스핑고신의 항진균 활성
효모 S.cerevisiae 및 C.albicans에 대한 피토스핑고신의 항진균 활성은 액체 희석법을 사용하여 측정할 수 있다(실험을 참조). 얻어진 m.i.c 값은 아래에 나타내었다. 이들 값들은 피토스핑고신이 효모에 대하여 강력한 생장 억제 활성을 갖는다는 것을 가리킨다.

Claims (20)
- 스핑고이드 베이스, 및 비이온성 및 이온성 계면활성제들의 군에서 선택되는 계면활성제를 포함하는 국소적으로 일어나는 미생물 생장을 억제하기 위한 국소적 조성물.
- 제 1 항에 있어서, 상기 국소적 조성물은 비이온성 계면활성제들의 군에서 선택되는 계면활성제를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어서, 상기 미생물 생장은 박테리아, 효모, 및 진균류의 군에서 선택되는 미생물에 의해 초래되는 것을 특징으로 하는 국소적 조성물.
- 제 3 항에 있어서, 상기 박테리아는 그램-음성 박테리아인 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어서, 상기 국소적 조성물은 0.001 내지 5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어, 상기 국소적 조성물은 0.005 내지 5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어, 상기 국소적 조성물은 0.01 내지 2.5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어, 상기 국소적 조성물은 0.02 내지 1wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어, 상기 국소적 조성물은 0.02 내지 0.5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 국소적 조성물.
- 제 1 항 또는 제 2 항에 있어서, 스핑고이드 베이스가 스핑가닌, 스핑고신, 및 피토스핑고신의 군에서 선택되는 것을 특징으로 국소적 조성물.
- 제 10 항에 있어서, 스핑고이드 베이스는 피토스핑고신인 것을 특징으로 하는 국소적 조성물.
- 스핑고이드 베이스와 비이온성 및 이온성 계면활성제들의 군에서 선택되는 계면활성제를 포함하는 국소적 조성물을 적용하는 것을 포함하는, 국소적으로-일어나는 미생물 생장을 억제하는 방법.
- 제 12 항에 있어서, 상기 국소적 조성물은 0.001 내지 5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 방법.
- 제 12 항에 있어서, 상기 국소적 조성물은 0.005 내지 5wt% 범위 농도의 스핑고이드 베이스를 포함하는 것을 특징으로 하는 방법.
