Claims (13)
- 표적(target) 핵산과 상보적인 서열 범위 내에, 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소가 절단 가능한 인위적인 서열을 포함하는 핵산 탐침을 준비하는 단계;상기 준비된 핵산 탐침에 비드를 부착하여 비드가 부착된 핵산 탐침을 준비하는 단계 ;상기 비드가 부착된 핵산 탐침을, 그 직경이 20nm 내지 120nm인 나노 크기의 포어를 가진 고체 기판의 나노포어 주위에 고정시키는 단계 ;상기 핵산 탐침이 고정화된 기판에 표적 핵산 함유의 시료를 투입시키고, 표적 핵산과 핵산 탐침을 혼성화시키는 단계 ;상기 표적 핵산과 핵산 탐침이 혼성화된 고체기판에 상기 인위적인 서열을 절단할 수 있는 상기 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소를 반응시켜, 핵산 단편이 부착된 비드를 효소반응 산물로 제조하는 단계; 및상기 효소반응 산물에 포함된 핵산 단편 부착 비드가 나노포어를 통과할 때 나타나는 전기적 신호를 검출하는 단계를 포함하는 비드 및 나노포어를 이용한 핵산검출방법.
- 제1항에서, 상기 비드는 그 직경이 1nm 내지 10nm인 것을 특징으로 하는 비드 및 나노포어를 이용한 핵산검출방법.
- 삭제
- 제1항에서, 상기 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소가 절단 가능한 인위적인 서열은 표적 핵산과 미스매치되는 염기를 1~3개 가지는 것을 특징으로 하는 비드 및 나노포어를 이용한 핵산검출방법.
- 제4항에서, 상기 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소는 미스매치 염기 인식 제한효소(mismatch cleavage enzyme)인 것을 특징으로 하는 비드 및 나노포어를 이용한 핵산검출방법.
- 제1항에서, 상기 표적 핵산은 70bps 내지 300 bps 크기인 것을 특징으로 하는 비드 및 나노포어를 이용한 핵산검출방법.
- a) 핵산의 관통을 수용하는 크기로서 그 직경이 20nm 내지 120nm인 나노포어를 가진 고체 기판,b) 상기 고체기판의 나노포어에 전압을 인가하는 전극, 및c) 상기 고체기판의 나노포어 주위에 고정화 된, 비드가 부착되고 표적(target) 핵산과 상보적인 서열 범위 내에 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소가 절단 가능한 인위적인 서열을 포함하는 핵산 탐침으로 구성된 나노포어 검출부;상기 나노포어 검출부로 투입되는 표적 핵산을 포함하는 시료가 보관된 시료 저장챔버; 및,상기 나노포어 검출부로 투입되는 상기 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소가 보관된 효소 저장챔버를 포함하는 나노포어를 통과하는 핵산 단편 부착의 비드를 검출하는 핵산검출장치.
- 제7항에서, 상기 시료 저장챔버 및 상기 효소 저장챔버와 상기 나노포어검출부를 각각 연결하는 연결부가 더 포함되는 것을 특징으로 하는 나노포어를 통과하는 핵산 단편 부착의 비드를 검출하는 핵산검출장치.
- 제7항에서, 상기 비드는 그 직경이 1nm 내지 10nm인 것을 특징으로 하는 나노포어를 통과하는 핵산 단편 부착의 비드를 검출하는 핵산검출장치.
- 삭제
- 제7항에서, 상기 핵산의 특정서열 및 구조에 특이적으로 작용하는 효소가 절단 가능한 인위적인 서열은 표적 핵산과 미스매치되는 염기를 1~3개 가지는 것을 특징으로 하는 나노포어를 통과하는 핵산 단편 부착의 비드를 검출하는 핵산검출장치.
