Abstract
Description
도 2는 포도당 대신 당화액으로 제조한 CGM 고체배지에서 2일 배양 후 대조군과 비교한 TM1-3 균주이다.
도 3은 자일로스-포도당 동시발효균주인 TM2-1 내지 TM2-20의 부탄올 생성능을 나타낸다.
도 4는 TM2-1, TM2-16 및 TM2-19 균주들의 포도당과 자일로스의 동시발효능을 나타낸다.
도 5는 자일로스를 40% 포함하는 배지에서 TM2-1 균주의 동시발효능을 나타낸다.
도 6은 pGS1-AdhE1 플라스미드를 나타낸다.
도 7은 pGS1-CtfAB 플라스미드를 나타낸다.
도 8은 pGS1-E1AB의 플라스미드를 나타낸다.
도 9는 혼합당을 포함하는 목질계 당화액으로 TM2-1-C(pGS1-E1AB) 균주를 162.5 시간 동안 연속 발효 시 발효 시간에 따른 배지 내 당 프로파일을 나타낸다.
Claims (20)
- 글루코스 및 자일로스에 대하여 동시발효능을 갖고,
또한 부탄올 생성능을 갖는 클로스트리디움 아세토부틸리쿰 TM2-1-C(기탁번호 KCTC 12604BP).
- 제 1항에 있어서,
목질계 당화액에 내성을 갖는 것을 특징으로 하는 클로스트리디움 아세토부틸리쿰 TM2-1-C.
- 삭제
- 제 1항에 있어서,
글루코스 및 자일로스의 동시발효능은 배치발효 기준으로 총 대사된 당 량의 30%이상이 자일로스이고, 부탄올 생산성이 1.0 g/L/h 이상인 것을 특징으로 하는 클로스트리디움 아세토부틸리쿰 TM2-1-C.

- 글루코스를 포함하는 배지를 준비하는 단계;
상기 배지에 제 1항의 클로스트리디움 아세토부틸리쿰 TM2-1-C를 접종하는 단계;및
상기 클로스트리디움 아세토부틸리쿰 TM2-1-C를 배양하는 단계를 포함하는 부탄올의 생산 방법.
- 제 1항의 클로스트리디움 아세토부틸리쿰 TM2-1-C에서,
알데히드/알코올 디하이드로게나아제에 의하여 부티릴 코에이를 부탄올로 전환하는 경로 또는 코에이트랜스퍼라제에 의하여 부티레이트를 부티릴 코에이로 전환하는 경로가 촉진된,
부탄올 생성능이 증가된 재조합 미생물.
- 제 9항에 있어서,
알데히드/알코올 디하이드로게나아제의 활성이 증가됨으로써 부티릴 코에이를 부탄올로 전환하는 경로가 촉진되는 것을 특징으로 하는 재조합 미생물.
- 제 9항에 있어서,
코에이트랜스퍼라제의 활성이 증가됨으로써 부티레이트를 부티릴 코에이로 전환하는 경로가 촉진되는 것을 특징으로 하는 재조합 미생물.
- 제 9항에 있어서,
자일로스, 아라비노스 및 셀로비오스로 구성된 군으로부터 선택된 하나 이상의 당과 글루코스에 대하여 동시발효능을 갖는 것을 특징으로 하는 재조합 미생물.

- 제 9항에 있어서,
유가식 배양을 기준으로 부탄올 선택도가 70% 이상인 것을 특징으로 하는 재조합 미생물.
- 제 9항에 있어서,
유가식 배양을 기준으로 아세톤 선택도가 20% 미만인 것을 특징으로 하는 재조합 미생물.
- 제 9항에 있어서,
유가식 배양을 기준으로 에탄올 선택도가 20% 미만인 것을 특징으로 하는 재조합 미생물.
- 제 9항에 있어서,
유가식 배양을 기준으로 부탄올 생산성이 0.5 g/L/h 이상인 것을 특징으로 하는 재조합 미생물.
- 자일로스, 아라비노스 및 셀로비오스로 구성된 군으로부터 선택된 하나 이상의 당과 글루코스를 포함하는 배지를 준비하는 단계;
상기 배지에 제 9항의 재조합 미생물을 접종하는 단계;및
상기 재조합 미생물을 배양하는 단계를 포함하는 부탄올의 생산 방법.
