Abstract
Description
도 2는 재조합 대장균 균주(Inha254 및 Inha256)에서 세포 성장 및 안트라닐레이트 생산 수율의 비교 결과를 나타낸다. (A) OD600에서 측정한 세포 성장, (B) 안트라닐레이트 생산 수율의 정량 분석.
도 3은 7L 바이오리액터에서 안트라닐레이트 생산을 위한 유가식 배양(Fed-batch fermentation) 결과를 나타낸다. RPM의 시간 경과, DO 농도, pH, DCW 및 안트라닐레이트 생산 수율을 각각 나타냈다. 공급은 15 hr에 0.1701 ml/min으로 시작하여 27 hr 및 42 hr에 각각 0.1323과 0.1701 ml/min으로 변경하였다.

Claims (5)
- 시키메이트(shikimate) 고생산 균주에 aroL 및 aroK 유전자를 과발현시키고, trpD, pheA , tyrA , pabA, entC 및 ubiC 유전자를 결실시킨, 안트라닐레이트(anthranilate) 고생산 재조합 대장균 균주.
- 제1항에 있어서, 상기 시키메이트(shikimate) 고생산 균주는 E. coli AB2834 균주에 aroB , aroD , aroF , aroG , galP, ppsA , aroE 및 tktA 유전자를 과발현시키고, tyrR , ptsG , pykA , aroL , aroK 및 shiA 유전자를 결실시킨 것을 특징으로 하는 안트라닐레이트(anthranilate) 고생산 재조합 대장균 균주.
- 제1항에 따른 균주의 유전체(chromosome)에 aroE 유전자를 삽입하고, trpR 유전자를 결실시키고, tktA 유전자는 J23119 프로모터의 조절 하에서 과발현시키고, aroL 및 aroK 유전자는 종결코돈과 개시코돈이 결합된 형태로 치환시킨, 안트라닐레이트(anthranilate) 고생산 재조합 대장균 균주.
- 제3항에 있어서, 상기 재조합 대장균 균주는 수탁번호 KCTC19060P로 기탁된 균주인 것을 특징으로 하는 안트라닐레이트(anthranilate) 고생산 재조합 대장균 균주.
- 제1항 내지 제4항 중 어느 한 항에 따른 재조합 대장균 균주를 배양하는 단계를 포함하는 안트라닐레이트(anthranilate) 생산 방법.
