Claims (35)
- 본질적으로 화학적으로 정의된 구성성분들로 구성된 화학적 합성 발효 배지에서 산업적 규모로 미생물 균주를 발효시키는 단계와, 발효 육즙으로부터 가치있는 화합물을 회수하는 단계를 포함하는 가치있는 화합물의 제조방법.
- 제 1항에 있어서, 화학적 합성 배지가 본질적으로 소량의 복합 탄소 및/또는 질소원을 함유하는 것을 특징으로 하는 방법.
- 제 1항 또는 제 2항에 있어서, 화학적 합성 배지의 화학적으로 정의된 구성성분들이 글루코스, 락토스, 프룩토스, 수크로스, 말토덱스트린, 녹말 및 이눌린 등의 탄수화물류, 글리세롤, 식물성 오일류, 탄화수소류, 메탄올 및 에탄올 등의 알코올류, 아세테이트 및 고급 알카노산 등의 유기산류로 이루어지는 군으로부터 선택된 탄소원과, 유레아, 암모니아, 질산염, 황산 암모늄, 인산 암모늄 및 질산 암모늄 등의 암모늄염류 및 글루타메이트 및 리신 등의 아미노산류로 이루어지는 군으로부터 선택된 질소원을 포함하는 것을 특징으로 하는 방법.
- 제 3항에 있어서, 탄소원이 글루코스이고 질소원이 암모니아 및/또는 암모늄염인 것을 특징으로 하는 방법.
- 제 1항 내지 제 4항 중 어느 한 항에 있어서, 발효가 회분식, 반복 회분식, 유가식, 반복 유가식 또는 연속 발효 공정을 통해 일어나는 것을 특징으로 하는 방법.
- 제 5항에 있어서, 발효가 유가식 공정을 통해 일어나는 것을 특징으로 하는 방법.
- 제 6항에 있어서, 탄소 및/또는 질소원을 그 공정에 피딩하는 것을 특징으로 하는 방법.
- 제 7항에 있어서, 탄소원이 글루코스이고 질소원이 암모니아 및/또는 암모늄염인 것을 특징으로 하는 방법.
- 제 1항 내지 제 8항 중 어느 한 항에 있어서, 가치있는 화합물이 약제학적 단백질 또는 펩티드, 일차 또는 이차 대사산물, 또는 산업적 효소인 것을 특징으로 하는 방법.
- 제 9항에 있어서, 가치있는 화합물이 이차 대사산물인 것을 특징으로 하는 방법.
- 제 10항에 있어서, 이차 대사산물이 β-락탐 화합물인 것을 특징으로 하는 방법.
- 제 9항에 있어서, 가치있는 화합물이 효소인 것을 특징으로 하는 방법.
- 제 1항 내지 제 9항 중 어느 한 항에 있어서, 미생물 균주가 효모인 것을 특징으로 하는 방법.
- 제 13항에 있어서, 효모가 Phaffia rhodozyma이고 가치있는 화합물이 아스탁산틴인 것을 특징으로 하는 방법.
- 제 1항 내지 제 9항 중 어느 한 항에 있어서, 미생물 균주가 사상 미생물 균주인 것을 특징으로 하는 방법.
- 제 15항에 있어서, 사상 균주가 균류인 것을 특징으로 하는 방법.
- 제 16항에 있어서, 균류가 Aspergillus 균주인 것을 특징으로 하는 방법.

- 제 21항에 있어서, Mucorales 균주가 Blakeslea 균주인 것을 특징으로 하는 방법.
- 제 25항에 있어서, Mucorales 균주가 Blakeslea trispora이고 가치있는 화합물이 β-카로틴인 것을 특징으로 하는 방법.
- 제 15항에 있어서, 사상 균주가 박테리아인 것을 특징으로 하는 방법.
- 제 27항에 있어서, 박테리아가 Actinomycete인 것을 특징으로 하는 방법.
- 제 28항에 있어서, Actinomycete가 Streptomyces 균주이고 가치있는 화합물이 글루코스 이소머라제인 것을 특징으로 하는 방법.
- 제 28항에 있어서, Actinomycete가 Streptomyces clavuligerus이고 가치있는 생성물이 클라불란산인 것을 특징으로 하는 방법.
- 제 28항에 있어서, Actinomycete가 Saccharopolyspora erythraea이고 가치있는 화합물이 에리쓰로마이신인 것을 특징으로 하는 방법.
- 화학적 합성 배지에서 산업적 규모로 발효될 수 있는 원하는 가치있는 화합물을 생산하는 미생물 균주의 제조 및/또는 개선 방법으로서 하기 단계들을 포함하는 방법:* 적합한 부모 균주를 물리적 수단 및 화학적 돌연변이원들로 이루어지는 군으로부터 선택된 돌연변이유발 처리 및/또는 DNA 형질전환시키는 단계,* 결과된 돌연변이체 및/또는 형질전환체들을 화학적 합성 배지 상에서의 그들의 생장 성능 및 원하는 상기 가치있는 화합물의 생산성 레벨에 대해 스크리닝하는 단계,* 상기 부모 균주와 비교하여 화학적 합성 배지 상에서의 생장 성능이 좋고/좋거나 원하는 상기 가치있는 화합물의 생산성 레벨이 향상된 돌연변이체들 및/또는 형질전환체들을 셀렉션하는 단계.
- 제 32항에 있어서, 부모 균주가 화학적 합성 배지 상에서 생장 성능은 좋지만 생산성 레벨이 향상될 필요가 있는 균주들로 이루어지는 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 32항에 있어서, 부모 균주가 원하는 화합물의 생산성 레벨은 높으나 상대적으로 화학적 합성 배지 상에서의 생장 성능이 나쁜 균주들로 이루어지는 군으로부터 선택되는 것을 특징으로 하는 방법.
- 미생물 균주의 발효에 의해 가치있는 화합물을 산업적 규모로 생산하기 위한 화학적 합성 발효 배지의 이용.
