Definitions
- the present invention relates to a chip for manufacturing lipid nanoparticles containing a spiral structure and a method for manufacturing lipid nanoparticles using the same.
- Lipid nanoparticles are effective drug delivery systems for biologically active compounds such as cell-impermeable therapeutic nucleic acids, proteins, and peptides.
- vaccines are subdivided into “first generation”, “second generation” and “third generation” vaccines, and genetic vaccines, i.e. vaccines for genetic vaccination, are usually understood as “third generation” vaccines.
- Genetic vaccines typically consist of genetically engineered nucleic acid molecules that allow expression in vivo of peptides or protein (antigen) fragments characteristic for pathogens or tumor antigens. When administered to a patient, genetic vaccines are expressed after uptake by target cells. Expression of the administered nucleic acid results in the production of the encoded protein. When these proteins are recognized as foreign by the patient's immune system, an immune response is triggered.
- DNA as well as RNA can be used as nucleic acid molecules for administration.
- DNA is known to be relatively stable and easy to handle.
- DNA carries the risk of unwanted insertion of administered DNA fragments into the patient's genome, potentially resulting in mutagenic events such as loss of function of the damaged gene.
- RNA is quite unstable and can be easily degraded by ubiquitous RNases, resulting in problems of impermeability, fragility and immunogenicity.
- lipid nanoparticle formulations have been used as an efficient method for mRNA vaccination that can induce adaptive immune responses.
