Abstract

Description
발명의 배경
본 발명은 일반적으로 콜라겐 및 콜라겐-유도된 조성물 및 그들의 제조 방법에 관한 것이다. 특히, 본 발명은 향상된 비율로 물을 흡수할 수 있는 건성의 가교된 젤라틴 또는 다른 콜라겐 또는 콜라겐-유도된 조성물을 제조하는 방법에 관한 것이다.
본 출원의 양수인인 퓨전 메티칼 사(Fusion Medical Technologies, Inc.)는 플로실(FloSeal)(R)이라는 상품명 하에 지혈 조성물을 생산한다. 플로실(R) 제품은 두 개의 시린지(syringe; 이후 "주입기"라고도 함)를 포함하는 포장으로 시판된다. 첫번째 시린지는 완충 용액으로 미리-수화된 가교된 소 젤라틴의 과립으로 채워진다. 상기 젤라틴 수화겔은 약 85%(w/w)의 물을 함유하며 유동성 수화겔의 형태이다. 수술실에서 사용 직전에, 염수 중 트롬빈이 젤라틴 수화겔과 혼합된다. 트롬빈은 염수 중에 제조되며 두번째 시린지에 흡인되고, 시린지들은 트롬빈과 젤라틴의 혼합을 허용하도록 함께 연결되어 있다.

퓨전 메디칼 사에 의해 투자된 Abstract]에 기재되어 있다. 플로실(R) 제품을 포함하는 특허는 미국 특허 제 6,063,061 호 및 6,066,325 호를 포함한다. 플로실(R) 제품의 콜라겐, 젤라틴 또는 다른 콜라겐-유도된 성분 및 트롬빈 성분을 혼합 및 운반하기 적절한 두 개의 시린지 계는 미국 특허 제 5,908,054 호에 기재되어 있다. 상기 특허 참고문헌 각각의 완전한 개시가 여기에 참고문헌으로 도입된다.
발명의 간단한 요약
본 발명은 개선된 지혈 봉함 조성물, 그러한 개선된 조성물을 제조하는 방법 및 상기 개선된 조성물을 포함하는 키트를 제공한다. 상기 방법 및 조성물은 외과적 출혈 부위, 외상성 출혈 부위 등을 포함하는 출혈 부위에서 지혈을 제공하는 데 특히 유용할 것이다. 상기 조성물의 예시적 용도는 혈관 카테테르의 경우 생긴 혈관 관통의 위에 조직 관을 봉함하는 것일 것이다.
상기 조성물은 신속히 재-수화하도록 제조된 건성의 가교된 젤라틴 분말을 포함한다. 상기 젤라틴 분말은 바람직하게는, 미국 특허 제 6,063,061 호 및 6,066,325 호(이들의 전체 개시는 앞에서 참고문헌으로 도입되었다)에 기재된 바와 같이 단편 또는 부-단위라고도 일컬어지는 비교적 큰 입자를 포함한다. 바람직한 입자 크기는 150 μm 내지 750 μm의 범위일 것이나, 상기 바람직한 범위 외의 입 자 크기도 많은 경우에 사용될 수 있다. 건성 조성물은 또한 수성 재-수화 매질에 노출될 때 상당한 "평형 팽윤율"을 나타낼 것이다. 바람직하게는, 상기 팽윤율은 400% 내지 1000%의 범위일 것이나 상기 언급된 특허에 기재된 바와 같이 상기 범위 밖에 해당할 수도 있다. "평형 팽윤율"은 완전히 수화되어 완전히 팽윤될 때의 중량으로부터 젤라틴 수화겔 분말의 건조 중량을 감하여 결정될 수 있다. 다음 상기 차이를 건조 중량으로 나누고 100을 곱하여 팽윤의 척도를 수득한다. 건조 중량은 모든 잔류 수분을 실질적으로 제거하기에 충분한 상승된 온도 및 시간 동안, 예를 들면 120℃에서 2 시간 동안, 물질을 노출시킨 후 측정되어야 한다. 상기 물질의 평형 수화는 상기 건조 물질을 염수와 같은 적절한 재-수화 매질 중에, 수분 함량이 일정해지기 충분한 시간 동안, 전형적으로 실온에서 18 내지 24 시간 동안, 침지시킴으로써 수행될 수 있다.
본 발명의 건성 가교된 젤라틴 분말은 일반적으로 약간의 잔류 수분을 가질 것이나, 바람직한 안정성 및 연장된 보관 수명을 얻기 위해 충분히 건조될 것이다. 전형적으로, 상기 건성 조성물은 20 중량%(w/w) 이하의 수분 함량, 바람직하게는 5 중량% 내지 15 중량% 범위의 수분 함량을 갖는다. 건성을 유지하기 위해, 상기 조성물은, 본 발명의 키트에 관련하여 더 상세하게 기재하는 바와 같이, 일반적으로 수분 침입을 방지하기 적절한 방식으로 포장될 것이다.
본 발명의 한 특정 국면에서, 조성물은 20%(w/w) 이하의 수분 함량을 갖는 가교된 젤라틴 분말을 포함할 것이며, 여기에서 상기 분말은 상기 분말이 재-수화 보조제 없이 제조된 유사한 분말의 재-수화율에 비하여 5% 이상 더 높은 수성 재- 수화율를 갖도록 재수화 보조제의 존재 하에 가교되었다. 여기에서 정의된 "재-수화율"은 30 초 내에 분말 1 그램(건조 중량 기준)에 의해 흡수된, 일반적으로 0.9%(w/w) 염수인, 수용액의 양을 gm/gm으로 나타낸 것을 의미한다. 상기 비율을 측정하기 위한 특별한 기술은 이하의 실험 부분에서 기재한다. 본 발명의 바람직한 조성물은 3 gm/gm 이상, 바람직하게는 3.5 gm/gm 이상, 종종 3.75 gm/gm 이상의 재-수화율을 가질 것이다. 재-수화 보조제 없이 제조된 유사한 분말의 재-수화율은 전형적으로 3 미만이며, 재-수화율의 백분율 증가는 일반적으로 5% 이상, 바람직하게는 10% 이상, 더욱 바람직하게는 25% 이상일 것이다.
향상된 재-수화율을 갖는 본 발명의 건성 가교된 젤라틴 분말은 바람직하게는 특정 재-수화 보조제의 존재 하에 분말을 제조함으로써 수득된다. 그러한 재-수화 보조제는 분말의 제조 도중에 존재할 것이지만, 최종 생성물로부터는 일반적으로 제거될 것이다. 예를 들면, 총 고형분 약 20%로 존재하는 재-수화 보조제는 일반적으로 최종 생성물에서 1% 미만, 종종 0.5 중량% 미만으로 감소될 것이다. 예시적인 재-수화 보조제는 바람직하게는 약 1000의 분자량을 갖는 폴리에틸렌 글리콜(PEG); 바람직하게는 약 50,000의 평균 분자량을 갖는 폴리비닐피롤리돈(PVP); 및 전형적으로 약 40,000의 평균 분자량을 갖는 덱스트란을 포함한다. 본 발명의 조성물을 제조할 때 적어도 2 종의 상기 재-수화 보조제를 사용하는 것이 바람직하며, 세 가지 모두를 사용하는 것이 더욱 특히 바람직하다.
따라서 본 발명의 방법은 재-수화 보조제와 조합된 비-가교된 젤라틴의 수용액을 제공하는 것을 포함한다. 비-가교된 젤라탄은 일반적으로 5%(w/w) 내지 15%(w/w)로 수용액 중 존재할 것이고, 재-수화 보조제는 수용액 중 젤라틴의 중량을 기준으로 5% 내지 30%(w/w)로 존재할 것이다. 바람직하게는, 재-수화 보조제는 젤라틴의 중량을 기준으로 2.5% 내지 20%(w/w)의 PEG, 1.25% 내지 20%(w/w)의 PVP, 및 1.25% 내지 20%(w/w)의 덱스트란을 포함한다.
재-수화 보조제와 함께 상기 비-가교된 젤라틴을 수화겔을 형성하기 적절한 임의의 방법으로 가교시킨다. 예를 들면, 중합체 분자는 두 개 이상의 중합체 분자 쇄에 공유결합적으로 부착된 2-작용성 또는 폴리-작용성 가교제를 이용하여 가교될 수 있다. 예시적인 2-작용성 가교제는 알데히드, 에폭시류, 숙신이미드, 카보디이미드, 말레이미드, 아지드, 카보네이트, 이소시아네이트, 디비닐 술폰, 알코올, 아민, 이미데이트, 무수물, 할라이드, 실란, 디아조아세테이트, 아지리딘 등을 포함한다. 그렇지 않으면, 가교는, 중합체 상의 측쇄 또는 잔기를 활성화하여 다른 측쇄 또는 잔기와 반응함으로써 가교 결합을 형성하게 하는, 페리오데이트와 같은 산화제 및 기타 시약을 이용하여 수행될 수 있다. 가교의 추가적인 방법은 중합체를 감마 방사선 같은 방사선에 노출시켜 중합체 사슬이 가교 반응을 허용하도록 하는 것을 포함한다. 탈수열(dehydrothermal) 가교 방법이 또한 적절할 수 있다. 젤라틴 분자를 가교시키는 바람직한 방법을 이하에 기재한다.
가교된 젤라틴을 제조하는 예시적 방법은 다음과 같다. 젤라틴을 수득하고 수용액에 현탁시켜, 전형적으로 1% 내지 70 중량%, 일반적으로 3% 내지 10 중량%의 고형분을 갖는 비-가교된 수화겔을 형성한다. 젤라틴은 전형적으로 글루타르알데히드(예를 들면, 0.01% 내지 0.05% w/w, 수성 완충액 중 0℃ 내지 15℃에서 밤새), 소듐 페리오데이트(예를 들면, 0.05 M, 48 시간 동안 0℃ 내지 15℃에서 유지) 또는 1-에틸-3-(3-디메틸아미노프로필)카보디이미드("EDC")(예를 들면, 0.5% 내지 1.5% w/w, 실온에서 밤새)에 노출시키거나, 약 0.3 내지 3 메가래드의 감마 또는 전자 빔 방사에 노출시킴으로써 가교된다. 그렇지 않으면, 젤라틴 입자를 바람직하게는 메틸 알코올 또는 에틸 알코올 같은 알코올 중, 1% 내지 70 중량%, 일반적으로 3% 내지 10 중량%의 고형분으로 현탁시키고, 전형적으로 글루타르알데히드(예, 0.01% 내지 0.1% w/w, 실온에서 밤새) 같은 가교제에 노출시킴으로써 가교시킬 수 있다. 알데히드의 경우, pH는 약 6 내지 11, 바람직하게는 7 내지 10으로 유지되어야 한다. 글루타르알데히드로 가교될 경우, 가교는 쉬프 염기를 통하여 형성되고, 이는 이어지는 환원, 예를 들면 수소화 붕소 나트륨 처리에 의해 안정화될 수 있다. 가교 후, 수득되는 과립을 물에 세척하고, 선택적으로 알코올로 헹군 다음, 건조시킬 수 있다. 수득되는 건성 분말을, 이후에 상세히 기재하는 바와 같이 본 발명의 도포기 내에 부하할 수 있다.
가교 후, 재-수화 보조제의 적어도 50%(w/w)가 수득되는 수화겔로부터 제거될 것이다. 일반적으로, 재-수화 보조제는 수화겔을 여과한 다음 수득되는 여과 케이크를 세척함으로써 제거된다. 그러한 여과/세척 단계는 상기 생성물을 원하는 수준까지 세정하고 재-수화 보조제의 50% 이상을 제거, 바람직하게는 원래 존재하던 재-수화 보조제의 90%(w/w) 이상을 제거하기 위해 1회 이상 더 반복될 수 있다.
여과 후, 젤라틴을, 생성된 최종 여과 케이크를 건조시킴으로써 전형적으로 건조시킨다. 다음 건조된 여과 케이크를 부수거나 분마하여 전술한 바람직한 범위 의 입자 크기를 갖는 가교된 분말을 제조할 수 있다.
본 발명에 따르는 키트는 전술한 본 발명의 건성 가교된 젤라틴 분말을 보유하는 첫번째 용기를 포함할 것이다. 상기 키트는, 젤라틴이 재-수화될 때 젤라틴과 함께 혼합되도록 의도된 트롬빈을 포함하는 염수 또는 다른 수용액인, 수성 재-수화 매질을 보유하는 두번째 용기를 더 포함할 것이다. 상기 용기는 임의의 형태일 수 있으나 건조 젤라틴과 재-수화 매질의 혼합을 허용하는 시린지의 형태인 것이 바람직할 것이다.
도 1은 본 발명의 원리에 따라 구성된 키트를 도시한다.
구체적 구현예의 설명
이하의 실시예는 예시를 위해 제공되며, 한정하고자 함이 아니다.
실시예 1: 젤라틴 분말의 제조
소의 진피 조각을 실온에서 1 시간 동안 1 M 내지 2 M 농도의 수산화 나트륨 용액에 현탁시키고, 인산으로 중화시킨 다음 헹구었다. 상기 처리된 조각을 다시 탈이온수에 재현탁시키고, pH를 7-8로 조정한 다음 70℃로 가열하였다. 조각의 크기를 더 감소시키기 위해 호모지나이저를 사용하였다. 70℃에서 1 시간 후, 상기 진피는 대부분 젤라틴으로 용해되었다. 진피의 양은 수득되는 젤라틴 용액의 고형분이 약 3-10%(w/w), 전형적으로 7-10%가 되도록 선택되었다. 상기 용액을 테플론(Teflon)(R) 피복된 금속 트레이 상에서 얇은 층으로 주조하고, 건조시킨 다 음 분마하여 젤라틴 분말을 형성하였다.
실시예 2: "개질된 젤라틴 분말"의 제조
재-수화 보조제(표 1)를 50℃의 탈이온수 500 mL에 용해시킨 다음, 실시예 1에서 제조된 일정 량의 소 유래 젤라틴 분말을 상기 용액에 가하였다. 용액 중 젤라틴의 최종 농도는 약 8%(w/w, 벌크 젤라틴 분말 기준)로 선택되었고, 재-수화 보조제의 용액 중 총량은 실시예 9-44에서와 같이 선택되었다(표 1 및 표 2). 젤라틴이 용해된 후, 용액을 테플론(R) 피복된 금속 트레이 상에 붓고 건조시켰다. 건조된 젤라틴 시트를 분마하여 "개질된 젤라틴 분말"을 형성한다.
그렇지 않으면, 소 진피의 조각을 1 M 내지 2 M 농도의 수산화 나트륨 용액 중 실온에서 1 시간 동안 현탁시키고, 인산으로 중화시킨 다음 헹구었다. 다음, 처리된 조각을 탈이온수에 재현탁시키고, pH를 7-8로 조절하여 70℃로 가열하였다. 조각의 크기를 더 감소시키기 위해 호모지나이저를 사용하였다. 70℃에서 1 시간 후, 상기 진피는 대부분 젤라틴으로 용해되었다. 진피의 양은 수득되는 젤라틴 용액의 고형분이 약 3-10%(w/w), 전형적으로 7-10%가 되도록 선택되었다. 재-수화 보조제의 양은 실시예 9-44(표 1 및 2)에서와 같이 선택되었고, 이어서 이는 상기 젤라틴 용액에 고체 형태로 또는 소량 부피의 물에 용해된 상태로 첨가되었다. 상기 용액을 테플론(R) 피복된 금속 트레이 상에서 얇은 층으로 주조하고, 건조시킨 다음, 분마하여 "개질된 젤라틴 분말"을 형성하였다. 개질된 젤라틴의 경우 몇가지 조성의 예를 표 1 및 표 2에 나타낸다.
실시예 3: "개질된 젤라틴 분말"로부터 가교된 젤라틴 분말의 제조
600 mL의 0.2 M 인산염 완충액(pH 9.2 ± 0.2)을 12℃ 미만의 온도로 냉각시켰다. 0.32 mL의 글루타르알데히드(25%)를 상기 완충 용액에 가한 다음, 개질된 젤라틴 분말 20 g을 가하여, 글루타르알데히드 농도가 4000 ppm(글루타르알데히드 대 개질된 젤라틴, 벌크 중량 기준)이 되게 하였다. 젤라틴을 교반 막대를 이용하여 글루타르알데히드 용액 중 현탁시켰다. 각 현탁액의 pH를 9.2 ± 0.2의 범위로 조절한 다음 9 내지 12℃의 온도 및 pH 9.2 ± 0.2에서 19 시간 동안 유지시켰다.
현탁액을 여과하고, 여과 케이크를 탈이온수로 완전히 덮이게 함으로써 탈이온수로 3회 세척한 다음, 진공에 의해 케이크를 통해 헴굼 물을 빨아들였다. 각 헹굼의 동안 깔때기에 여과 케이크가 남았다.
0.2 g의 NaBH4를 비커 내에서 600 mL의 25 mM 인산염 완충액(pH 7.4 ± 0.2)에 용해시켰다. 상기 여과 케이크를 실온(약 22℃)에서 3 시간 동안 NaBH4 용액에 현탁시킨 다음 여과하여 액체를 제거하였다.
다음, 여과 케이크를 실온(약 22℃)에서 30 분 동안 600 mL 완충 용액에 현탁시키고, 다시 여과하였다. 완충액은 인산 나트륨(이염기 무수 및 단일염기 단일수화물) 및 소듐 아스코르베이트로 구성되었다. 상기 과정을 2 회 반복하여 염 대 젤라틴의 적절한 비가 존재하여 재구성 시 바람직한 완충액 조성물을 형성할 것을 보장하였다. 상기 여과 케이크를 건조시킨 다음 워링(Waring) 블렌더로 분마하여 "가교된 젤라틴 분말"을 수득하였다.
상기 방법은 개질되지 않은 젤라틴 분말; 즉, 그 제조 도중 재-수화 보조제가 첨가되지 않은 젤라틴으로부터 가교된 젤라틴 분말을 제조하기 위해서도 사용되었다.
실시예 4: 가교된 젤라틴 분말로부터 방사선 조사된 생성물의 제조
실시예 2에서 제조된 약 800 mg(벌크 중량)의 가교된 젤라틴 분말을 몇 개의 5 cc 시린지 각각에 넣었다. 분말을 함유하는 시린지를 주위 온도에서 감마선 조사로 소독하였다.
실시예 5: 생성물의 지혈제로서의 용도
개질된 젤라틴 분말로부터 약 0.8 g의 방사선 조사된 가교 젤라틴 분말을 함유하는 생성물의 시린지를 제조하였다. 상기 개질된 젤라틴 분말은 실시예 2에서와 같이 제조되었다. 상기 개질된 젤라틴을 실시예 3 및 실시예 4에서와 같이 더 가교시키고 방사선을 조사하였다. 상기 생성물을 1 밀리리터 당 소 트롬빈 약 1000 단위를 함유하는 염수 4 mL와 혼합하였다. 혼합은 암컷-암컷(female-female) 직접-통과(straight-through) 루어(Luer) 연결기를 이용하여 연결된 두 시린지 사이에서 왕복 통과시킴으로써 수행되었다. 시린지 안의 분말은 그것이 트롬빈 용액과 혼합되면서 수화되어 수화겔의 과립을 형성하였다.
약 1 cm x 1 cm x 0.2 cm 깊이의 정사각형 상해를 농장-급(farm-grade) 돼지의 간에 만들었다. 상기 돼지는 헤파린으로 항응고처리되어, 그 활성화된 응혈 시간(ACT)이 기본선 값의 3 내지 5 배였고, 상기 상해는 처리 이전에 자유롭게 출혈되었다. 혼합을 시작한 지 약 30 초후, 약 2 mL의 수화된 분말을 시린지로부터 상 기 상해 위로 압출하고, 2 분 동안 압축하여 그 자리에 유지시켰다. 압축을 제거한 후, 처리된 상해를 적용 후 3분, 10 분 및 50 분에 출혈에 대하여 관찰하였다. 처리된 상해에서는 3 분 및 10 분 관찰 시에 출혈이 보이지 않았다. 10 분 관찰 후, 처리된 상해를 염수로 세척하였다. 과량의 물질이 제거되는 동안, 재-출혈이 관찰되지 않았다. 적용 후 50 분에, 상해를 다시 관찰하였고, 출혈이 보이지 않았다.
실시예 6: 분말의 재-수화율의 측정
분말의 "재-수화율"은 다음과 같이 측정되었다. 5 cc 시린지에 충진된 분말은, 루어 장치로 연결된 두 시린지 사이에서 30 초 동안 통과시킴으로써 일정 부피의 수용액을 함유하는 시린지와 혼합되었다. 수용액의 부피는 30 초 동안 흡수될 것으로 예상될 수 있는 과량으로 선택되었다. 전형적으로 0.8 g(벌크 중량)의 분말을 3 mL의 0.9% 염화 나트륨 용액과 혼합하였다. 다음 수득된 혼합물을 즉시 여과하여 임의의 흡수되지 않은 액체를 제거하였다. 습윤 여과된 물질을 칭량한 다음 2 시간 동안 120℃ 오븐에서 건조시키고 다시 칭량하였다. 상기 측정은 습윤 물질로부터 제거된 물의 총량 및 건조 분말의 중량을 제공하였다. 원래 분말에 존재했던 잔류 습기에 대하여 약간 보정한 후, 분말에 의해 흡수된 물의 양을 계산하였다. "재-수화율"은 그 30 초 간격 동안 분말의 건조 중량 1 그램 당 흡수된 염수의 질량으로 주어졌다.
아래의 계산에서, 벌크 분말의 고형분("S")은 벌크 분말을 120℃에서 2 시간 동안 건조시키고 건조 이전 및 이후 분말을 칭량함으로써 독립적으로 측정되었다. S 값은 다음 수학식에 의해 주어진다:

실시예 7: 몇 가지 분말 생성물 배치(batch)의 재-수화율 및 물성 측정
표 1 및 표 2는 하나 내지 몇 가지 분말 생성물(실시예 9-23)에 대하여 수행된 재-수화율 측정의 결과를 보여준다. 이들은 실시예 1, 2, 3 및 4에서와 같은 방법을 이용하여 제조되었다. 실시예 9 및 17을 제외하고는, 이들은 다양한 비율의 젤라틴 및 다음 재-수화 보조제: 폴리에틸렌 글리콜(PEG), 평균 분자량 1000; 폴리비닐피롤리돈(PVP), "k-30"이라 불리우며 약 50,000의 평균 분자량에 해당; 덱스트란, 평균 분자량 40,000:를 사용하여 만들어진 개질 젤라틴으로부터 제조되었다.
젤라틴과 재-수화 보조제의 여러 가지 상이한 조합을 사용하는 것은 재-수화 보조제가 첨가되지 않은 젤라틴으로부터 제조된 분말 생성물에 비하여 분말 1 그램 당 30 초 동안 더 많은 염수를 흡수하는 분말 생성물의 결과를 가져올 수 있음이 나타난다. 개질 젤라틴 중 젤라틴, PEG, PVP 및 덱스트란이 80:10:5.5의 벌크 중량 비로 조합된 것(실시예 10)이 개질되지 않은 젤라틴으로부터 만들어진 분말 생성물에 비하여 30초 동안 그램 당 약 33%의 염수를 더 흡수하는 분말 생성물을 생성하는 것이 또한 나타난다.
표 1은 또한 분말 제품 로트의 경우 측정된 다른 물성들의 값을 제공한다. "% 고형분"은 잔류 습기를 증발시키기 위해 120℃에서 2 시간 동안 건조시키기 전과 후에 분말을 칭량하여 결정되었다. "DSC 피크 온도"는 1℃ 내지 70℃에서 수행된 시차 주사 열량계 측정의 온도기록도에 피크가 나타나는 온도를 의미한다. "평형 팽윤율"은 분말을 과량의 염수에 실온에서 18 시간 이상 현탁시켜 측정하였다. 수화된 분말을 칭량하여 그의 "평형 습윤 중량"을 측정하고, 120℃에서 2 시간 동안 건조시키고 다시 칭량하여 그의 "건조 중량"을 측정하였다. 평형 팽윤율은 다음 수학식에 의해 주어진다:

표 1에 나타난 데이터로부터, 재-수화 보조제의 적절한 사용이 다른 물성을 실질적으로 변화시키지 않고 분말 제품의 재-수화율을 변화시킬 수 있음이 나타났다.
실시예 8: 개질된 젤라틴 분말 및 가교된 분말 중 폴리에틸렌 글리콜, 폴리비닐피롤리돈 및 덱스트란 수준의 측정
약 50 mg의 개질된 젤라틴 또는 250 mg의 가교되고 방사선 조사된 분말 생성물을 10 mL의 탈이온수에 현탁시키고 65℃에서 3 시간 동안 가열하였다. 다음 시료를 2000 rpm에서 15 분동안 원심분리하였다. 수득된 상청액을 0.45 μm 겔만아크로디스크(Gelman Acrodisc) 필터를 통해 여과하고, 처음 1 mL는 폐기하였다. 다음 수득된 시료를 3 가지 상이한 고성능 액체 크로마토그래피(HPLC) 법으로 분석하여 시료 중 폴리에틸렌 글리콜(PEG), 폴리비닐피롤리돈(PVP) 및 덱스트란을 정량하였다. PEG의 경우, 이동상으로 탈이온수를 사용하는, 보호 컬럼 및 예비필터를 갖는 워터스 울트라히드로겔(Waters Ultrahydrogel) 120 컬럼, 7.8 x 300 mm 상에 100 μL의 시료가 주입되었다. 굴절율 검출기가 배출물을 감시하는 데 사용되었다. PVP의 경우, 이동상으로 메탄올과 인산 나트륨 수용액의 구배를 사용하는, 보호 컬럼 및 예비필터를 갖는 피닉스 킹소르브(Phenomenex Kingsorb) C18 5 μm 컬럼, 4.6 x 150 mm 상에 100 μL의 시료가 주입되었다. 자외선 흡광도 검출기가 배출물을 감시하는 데 사용되었다. 덱스트란의 경우, 이동상으로 0.1 M 인산 나트륨, pH 7과 아세토니트릴을 90:10 비율로 사용하는, 보호 컬럼과 예비필터를 갖는 워터스 울트라히드로겔(Waters Ultrahydrogel) 직선형 컬럼, 7.8 x 300 mm 상에 100 μm의 시료가 주입되었다. 굴절율 검출기가 배출물을 감시하는 데 사용되었다. 모든 컬럼은 분석을 위해 40℃로 가열되었다. 정량의 한계는 PEG 및 PVP의 경우 약 0.1 %(w/w 시료), 덱스트란의 경우 0.2 %(w/w 시료)였다.
실시예 2에서와 같이 개질된 젤라틴을 제조하였다. 개질된 젤라틴을 전술한 방법으로 PEG, PVP 및 덱스트란에 대하여 분석하였다. 결과는 PEG, PVP 및 덱스트 란이 각각 16%, 8% 및 3%(w/w 벌크)로 존재함을 나타내었다. 개질된 젤라틴을 이어서 가교하고, 수소화붕소 나트륨 처리한 다음 실시예 3에서와 같이 헹구어 가교된 개질 젤라틴 분말을 형성하였다. 상기 분말을 PEG, PVP 및 덱스트란에 대하여 전술한 방법으로 HPLC로 분석할 때, 세 가지 재-수화 보조제 각각의 함량은 정량의 한계 미만인 것으로 밝혀졌다.
실시예 9: 재-수화 보조제 없이 제조된 분말 생성물
실시예 1에서와 같이 소 진피 조각으로부터 개질되지 않은 젤라틴 - 즉, 가공 보조제가 첨가되지 않은 젤라틴 - 을 제조하고 실시예 3에서와 같이 가교하였다. 가교된 개질되지 않은 젤라틴을 실시예 4에서와 같이 시린지 내에 충진하고 감마선을 조사하였다. 수득되는 생성물의 물성을 실시예 6 및 7에서와 같이 측정하여 표 1에 나타낸다.
실시예 10-23: 재-수화 보조제를 이용하여 만들어진 분말 제품
젤라틴 분말 또는 진피 조각 및 1종, 2종 또는 3종의 재-수화 보조제로부터 실시예 2에서와 같이 개질된 젤라틴 배치를 제조하였다. 표 1은 사용된 벌크 젤라틴 및 재-수화 보조제의 비율을 나타낸다. 다음, 개질된 젤라틴을 실시예 3에서와 같이 가교하였다. 실시예 17의 경우를 제외하고는, 사용된 재-수화 보조제는 다음 목록에서 선택되었다: 평균 분자량이 약 1000인 폴리에틸렌 글리콜(PEG); 평균 분자량이 약 50,000인 "k-30"이라 불리는 폴리비닐피롤리돈(PVP), 및 평균 분자량이 약 40,000인 덱스트란. 실시예 17에서는, 평균 분자량이 약 400인 PEG가 사용되었다. 다음 가교된 개질 젤라틴을 실시예 4에서와 같이 시린지에 충진하고 감마선을 조사하였다. 이들 조제 각각으로부터 수득되는 분말 생성물의 물성을 실시예 6 및 7에서와 같이 측정하여 표 1에 나타낸다. 실시예 10의 경우 조성과 함께 주어진 데이터는 그 조성에 따라 제조된 9 개 배치의 평균 및 표준 편차이다.
실시예 24-44: 다양한 재-수화 보조제를 사용하여 제조된 분말 생성물
젤라틴 분말 또는 진피 조각 및 몇 가지 재-수화 보조제의 1종으로부터 실시예 2에서와 같이 개질된 젤라틴의 배치를 제조하였다. 표 2는 각 배치에 사용된 재-수화 보조제의 종류 및 농도를 벌크 젤라틴 중량 대 재-수화 보조제의 비율로서, 및 상기 개질된 젤라틴을 제조하는 데 사용된 총 벌크 용질의 백분율로서 보여준다. 다음 개질된 젤라틴을 실시예 3에서와 같이 가교하였다. 다음 가교된 개질 젤라틴을 실시예 4에서와 같이 시린지에 충진하고 감마선을 조사하였다. 상기 제조 각각으로부터 수득되는 분말 생성물의 물성을 실시예 6 및 7에서와 같이 측정하여 표 2에 나타낸다. 실시예 9 조성의 데이터는 비교를 위해 표 2에 나타낸다.
이상은 본 발명의 바람직한 구현예의 완전한 기재이나, 다양한 선택의 여지, 수정 및 동등물이 사용될 수 있다. 따라서 상기 기재는 첨부된 청구범위에 의해 정의되는 본 발명의 범위를 제한하는 것으로 여겨져서는 안된다.
Claims (22)
1종 이상의 재-수화 보조제와 조합된 비-가교된 젤라틴을 포함하는 수용액을 제공하고;
상기 젤라틴을 가교시키고;
상기 재-수화 보조제의 50%(w/w) 이상을 제거하고;
상기 가교된 젤라틴을 건조시키고 분마하여 20%(w/w) 이하의 수분 함량을 갖는 분말을 생성하는 것을 포함하며, 상기 재-수화 보조제는 글리세롤, 덱스트란, 폴리비닐피롤리돈(PVP) 및 폴리에틸렌 글리콜(PEG)로 이루어진 군에서 선택된 것을 포함하는 것인, 가교된 젤라틴 분말의 제조 방법.

제 1 항에 있어서, 상기 재-수화 보조제가 상기 수용액에 존재하는 젤라틴의 중량을 기준으로 5 중량% 내지 30 중량% 범위의 농도로 존재하는 것인 방법.
제 3 항에 있어서, 상기 재-수화 보조제가 상기 수용액에 존재하는 젤라틴의 중량을 기준으로 2.5 중량% 내지 20 중량%의 PEG, 상기 수용액에 존재하는 젤라틴의 중량을 기준으로 1.25 중량% 내지 20 중량%의 PVP, 및 상기 수용액에 존재하는 젤라틴의 중량을 기준으로 1.25 중량% 내지 20 중량%의 덱스트란을 포함하는 것인 방법.
제 1 항에 있어서, 상기 가교가 젤라틴 용액에 가교제를 가하는 것을 포함하는 방법.
제 5 항에 있어서, 상기 가교제가 글루타르알데히드를 포함하는 방법.
제 1 항에 있어서, 재-수화 보조제의 50%(w/w) 이상을 제거하는 것이, 용매 중 가교된 분말의 현탁액을 여과하여 여과 케이크를 수득하고, 상기 여과 케이크를 세척하여 재-수화 보조제를 제거하는 것을 포함하는 방법.
제 7 항에 있어서, 상기 여과 케이크를 세척하는 것이, 원래 수용액 중에 존재하던 재-수화 보조제의 90%(w/w) 이상을 제거하는 것인 방법.
제 7 항에 있어서, 상기 건조는 세척 후 여과 케이크를 건조하는 것을 포함하며, 여기에서 상기 방법은 건조된 여과 케이크를 분마하여 건성의 분마된 젤라틴 분말을 제조하는 것을 더 포함하는 것인 방법.
20 %(w/w) 이하의 수분 함량을 갖는 가교된 젤라틴 분말을 포함하며, 상기 분말이 재-수화 보조제 없이 제조된 분말의 재-수화율보다 5% 이상 더 높은 수성 재-수화율을 갖도록 재-수화 보조제의 존재 하에 가교되고, 상기 재-수화 보조제는 글리세롤, 덱스트란, 폴리비닐피롤리돈(PVP) 및 폴리에틸렌 글리콜(PEG)로 이루어진 군에서 선택된 것을 포함하는 것인, 조성물.
제 10 항에 있어서, 상기 분말이 재-수화 보조제 없이 제조된 분말의 재-수화율보다 10% 이상 더 높은 재-수화율을 갖는 조성물.

제 10 항에 있어서, 상기 분말이 150 μm 내지 750 μm 범위의 평균 입자 크기를 갖는 조성물.

제 15 항에 있어서, 상기 수성 재-수화 매질이 트롬빈을 포함하는 키트.
제 15 항에 있어서, 상기 가교된 젤라틴 분말과 재-수화 매질을 조합하여 트롬빈-함유 젤라틴 수화겔 단편을 제조하고 상기 수화겔을 상처 부위에 적용하는 방법이 기재된 사용 지침을 더 포함하는 키트.
제 15 항에 있어서, 상기 첫번째 및 두번째 용기를 보유하는 포장을 포함하는 키트.
제 10 항에 있어서, 상기 분말이 재-수화 보조제 없이 제조된 분말의 재-수화율보다 25% 이상 더 높은 재-수화율을 갖는 조성물.
20 ％(w/w) 이하의 수분 함량을 갖는 가교된 젤라틴 분말을 포함하며, 상기 분말은 재-수화 보조제의 존재하에 가교되어 3 gm/gm 이상의 재-수화율을 갖고, 상기 재-수화 보조제는 글리세롤, 덱스트란, 폴리비닐피롤리돈 및 폴리에틸렌 글리콜로 이루어진 군에서 선택된 것인 조성물.
