Abstract
Description
도 2: 인간 말초 혈액 단핵 세포 사이토카인 IFN-γ 분비 시험. 결과는 시험 PD-1 항체 mAb005가 13ng/㎖의 EC50으로 PBMC 증식을 자극하고, 동시에 사이토카인 IFN-γ의 분비를 효과적으로 자극할 수 있다는 것을 보여준다.

Claims (27)
- 경쇄 가변 영역 및 중쇄 가변 영역을 포함하는 PD-1 항체 또는 이의 항원-결합 단편으로서,
상기 경쇄 가변 영역은 각각 서열 번호 6, 서열 번호 7 및 서열 번호 8에 나타낸 LCDR1, LCDR2 및 LCDR3을 포함하고; 상기 중쇄 가변 영역은 각각 서열 번호 3, 서열 번호 4 및 서열 번호 5에 나타낸 HCDR1, HCDR2 및 HCDR3을 포함하는, PD-1 항체 또는 이의 항원-결합 단편. - 제1항에 있어서, 상기 항체 또는 상기 이의 항원-결합 단편은 뮤린(murine) 항체 또는 이의 단편인, PD-1 항체 또는 이의 항원-결합 단편.
- 제1항에 있어서, 상기 항체 또는 상기 이의 항원-결합 단편은 키메라 항체 또는 이의 단편인, PD-1 항체 또는 이의 항원-결합 단편.
- 제3항에 있어서, 상기 키메라 항체의 상기 경쇄 가변 영역 서열은 서열 번호 10인, PD-1 항체 또는 이의 항원-결합 단편.
- 제3항에 있어서, 상기 키메라 항체의 상기 중쇄 가변 영역 서열은 서열 번호 9인, PD-1 항체 또는 이의 항원-결합 단편.
- 제1항에 있어서, 상기 항체 또는 상기 이의 항원-결합 단편은 인간화 항체 또는 이의 단편인, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체의 상기 경쇄 가변 영역의 경쇄 FR 서열은, IGKV 1-39의 FR1, FR2 및 FR3 및 JK4의 FR4를 포함하는, 서열 번호 14에 나타낸 인간 생식선(germline) 경쇄 IGKV1-39 및 JK4의 조합 서열로부터 유래한, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체 경쇄의 서열은 서열 번호 12에 나타낸 서열 또는 이의 변이체이고; 상기 변이체는 상기 경쇄 가변 영역에서 A43S 아미노산 돌연변이를 포함하는, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체의 상기 중쇄 가변 영역은 인간 IgG1, IgG2, IgG3 또는 IgG4의 중쇄 FR을 추가로 포함하는, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체의 상기 중쇄 가변 영역은 인간 IgG2 또는 IgG4의 중쇄 FR을 추가로 포함하는, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체의 상기 중쇄 가변 영역의 중쇄 FR 서열은, IgHV3-7의 FR1, FR2 및 FR3 및 JH6의 FR4를 포함하는, 서열 번호 13에 나타낸 인간 생식선 중쇄 IgHV3-7 및 JH6의 조합 서열로부터 유래한, PD-1 항체 또는 이의 항원-결합 단편.
- 제6항에 있어서, 상기 인간화 항체 중쇄의 서열은 서열 번호 11에 나타낸 서열 또는 이의 변이체이고; 상기 변이체는 상기 중쇄 가변 영역에서 G44R 아미노산 돌연변이를 포함하는, PD-1 항체 또는 이의 항원-결합 단편.
- 제1항 내지 제12항 중 어느 한 항에 따른 항체를 코딩하는 DNA 분자.
- 제13항에 따른 DNA 분자를 포함하는 발현 벡터.
- 제14항에 따른 발현 벡터에 의해 형질전환된 숙주 세포.
- 제15항에 있어서, 상기 숙주 세포는 박테리아인, 숙주 세포.
- 제16항에 있어서, 상기 숙주 세포는 이. 콜라이(E. coli)인, 숙주 세포.
- 제15항에 있어서, 상기 숙주 세포는 효모인, 숙주 세포.
- 제18항에 있어서, 상기 숙주 세포는 피치아 파스토리스(Pichia pastoris)인, 숙주 세포.
- 제1항 내지 제12항 중 어느 한 항에 따른 PD-1 항체 또는 이의 항원-결합 단편 및 약제학적으로 허용되는 부형제, 희석제 또는 담체를 포함하는, 암을 치료하기 위한, 약제학적 조성물.
