Abstract
La présente invention concerne la production et la purification d'antigène spécifique de la prostate humaine (hPSA) par une technique de recombinaison de l'ADN. Ce hPSA a été produit sous une forme active et sous une inactive au moyen du système de vecteurs d'expression du baculovirus. La protéine de recombinaison a été sécrétée en grande quantité dans le milieu de culture par des cellules d'insectes infectés et homogénéisée par purification grâce à un mode opératoire en trois temps faisant intervenir la chromatographie par échange cationique et la filtration sur gel. La protéine active obtenue diffère de la protéine naturelle en ce qu'elle n'est pas glycosilatée et qu'elle ne dégrade pas le substrat H-D-Pro-Phe-Arg-pNA.Hcl spécifique de la kallikréine. La forme inactive du hPSA obtenu comporte sur son extrémité N terminale deux résidus aminoacides supplémentaires et elle ne forme aucun complexe. L'invention concerne également des utilisations possibles de cette protéine de recombinaison.
