Abstract

La présente invention concerne un procédé de détection et/ou de quantification d'une séquence nucléotidique cible (2) présente dans un échantillon biologique caractérisé en ce qu'il comprend une mise en contact de type 'sandwich' de la séquence nucléotidique cible (2) avec une séquence nucléotidique monobrin trappeur (5) fixée sur un support solide insoluble (3) et complémentaire d'une partie (7) de ladite séquence nucléotidique cible (2) et avec une ou plusieurs séquences (6, 11) nucléotidiques dont au moins une est marquée (6), la ou lesdites séquences nucléotidiques (6, 11) étant complémentaires d'une autre partie (8) de la séquence nucléotidique cible (2); en ce que la séquence nucléotidique trappeur (5) est fixée de manière covalente par l'une de ses extrémités sur le support solide (3), comporte une longueur comprise entre 50 et 500 bases; et en ce que la partie (10) de la séquence nucléotidique trappeur (5) qui ne s'hybride pas avec la partie (7) de la séquence nucléotidique cible (2) est inférieure à 60 bases. La présente invention concerne également la trousse de détection et/ou de quantification pour la mise en oeuvre de ce procédé et l'utilisation d'une séquence nucléotidique standard de quantification.
