Abstract

L'invention concerne une méthode de mesure quantitative par spectrométrie de masse d'un peptide ou de peptides générés par une ou plusieurs protéines d'analysat par clivage enzymatique ou chimique. La méthode consiste à exposer ensemble l'échantillon à analyser et un ou plusieurs précurseurs de normes internes peptidiques marquées en masse à un clivage enzymatique ou chimique de façon à générer à partir dudit précurseur une ou plusieurs normes internes peptidiques marquées en masse. Le précurseur de normes internes peptidiques marquées en masse comprend une extension d'au moins un acide aminé N-terminal vers la terminaison N de la norme interne peptidique marquée en masse la plus N-terminale qui en est issue; et une extension d'au moins un acide aminé C-terminal vers la terminaison C de la norme interne peptidique marquée en masse la plus C-terminale qui en est issue.
